Clinical usefulness of ankle brachial index and brachial-ankle pulse wave velocity in patients with ischemic stroke

Ankle brachial index （ABI） and brachial-ankle pulse wave velocity （baPWV） are widely used noninvasive modalities to evaluate atherosclerosis. Recently, evidence has increased supporting the use of ABI and baPWV as markers of cerebrovascular disease. This study sought to examine the relationship between ABI and baPWV with ischemic stroke. This study also aimed to determine which pathogenic mechanism, large artery disease （LAD） or small vessel disease （SVD）, is related to ABI or baPWV. Retrospectively, 121 patients with ischemic stroke and 38 subjects with no obvious ischemic stroke history were recruited. First, ABI and baPWV were compared between the groups. Then, within the stroke group, the relevance of ABI and baPWV with regard to SVD and LAD, which were classified by brain magnetic resonance image （MRI） and magnetic resonance angiography （MRA） or computed tomography angiography （CTA） findings, was assessed. The baPWV was higher in the stroke group than non-stroke group （1,944.18±416.6 cm/s vs. 1,749.76±669.6 crn/s, P 〈 0.01）. Regarding LAD, we found that mean ABI value was lower in the group with extracranial large artery stenosis （P 〈 0.01）, and there was an inverse linear correlation between ABI and the grade of extracranial large artery stenosis （P〈 0.01）. For SVD, there was a significant correlation between SVD and baPWV （2,057.6±456.57 cm/s in the SVD （＋） group vs. 1,491±271.62 cm/s in the SVD （-） group; P 〈 0.01）. however, the grade of abnormalities detected in SVD did not correlate linearly with baPWV. These findings show that baPWV is a reliable surrogate marker ofischemic stroke. Furthermore, baPWVand ABI can be used to indicate the presence of small vessel disease and large arterial disease, respectively.

Genevestigator analysis has indicated heat shock induction of transcripts for NADPH-thioredoxin reduc-tase, type C （NTRC） in the light. Here we show overexpression of NTRC in Arabidopsis （NTRC°E） resulting in enhanced tolerance to heat shock, whereas NTRC knockout mutant plants （ntrcl） exhibit a temperature sensitive phenotype. To investigate the underlying mechanism of this phenotype, we analyzed the protein＇s biochemical properties and protein structure. NTRC assembles into homopolymeric structures of varying complexity with functions as a disulfide reductase, a foldase chaperone, and as a holdase chaperone. The multiple functions of NTRC are closely correlated with protein structure.. Complexes of higher molecular weight （HMW） showed stronger activity as a holdase chaperone, while low molecular weight （LMW） species exhibited weaker holdase chaperone activity but stronger disulfide reductase and fol-dase chaperone activities. Heat shock converted LMW proteins into HMW complexes. Mutations of the two active site Cys residues of NTRC into Ser （C217/454S-NTRC） led to a complete inactivation of its disulfide reductase and foldase chaperone functions, but conferred only a slight decrease in its holdase chaperone function. The overexpression of the mutated C217/454S-NTRC provided Arabidopsis with a similar degree of thermotolerance compared with that of NTRC°E plants. however, after prolonged incubation under heat shock, NTRC°E plants tolerated the stress to a higher degree than C217/454S-NTRC°E plants. The results suggest that the heat shock-mediated holdase chaperone function of NTRC is responsible for the increased thermotolerance of Arabidopsis and the activity is significantly supported by NADPH.

Jeju seaweeds suppress lipopolysaccharide-stimulated proinflammatory response in RAW 264.7 murine macrophages

Objective:To investigate the anti-inflammatory effects of Jeju seaweeds on macrophage RAW264.7 cells under lipopolysaccharide(LPS)***:Ethyl acetate fractions were prepared from five different types of Jeju seaweeds,Dictyopteris divaricata(***),Dictyopteris prolifera(***),Prioutis cornea(***,Grateloupia laceolata(G,lanceolate,and Cralcloupia filicina(***)They were screened for inhibitory effects on proinflammatory mediators and cytokines such as nitric oxide(NO),prostaglandin E,,tumor necrosis factor-a(TNF-a),and interleukin-6(11.-6).Results:Our results revealed that ***,***,***,***,and *** potently inhibited ***-stimulaled NO production(IC_(50),values were 18.0,38.36,38.43,32.81 and 37.14μg/mL,respectively).Consistent with these findings,***,***,***,and *** also reduced the IPS-induced and prostaglandin E,production in a concentration-dependent ***,they suppressed the expression of inducible NO synthase and cyclooxygenase-2 at the protein level in a dose-dependent manner in the RAW264.7 cells,as detennined by western *** addition,the levels of TNF-a and IL-6,released into the medium,were also reduced by ***,***,***,G,lanceolata,and *** in a dose-dependent manner(IC_(50)values for TNF-a were 16.11,28.21,84.27,45.52 and74.75μg/mL,respectively;IC_(50),values for IL-6 were 37.35,80.08,103.28,62.53 and 84.28μg/mL,respectively).The total phlorotannin content was measured by the Folin-Ciocalteu method and expressed as phloroglucinol *** content was 92.0μg/mg for ***,151.8μg/mg for ***,57.2μg/mg for ***,53.0 pg/mg for ***,and 40.2μg/mg for ***:Thus,these findings suggest that Jeju seaweed extracts have potential therapeutic applications for inflammatory responses.

Association between hepatocellular carcinoma and tumor necrosis factor alpha polymorphisms in South Korea

AIM: To investigate associations between the tumor necrosis factor alpha(TNF-α)-1031 T>C,-863 C>A,-857 C>T,-308 G>A,and-238 G>A polymorphisms and HCC in ***: Hepatocellular carcinoma(HCC) cases were diagnosed at CHA Bundang Medical Center from June 1996 to August 2008. The association between TNF-α polymorphisms and HCC was analyzed in 157HCC patients and 201 controls using a polymerase chain reaction-restriction fragment length polymorphism assay. We investigated five TNF-α polymorphisms,which are TNF-α-1031 T>C,-863 C>A,-857 C>T,-308 G>A,and-238 G>A. The TNF-α genotype frequencies,genotype combinations and haplotypes were analyzed to disclose the association with ***: None of the TNF-α polymorphisms was significantly associated with HCC. however,nine genotype combinations had associations with increased likelihood of HCC. Among them,TNF-α-1031/-857/-238 TT/CC/GA(AOR = 18.849,95%CI: 2.203-161.246,P = 0.007),TNF-α-1031/-308/-238 TT/GG/GA(AOR = 26.956,95%CI: 3.071-236.584,P = 0.003),and TNF-α-1031/-238 TT/GA(AOR = 21.576,95%CI: 2.581-180.394,P = 0.005) showed marked association with HCC. There were five haplotypes of TNF-α polymorphisms which were significantly associated with HCC. They are TNF-α-1031/-863/-857/-308/-238 T-C-C-G-A(OR = 25.824,95%CI: 1.491-447.223,P = 0.0005),TNF-α-1031/-857/-308/-238 T-C-G-A(OR = 12.059,95%CI: 2.747-52.950,P hose with the TT genotype(AOR = 5.795,95%CI: 1.145-29.323). CONCLUSION: Although no single TNF-α polymorphism is associated with HCC in this study,some TNF-α genotype combinations and haplotypes are associated with HCC. Apocrine carcinoma is a rare malignant adnexal neoplasm. The differential diagnosis between apocrine carcinoma and cutaneous metastasis is often difficult. Here, we report a case of locally recurrent penile apocrine carcinoma initially diagnosed as metastatic adenocarcinoma of the colon. A 75-year-old man with a history of surgical resection due to sigmoid colon cancer and penile metastasis two years prior to this study presented with a nodule at the left penile base. He underwent a wide local resection of the penile mass under a suggested preoperative diagnosis of extra-mammary Paget＇s disease （EMPD） associated with previous sigmoid colon cancer. however, the previously and currently resected penile masses were identified as primary apocrine carcinoma upon hematoxylin and eosin （H＆E） staining and immunohistochemical staining. Although the incidence is extremely rare, both clinicians and pathologists should be alert to the possibility of synchronous double primary apocrine carcinoma in cancer patients with malignant cutaneous lesions.

Electrical stimulation has recently received attention as noninvasive treatment in skin wound healing with its outstanding biological property for clinical ***,the complexity of equipment for applying appropriate electrical stimulation remains an ongoing ***,we proposed a strategy for skin scar inhibition by providing electrical stimulation via a multilayer stacked electret(MS-electret),which can generate direct current(DC)electric field(EF)without any power supply *** addition,the MS-electret can easily control the intensity of EFs by simply stacking electret layers and maintain stable EF with the surface potential of 3400 V over 5 days owing to the injected charges on the electret *** confirmed inhibition of type 1 collagen andα-SMA expression of human dermal fibroblasts(hDFs)by 90%and 44%in vitro,indicating that the transition of hDFs to myofibroblasts was restricted by applying stable electrical *** further revealed a 20%significant decrease in the ratio of myofibroblasts caused by the MS-electret in *** findings present that the MS-electret is an outstanding candidate for effective skin scar inhibition with a battery-free,physiological electrical microenvironment,and noninvasive treatment that allows it to prevent external infection.

Effects of PEG Priming and Fungicide Treatment on Kenaf(Hibiscus cannabinus L.)Seed Germination

An experiment was conducted in the Jeollabuk-do Agricultural Research and Extension Services,Korea from February to June 2017,to study the effect of polyethylene glycol(PEG)on the germination of kenaf seeds variety‘Jangdae’,which treated with seven PEG concentrations(0%,10%,20%,25%,30%,35%and 40%)for 24 h at 20°C in the dark,and evaluate the effect of fungicide on surging kenaf seed germination in the *** showed that the critical concentration rate needed to hasten germination based on germination percentage,times to reach 50%of the final germination rate(T50)and mean *** days to germination(MDG)is using 10%PEG.A maximum germination rate of 83.8%using hydro-priming(HP)(seed treated with HP)was observed during the first 12 h of *** control’s(Jangdae seed)germination increased sharply and reached a maximum germination rate of 53.2%on the 21st day in the *** 21 d,germination of all priming treatments ranged from 13.8%to 26.3%.The palisade layer(PAL)after priming treatment was more damaged than that of the ***,a significant difference on the acid level between the control and PEG priming treatment(phowed that application of fungicide after priming optimizes seed germination and ***,it is recommended to invigorate the kenaf seed before planting.

Characteristics of Kenaf(Hibiscus cannabinus L.) Mutants Induced by Gamma-Ray

Kenaf (Hibiscus cannabinus L.) is known as a multipurpose crop producing biomass for energy thus, the study was conducted to select the elite mutants with higher biomass produced through mutation. Obtained from the National Agricultural Genetic Resources Center (NAGRC) in Korea, the seeds of the original natural resource named IT202801 (control) were irradiated with 250 Gy gamma-ray. The agronomic performances, genetic variation and histological analysis of the elite mutant lines were investigated at M2 generation in comparison with the control. Significant differences were observed from the previous three evaluations of the eight mutants. Nine plants, including the control and eight mutants, had little difference in the number of flowering days from July 23 to July 25. The leaf shape of the control was entire, while the leaf shape of the mutant was palmate. Of the eight lines, M2IT20-5 showed better performance in regard to diameter, dry weight and seed weight per plant. The random amplified polymorphic DNA (RAPD) analysis produced a genetic diversity of 72.7% within the genotypes used. A dendrogram was constructed based on the RAPD fragments. Differentiation was demonstrated between the control and kenaf mutants. Taken together, the mutant line, M2IT20-5, can be useful as a resource for high biomass production.
