Design of dual targeting immunomicelles loaded with bufalin and study of their anti-tumor effect on liver cancer

Objective: Bufalin is an effective drug for the treatment of liver cancer. But its high toxicity, poor watersolubility, fast metabolism and short elimination half-life limit its use in tumor treatment. How to make the drug accumulate in the tumor and reduce side effects while maintaining its efficacy are urgent problems to be solved. The goal of this study is to solve these ***: A copolymer with tunable poly-N-isopropylacrylamide and polylactic acid was designed and synthesized. The corresponding dual targeting immunomicelles(DTIs) loaded with bufalin(DTIs–BF)were synthesized by copolymer self-assembly in an aqueous solution. The size and structure of DTIs–BF were determined by ZetaSizer Nano-ZS and transmission electron microscopy. Then, its temperature sensitivity, serum stability, critical micelle concentration(CMC), entrapment efficiency(EE), drug release and non-cytotoxicity of blank block copolymer micelles(BCMs) were evaluated. Next, the effects of DTIs–BF on cellular uptake, cytotoxicity, and tumor cell inhibition were evaluated. Finally, the accumulation of DTIs–fluorescein isothiocyanate(FITC) and the in vivo anti-tumor effect were observed using an interactive video information ***: DTIs–BF had a small size, spherical shape, good temperature sensitivity, high serum stability, low CMC, high EE, and slow drug release. The blank BCMs had very low cytotoxicity. Compared with free bufalin, the in vitro cellular internalization and cytotoxicity of DTIs–BF against SMMC-7721 cells were significantly enhanced, and the effects were obviously better at 40 ℃ than 37 ℃. In addition, the therapeutic effect on SMMC-7721 cells was further enhanced by the programmed cell death specifically caused by bufalin. Effects of titanium on microstructure and mechanical properties of ZnAl4Y alloy

The effect of Ti on the microstructure and mechanical properties of zinc-aluminum ZnAl4Y alloy has been investigated in this work. Small amount of Ti was added into ZnAl4Y alloy in the form of Al-10wt.%Ti alloy. The results show that Ti addition into ZnAl4Y alloy refines the primary η-Zn phase and increases the amount of η-Zn + α-Al eutectic structure. There exists a ternary T phase in Zn-Al-Ti alloy. Fine TiAl3 particles and Ti-Zn compounds can serve as the nucleation sites of the α-Al and η-Zn phase, respectively, resulting in the refining of the microstructure of ZnAl4Y alloy. Ti addition changes the fracture characteristics of ZnAl4Y alloy. With 0.05wt%Ti, the fracture surface of the alloy shows a lot of dimples and tearing ridges connecting the microscopic dimples, which is mainly ductile fracture morphology. Ti addition into ZnAl4Y alloy also increases the mechanical properties of the alloy. When the content of Ti is 0.05wt.%, the ZnAl4Y alloy possesses the best comprehensive mechanical properties.

Individualized peri-operative fluid therapy facilitating early-phase recovery after liver transplantation

Electrolytic Passivation of Nitinol Shape Memory Alloy in Different Electrolytes

The corrosion behavior of the nitinol alloy was studied in various corrosion media of different Cl^- ion concentrations. The results demonstrate that the Cl^- ion concentration has significant influences on the corrosion behavior of the nitinol alloy. In order to enhance the corrosion resistance, protective films were generated on the surface of the nitinol alloy by means of the electrochemical passivation method, for which five different electrolytic solutions were investigated. The surface analysis indicates full growth of all samples passivated in the different electrolytic solutions with layers, however, showing different morphological features. Without any defects like micro-cracks and pores, the surface of the samples passivated in the molybdate solution turns out smoother and denser than those passivated in other solutions. It is shown that the electro-chemical passivation will reduce Ni content but increase Ti content in the surface, reaching the Mole ratio of Ti：Ni = 9.01：1 on the outermost surface. Potentiodynamic polarization test demonstrates that the samples electrochemically passivated in the molybdate solution present a significant increase in breakdown potential due to titanium enrichment on the outermost surface.

Complexation between DNA and Peptides with Precisely Controlled Charge Density and Distribution

Using three designed peptides with precisely-controlled charge density and three types of DNAs with different length and flexibility, the effect of charge density on the formation of PEC was studied. Highly charged（KKKK）5 interacts strongly with 21 bp ds DNA to form large complex, followed by precipitation; while the medium charged（KGKG）5 only form complex with 21 bp ds DNA at proper ＋/- charge ratios; and no prominent complex between weakly charged（KGGG）5 and 21 bp ds DNA is observed at the same conditions. Similar trend is observed when the peptides form complex with 2000 bp DNA or 21 nt ssD NA. It is also found that the complex formed by adding peptide to DNA is in random coil conformation, but the complex prepared by the inverse order is in molten globule state. Re-dissolution of the complex occurs only when DNA is added to peptides with similar or shorter length.

916 MHz electromagnetic field exposure affects rat behavior and hippocampal neuronal discharge

Wistar rats were exposed to a 916 MHz, 10 W/m2 mobile phone electromagnetic field for 6 hours a day 5 days a week. Average completion times in an eight-arm radial maze were longer in the exposed rats than control rats after 4-5 weeks of exposure. Error rates in the exposed rats were greater than the control rats at 6 weeks. Hippocampal neurons from the exposed rats showed irregular firing patterns during the experiment, and they exhibited decreased spiking activity 6-9 weeks compared with that after 2 5 weeks of exposure. These results indicate that 916 MHz electromagnetic fields influence leaming and memory in rats during exposure, but long-term effects are not obvious.

sudden cardiac arrest in a patient with epilepsy induced by chronic inflammation on the cerebral surface

The present study analyzed a patient with epilepsy due to chronic inflammation on the cerebral surface underwent sudden cardiac arrest. Paradoxical brain discharge, which occurred prior to epileptic seizures, induced a sudden cardiac arrest. However, when the focal brain pressure was relieved, cardiac arrest disappeared. A 27-year-old male patient underwent pre-surgical ram monitoring for 160 hours. During monitoring, secondary tonic-clonic seizures occurred five times. A burst of paradoxical brain discharges occurred at 2-19 seconds （mean 8 seconds） prior to epileptic seizures. After 2-3 seconds, sudden cardiac arrest occurred and lasted for 12-22 seconds （average 16 seconds）. The heart rate subsequently returned to a normal rate. Results revealed arachnoid pachymenia and adhesions, as well as mucus on the focal cerebral surface, combined with poor circulation and increased pressure, lntracranial electrodes were placed using surgical methods. Following removal of the arachnoid adhesions and mucus on the local cerebral surface, paradoxical brain discharge and epileptic seizures occurred three times, but sudden cardiac arrest was not recorded during 150-hour monitoring. Post-surgical histological examination indicated meningitis. Experimental findings suggested that paradoxical brain discharge led to cardiac arrest instead of epileptic seizures; the insult was associated with chronic inflammation on the cerebral surface, which subsequently led to hypertension and poor blood circulation in focal cerebral areas.
