Influence of biological sex and exercise on murine cardiac metabolism

Although the structural and functional effects of exercise on the heart are well established,the metabolic changes that occur in the heart during and after exercise remain *** this study,we used metabolomics to assess time-dependent changes in the murine cardiac metabolome following 1 session of treadmill *** the exercise bout,we also recorded blood lactate,glucose,and ketone body levels and measured cardiac mitochondrial *** both male and female mice,moderate-and high-intensity exercise acutely increased blood lactate *** both sexes,low-and moderate-intensity exercise augmented circulating 3-hydroxybutryrate levels immediately after the exercise bout;however,only in female mice did high-intensity exercise increase 3-hydroxybutyrate levels,with significant increases occurring 1 h after the exercise *** metabolomics analyses of sedentary female and male hearts suggest considerable sex-dependent differences in basal cardiac metabolite levels,with female hearts characterized by higher levels of pantothenate,pyridoxamine,homoarginine,tryptophan,and several glycerophospholipid and sphingomyelin species and lower levels of numerous metabolites,including acetyl coenzyme A,glucuronate,gulonate,hydroxyproline,prolyl-hydroxyproline,carnosine,anserine,and carnitinylated and glycinated species,as compared with male *** after a bout of treadmill exercise,both male and female hearts had higher levels of corticosterone;however,female mice showed more extensive exercise-induced changes in the cardiac metabolome,characterized by significant,time-dependent changes in amino acids(e.g.,serine,alanine,tyrosine,tryptophan,branched-chain amino acids)and the ketone body *** from experiments using isolated cardiac mitochondria suggest that high-intensity treadmill exercise does not acutely affect respiration or mitochondrial coupling;however,female cardiac mitochondria demonstrate generally higher adenosi

Individualization of Data-Segment-Related parameters for Improvement of EEG Signal Classification in Brain-Computer Interface

In electroencephalogram (EEG) modeling techniques, data segment selection is the first and still an important step. The influence of a set of data-segment-related parameters on feature extraction and classification in an EEG-based brain-computer interface (BCI) was studied. An auto search algorithm was developed to study four datasegment-related parameters in each trial of 12 subjects’ EEG. The length of data segment (LDS), the start position of data (SpD) segment, AR order, and number of trials (NT) were used to build the model. The study showed that, compared with the classification ratio (CR) without parameter selection, the CR was increased by 20% to 30% with proper selection of these data-segment-related parameters, and the optimum parameter values were subject-dependent. This suggests that the data-segment-related parameters should be individualized when building models for BCI.

The authors,one from China and one from the United States,present a theoretical framework for understanding the discursive fields of citizenship education as composed,in large part,of the discourses of nationalism,globalization,and *** framework is illustrated by examples from citizenship education in China and the United *** education in these examples is largely influenced by the discourse of *** discursive fields are fractured,context-specific,and *** conclusion,the authors call for awareness of how these discourses operate,and propose that the discourses of globalization and cosmopolitanism merge and strengthen within citizenship *** effect could be a new citizenship education that is responsive to the current needs of local and global democratic communities.

Sensitivity Analysis of Key Input parameters in Microscopic Simulation of Congested and Uncongested Arterial Signals

This study involved investigating the sensitivity of Measures of Effectiveness （MOEs） to different simulation initialization time （7, 10, and 13 minutes）; observing the trend of variation of MOEs with increasing simulation runs; and identifying the major contributors of variation in MOEs using CORSIM and SimTraffic. The results showed that （1） the MOEs of a simulated intersection approaches were indeed sensitive to initialization times; （2） the variation within MOEs reached a steady state with increased number of simulation runs, while CORSIM required at least 50 simulation runs, SimTraffic required even higher number of runs for congested approaches; （3） lane changing and gap acceptance parameters play a major role as a source of variation of MOEs （delay/vehicle） in CORSIM and SimTraffic respectively.

Genetic Relationships Among Soluble Carbohydrates, Anthocyanins and Growth Characteristics in Leymus (Gramineae) Detected with Molecular Markers

Low-temperature soluble carbohydrate accumulations are commonly associated with anthocyanin coloration, attenuated growth and cold adaptation of cool-season grasses. The vrn-1 gene has potent effects on vernalization requirement, growth, and soluble carbohydrate accumulations of the winter-annual Triticeae species. Two hundred and four unmapped AFLp markers and genome-specific DNA markers genetically linked to the vrn-1 gene were used to detect QTL controlling soluble carbohydrate accumulations, anthocyanin coloration and growth characteristics in a segregating population derived from open pollinated Leymus cinereus x L. triticoides hybrids. These perennial Triticeae grasses are distinguished by adaptation and growth habit. As expected, positive trait correlations and pleiotropic gene effects were detected for soluble carbohydrate accumulations and anthocyanin coloration. Likewise, positive trait correlations and pleiotropic gene effects were detected for tillering, leaf development, leaf growth, regrowth and rhizome spread. However, soluble carbohydrate accumulations were not associated with attenuated growth. In fact, several DNA marker alleles, including one near vrn-Ns1, had positive effects on soluble leaf carbohydrate concentrations and low temperature growth. The corresponding DNA marker near vrn-Ns1 had more specific effects on tillering. We speculate that vrn-1 exerts quantitative effects on low-temperature soluble leaf carbohydrate accumulations and growth habit of the perennial Leymus. However, a number of other DNA markers displayed highly significant effects on soluble carbohydrate accumulations and various growth characteristics. Findings indicate that anthocyanin coloration may be a useful phenotypic marker for soluble carbohydrate accumulation. Although variation for soluble carbohydrates was not associated with attenuated growth in this population, this trait was under genetic control.

A boronic acid-based anthracene fluorescent probe was functionalised with an acrylamide unit to incorporate into a hydrogel system for monosaccharide *** solution,the fluorescent probe displayed a strong fluorescence turn-on response upon exposure to fructose,and an expected trend in apparent binding constants,as judged by a fluorescence response where D-fructose>D-galactose>D-mannose>*** hydrogel incorporating the boronic acid monomer demonstrated the ability to detect monosaccharides by fluorescence with the same overall trend as the monomer in solution with the addition of D-fructose resulting in a 10-fold enhancement(≤0.25 mol/L).

Identification of neuron selective androgen receptor inhibitors

AIM To identify neuron-selective androgen receptor(AR) signaling inhibitors, which could be useful in the treatment of spinal and bulbar muscular atrophy(SBMA), or Kennedy's disease, a neuromuscular disorder in which deterioration of motor neurons leads to progressive muscle weakness. METHODS Cell lines representing prostate, kidney, neuron, adipose, and muscle tissue were developed that stably expressed the CFp-AR-YFp FRET reporter. We used these cells to screen a library of small molecules for cell typeselective AR inhibitors. Secondary screening in luciferase assays was used to identify the best cell-type specific AR inhibitors. The mechanism of action of a neuronselective AR inhibitor was examined in vitro using luciferase reporter assays, immunofluorescence microscopy, and immunoprecipitations. Rats were treated with the most potent compound and tissue-selective AR inhibition was examined using RT-q pCR of AR-regulated genes and *** We identified the thiazole class of antibiotics as com-pounds able to inhibit AR signaling in a neuronal cell line but not a muscle cell line. One of these antibiotics, thiostrepton is able to inhibit the activity of both wild type and polyglutamine expanded AR in neuronal GT1-7 cells with nanomolar potency. The thiazole antibiotics are known to inhibit FOXM1 activity and accordingly, a novel FOXM1 inhibitor FDI-6 also inhibited AR activity in a neuron-selective fashion. The selective inhibition of AR is likely indirect as the varied structures of these compounds would not suggest that they are competitive antagonists. Indeed, we found that FOXM1 expression correlates with cell-type selectivity, FOXM1 co-localizes with AR in the nucleus, and that sh RNA-mediated knock down of FOXM1 reduces AR activity and thiostrepton sensitivity in a neuronal cell line. The Gp130 cytokine receptor subunit encoded by IL6ST is the shared receptor for ten cytokines of the IL-6 family. We describe a homozygous non-synonymous variant in IL6 ST(p.R281 Q) in a patient with craniosynostosis and retained deciduous teeth. We characterize the impact of the variant on cytokine signaling in vitro using transfected cell lines as well as primary patient-derived cells and support these findings using a mouse model with the corresponding genome-edited variant Il6 st p.R279 Q. We show that human Gp130 p.R281 Q is associated with selective loss of IL-11 signaling without affecting IL-6, IL-27, OSM, LIF, CT1, CLC, and CNTF signaling. In mice Il6 st p.R279 Q lowers litter size and causes facial synostosis and teeth abnormalities. The effect on IL-11 signaling caused by the Gp130 variant shows incomplete penetrance but phenocopies aspects of IL11 RA deficiency in humans and mice. Our data show that a genetic variant in a pleiotropic cytokine receptor can have remarkably selective defects.
