Evidence Based Validation of Indian Traditional Medicine–Way Forward

Evidence based validation of the ethno-pharmacological claims on traditional medicine(TM) is the need of the day for its globalization and reinforcement. Combining the unique features of identifying biomarkers that are highly conserved across species, this can offer an innovative approach to biomarker-driven drug discovery and development. TMs are an integral component of alternative health care systems. India has a rich wealth of TMs and the potential to accept the challenge to meet the global demand for them. Ayurveda, Yoga, Unani, Siddha and Homeopathy(AYUSH) medicine are the major healthcare systems in Indian Traditional Medicine. The plant species mentioned in the ancient texts of these systems may be explored with the modern scientific approaches for better leads in the healthcare. TM is the best sources of chemical diversity for finding new drugs and leads. Authentication and scientific validation of medicinal plant is a fundamental requirement of industry and other organizations dealing with herbal drugs. Quality control(QC) of botanicals, validated processes of manufacturing, customer awareness and post marketing surveillance are the key points, which could ensure the quality, safety and efficacy of TM. For globalization of TM, there is a need for harmonization with respect to its chemical and metabolite profiling, standardization, QC, scientific validation,documentation and regulatory aspects of TM. Therefore, the utmost attention is necessary for the promotion and development of TM through global collaboration and co-ordination by national and international programme.

Monte Carlo study of B-cell receptor clustering mediated by antigen crosslinking and directed transport

It is known from experiments that in the presence of soluble antigen,B-cell receptors(BCRs)assemble into microclusters and then collect into a macrocluster known as a‘cap’.However,the mechanisms of BCR cluster formation during recognition of soluble antigens remain *** previous work,we demonstrated that effective intrinsic attractions among BCRs can lead to the formation of small microclusters of BCR *** effective intrinsic attractions could be caused by multivalent antigen binding,association with lipid rafts,or other biochemical *** the present study,we have developed and studied a Monte Carlo model of BCR clustering mediated by explicit binding and crosslinking of soluble bivalent *** crosslinking is shown to microcluster BCRs in an affinity-dependent manner and also in a biologically relevant timescale;however,antigen crosslinking alone does not appear to be sufficient for the formation of a single macrocluster of receptor *** show that directed transport of BCRs is needed to drive the formation of large *** constructed a simple model of directed transport,where BCR molecules diffuse towards the largest cluster or towards a random BCR microcluster,which results in a single macrocluster of receptor *** mechanisms for both types of directed transport are compared using network-based *** also develop and use appropriate network measures to analyze the effect of BCR and antigen concentration on BCR clustering,the stability of the formed clusters over time and the size of BCR–antigen crosslinked chains.

Molecular dynamics study on growth of carbon dioxide and methane hydrate from a seed crystal

In the current work,molecular dynamics simulation is employed to understand the intrinsic growth of carbon dioxide and methane hydrate starting from a seed crystal of methane and carbon dioxide *** comparison was carried out because it has relevance to the recovery of methane gas from natural gas hydrate reservoirs by simultaneously sequestering a greenhouse gas like *** seed crystal of carbon dioxide and methane hydrate was allowed to grow from a super-saturated mixture of carbon dioxide or methane molecules in water *** different concentrations(1:6 and 1:8.5)of CO2/CH4 molecules per water molecule were chosen based on gas–water composition in hydrate *** molecular level growth as a function of time was investigated by all atomistic molecular dynamics simulation under suitable temperature and pressure range which was well above the hydrate stability zone to ensure significantly faster growth *** concentration of CO2 molecules in water played a significant role in growth kinetics,and it was observed that maximizing the CO2 concentration in the aqueous phase may not result in faster growth of CO2 *** the contrary,methane hydrate growth was independent of methane molecule concentration in the aqueous *** have validated our results by performing experimental work on carbon dioxide hydrate where it was seen that under conditions appropriate for liquid CO2,the growth for carbon dioxide hydrate was very slow in the beginning.

Modeling the gene delivery process of the needle array-based tissue nanotransfection

Hollow needle array-based tissue nanotransfection(TNT)presents an in vivo transfection approach that directly translocate exogeneous genes to target tissues by using electric *** this work,the gene delivery process of TNT was simulated and experimentally *** adopted the asymptotic method and cell-array-based model to investigate the electroporation behaviors of cells within the skin *** distribution of nonuniform electric field across the skin results in various electroporation behavior for each *** underneath the hollow microchannels of the needle exhibited the highest total pore numbers compared to others due to the stronger localized electric *** percentage of electroporated cells within the skin structure,with pore radius over 10 nm,increases from 25%to 82%as the applied voltage increases from 100 to 150 V/***,the gene delivery behavior across the skin tissue was investigated through the multilayer-stack-based *** delivery distance increased nonlinearly as the applied voltage and pulse number increased,which mainly depends on the diffusion characteristics and electric conductivity of each *** was also found that the skin is required to be exfoliated prior to the TNT procedure to enhance the delivery *** work provides the foundation for transition from the study of murine skin to translation use in large animals and human settings.

Discrimination of membrane antigen affinity by B cells requires dominance of kinetic proofreading over serial engagement

B-cell receptor signaling in response to membrane-bound antigen increases with antigen affinity,a process known as affinity *** use computational modeling to show that B-cell affinity discrimination requires that kinetic proofreading predominate over serial *** find that if B-cell receptors become signaling-capable immediately upon antigen binding,which results in decreasing serial engagement as affinity increases,then increasing affinity can lead to weaker ***,antigen must stay bound to B-cell receptors for a threshold time of several seconds before becoming signaling-capable,a process similar to kinetic *** process overcomes the loss in serial engagement due to increasing antigen affinity,and replicates the monotonic increase in B-cell signaling with increasing affinity that has been observed in B-cell activation *** finding matches well with the experimentally observed time(,20 s)required for the B-cell receptor signaling domains to undergo antigen and lipid raft-mediated conformational changes that lead to Src-family kinase *** hypothesize that the physical basis for a threshold time of antigen binding might lie in the formation timescale of B-cell receptor *** time required for dimer formation decreases with increasing antigen affinity,thereby resulting in shorter threshold antigen binding times as affinity *** an affinity-dependent kinetic proofreading requirement results in affinity discrimination very similar to that observed in biological experiments.B-cell affinity discrimination is critical to the process of affinity maturation and the production of high-affinity antibodies,and thus our results have important implications in applications such as vaccine design.

Non-Gaussian normal diffusion in low dimensional systems

Brownian particles suspended in disordered crowded environments often exhibit non-Gaussian normal diffusion(NGND),whereby their displacements grow with mean square proportional to the observation time and non-Gaussian *** distributions appear to decay almost exponentially according to“universal”laws largely insensitive to the observation *** effect is generically attributed to slow environmental fluctuations,which perturb the local configuration of the suspension *** investigate the microscopic mechanisms responsible for the NGND phenomenon,we study Brownian diffusion in low dimensional systems,like the free diffusion of ellipsoidal and active particles,the diffusion of colloidal particles in fluctuating corrugated channels and Brownian motion in arrays of planar convective *** appears to be a transient effect related to the time modulation of the instantaneous particle’s diffusivity,which can occur even under equilibrium ***,we propose to generalize the definition of NGND to include transient displacement distributions which vary continuously with the observation *** this purpose,we provide a heuristic one-parameter function,which fits all time-dependent transient displacement distributions corresponding to the same diffusion ***,we reveal the existence of low dimensional systems where the NGND distributions are not leptokurtic(fat exponential tails),as often reported in the literature,but platykurtic(thin sub-Gaussian tails),i.e.,with negative excess *** actual nature of the NGND transients is related to the specific microscopic dynamics of the diffusing particle.

Thermal maturity evaluation using Raman spectroscopy for oil shale samples of USA:comparisons with vitrinite reflectance and pyrolysis methods

Thermal maturity is commonly assessed by various geochemical screening methods(e.g.,pyrolysis and organic petrology).In this contribution,we attempt to establish an alternative approach to estimating thermal maturity with Raman spectroscopy,using 24 North American oil shale samples with thermal maturity data generated by vitrinite reflectance(VRo%)and pyrolysis(Tmax)-based maturity calculation(VRe%).The representative shale samples are from the Haynesville(East Texas),Woodford(West Texas),Eagle Ford and Pearsall(South Texas)Formations,as well as Gothic,Mancos,and Niobrara Formation shales(all from Colorado).The Raman spectra of disordered carbonaceous matter(D1 and G bands separation)of these samples were directly obtained from the rock chips without prior sample *** the Gaussian and Lorentzian distribution approach,thermal maturities from VR were correlated with carbon G and *** found that the Raman band separation(RBS)displayed a better correlation for equivalent VRe%than vitrinite reflectance VRo%.The RBS(D1–G)distance versus total organic carbon,free hydrocarbons from thermal extraction(S1),and the remaining hydrocarbon generating potential(S2)indicate that the RBS(D1–G)distance is also related to kerogen *** presented here from three methods of maturity determination of shale demonstrate that Raman spectroscopy is a quick and valid approach to thermal maturity assessment.
