Role of mitophagy in head and neck squamous cell carcinoma:Prognosis and immune insights

Purpose:Head and neck squamous cell carcinoma(HNSCC) treatment is facing clinical *** in cancer cells is related to the tumor high energetic *** tumor immune microenvironment(TME) strongly influences clinical outcomes and treatment ***,the correlation of mitophagy and TME remains unknown in ***:Based on machine learning,a prognostic multigene signature was built with mitophagy-related differentially expressed genes(MPGs),which was associated with TME by gene set enrichment analysis(GSEA),in the TCGA ***,we systematically correlated risk signature with immunological characteristics in TME,which included immune checkpoints,tumor-infiltrating immune cells(TIICs),*** further invalidate CSNK2A2,we employed immunohistochemistry to examine its ***:MPGs-related prognostic model showed good prediction *** who had high-risk scores had significantly shorter progression-free survival(PFS) and overall survival(OS) than those with low-risk scores,according to the results of the survival analysis(p<0.0001).The CD8+ T cells infiltrated less in samples with higher risk *** immunological characteristic markers were expressed at higher levels in the low-risk ***,immune therapy might be effective for the low-risk subtype of HNSCC patients(p<0.001).Samples with higher risk scores were more sensitive to ***2A2was validated to be higher expressed in HNSCC tissues,according to ***:We have constructed a prognostic signature and provided innovative insights that may improve HNSCC management,which might give a more precise prognostic ***2A2 might be a novel biomarker to predict immune efficacy.

Background:We aimed to evaluate the optimal management for elderly patients with nasopharyngeal carcinoma(NPC) with intensity-modulated radiotherapy(IMRT).Methods:301 elderly patients with NPC diagnosed from 2015 to 2019 were *** survival(OS) was the primary *** univariate and multivariate Cox regression analysis were preformed to identify potential prognostic *** recursive partitioning analysis(RPA) was used for risk ***-Meier survival curves were applied to evaluate the survival endpoints,and log-rank test was used to assess the difference between *** prognostic index(PI) was constructed to further predict patients’ prognoses displayed by nomogram *** area under the receiver operating characteristic(ROC) curves(AUC) and the calibration curves were applied to assess this ***:Based on RPA-based risk stratification,we demonstrated that elderly NPC patients who were treated with IC followed by RT had similar OS compared with those with IC combined with CCRT in the intermediate-(stage Ⅰ-Ⅲ and pre-treatment EBV > 1840 copies/ml) and high-risk groups(stage IV).IMRT alone maybe an optimal treatment option in the low-risk group(stage Ⅰ-Ⅲ with pre-treatment EBV < 1840 copies/ml).We established an integrated PI which was indicted with stronger prognostic power than it based on each separately for elderly NPC(The AUC of PI was 0.87,0.81,and 0.79 for 1-,3-,5-year prediction of OS,respectively).Conclusion:We present a robust model for clinical stratification which could guide individual therapy for elderly NPC patients.

Background:It remains controversial that the application of chemotherapy has an impact on recurrent nasopharyngeal carcinoma(r NPC) patients with salvage ***,we aimed to evaluate treatment outcomes of r NPC patients and derive a prognostic model to assess the benefit of chemotherapy in patients with ***:Totally 340 r NPC patients treated with salvage intensity-modulated radiotherapy(IMRT) or chemoradiotherapy(CRT) from October 2006 to September 2019 were included in this *** survival(OS) was the primary ***-Meier method was employed to detect the prognostic difference with Log-rank *** Cox regression analysis was performed to explore the potential prognostic factors while the multivariate Cox analysis was used to identify candidate variables for the prognostic model of ***:The 5-year actuarial rates of OS,progression-free survival,loco-regional progression-free survival,and distant metastases-free survival didn’t show significant difference between the IMRT and CRT groups(ps > 0.05).Age at recurrence was found to be the only independent prognostic factor for OS(HR = 2.076,95% CI 1.490-2.894,p < 0.001).Three factors(age at recurrence,r Stage,and initial treatment) were identified for the prognostic *** found that r NPC patients suffered poor OS in the high-risk group(p < 0.001).In the low-risk group,CRT failed to induce survival benefits when compared to RT alone(p = 0.036).Conclusions:Salvage RT combined with chemotherapy cannot improve survival outcomes for r *** novel clinical trials should be explored to develop individualized strategies to improve survival and minimize toxicities.

Recombinant adenovirus-p53(Gendicine) sensitizes a pancreatic carcinoma cell line to radiation

Objective： In this study, we examine the effects of recombinant adenovirus-p53 （rAd-p53） on the pancreatic carcinoma cell line SW1990. Specifically, we determine if expression of rAd-p53 sensitizes these cells to radiation. Methods： Following transfection of SW1990 cells with rAd-p53, we measured expression of P53, P21 and Bax by immunocytochemistry. Both transfected and control cell lines were irradiated with a range of doses, and the survival fractions （SF） were calculated. Dose survival cttrves were constructed and modeled for comparison. Results： Transfection of SW1990 cells with rAd-p53 resulted in increased expression of P53, P21 and Bax in a time-dependent manner. At 96 h after transfection, 89.92% of cells expressed P53, 56.8% expressed P21, and 76.50% expressed Bax. The SF following radiation was lower in the rAd-p53 transfected cells compared to the control cells, suggesting that rAd-p53 sensitizes SW1990 cells to radiation （Do for the experimental and control groups was 2.199 and 2.462, respectively）. Conclusions： Use of the adenoviral vector is an effective means of transfecting SW1990 cells with wild-type P53, and this sensitizes the cell line to irradiation. This work suggests that combining rAd-p53 with radiation therapy in pancreatic cancer may be therapeutically beneficial.

Room temperature sodium-sulfur(Na-S)batteries,known for their high energy density and low cost,are one of the most promising next-generation energy storage ***,the polysulfide shuttling and uncontrollable Na dendrite growth as well as safety issues caused by the use of organic liquid electrolytes in Na-S cells,have severely hindered their ***-state electrolytes instead of liquid electrolytes are considered to be the most direct and effective solution to solve the above ***,its practical application is still greatly challenged due to the poor interfacial compatibility between the all-solid-state electrolytes and the anode/cathode,ionic conductivity,and the shuttle effect caused by the presence of liquid phase in the quasi-solid-state *** paper presents a comprehensive review of solid-state Na-S batteries from the perspective of regulating interfacial compatibility and improving ionic conductivity as well as suppressing polysulfide *** to different components,solid-state electrolytes were divided into five categories:solid inorganic electrolytes,solid polymer electrolytes,polymer/inorganic solid hybrid electrolytes,gel polymer electrolytes,and liquid–solid inorganic hybrid ***,the prospect of developing high performance solid-state electrolytes to improve the cycling stability of room temperature Na-S cells is envisaged.

Enhanced service stability and thermoelectric performance in Cu_(2)−δSebased composites

Thermoelectric(TE)materials can directly convert the industrial waste heat into useful electricity based on the Seebeck effect,providing a promising way to tackle the environmental pollution and global energy crisis[1].In the past several decades,many new high-performance TE materials have been discovered to enrich the TE *** them,Cu_(2)−δSe-based TE materials,which are in line with the“phonon-liquid electron-crystal”concept,have sparked widespread interest due to the high TE figure-of-merit(ZT)and the combination of elements that are nontoxic and earth-abundant[2].In 2012,Liu et al.[3]reported a ZT of 1.5 at 1000 K in the stoichiometric Cu_(2)*** alloying S at the Se-site,the ZT was enhanced to around 2.0 at 1000 K[4].Via incorporating carbon nanotubes,the ZT was further enhanced to 2.4 at 1000 K[5].Moreover,Yang et al.[6]and Zhou et al.[7]reported that the ZT of Cu_(2)Se/BiCuSeO composites can reach 2.7–2.8 around 1000 K.

y-Tuning: an efficient tuning paradigm for large-scale pre-trained models via label representation learning

With current success of large-scale pre-trained models(PTMs),how efficiently adapting PTMs to downstream tasks has attracted tremendous attention,especially for PTMs with billions of *** work focuses on designing parameter-efficient tuning paradigms but needs to save and compute the gradient of the whole computational *** this paper,we propose y-Tuning,an efficient yet effective paradigm to adapt frozen large-scale PTMs to specific downstream tasks.y-Tuning learns dense representations for labels y defined in a given task and aligns them to fixed feature *** computing the gradients of text encoder at training phrase,y-Tuning is not only parameterefficient but also *** results show that for DeBERTaxxL with 1.6 billion parameters,y-Tuning achieves performance more than 96%of full fine-tuning on GLUE Benchmark with only 2%tunable parameters and much fewer training costs.
