Anti-CD163-dexamethasone conjugate inhibits the acute phase response to lipopolysaccharide in rats

AIM: To study the effect of a new anti-CD163-dexamethasone conjugate targeting activated macrophages on the hepatic acute phase response in rats. METHODS: Wistar rats were injected intravenous with either the CD163 targeted dexamethasone-conjugate(0.02 mg/kg) or free dexamethasone(0.02 or 1 mg/kg) 24 h prior to lipopolysaccharide(LPS)(2.5 mg/kg intraperitoneal). We measured plasma concentrations of tumour necrosis factor-a(TNF-a) and interleukin 6(IL-6) 2 h post-LPS and liver m RNAs and serum concentrations of the rat acute phase protein a-2-macroglobulin(a-2-M) 24 h after LPS. Also, plasma concentrations of alanine aminotransferase and bilirubin were measured at termination of the study. Spleen weight served as an indicator of systemic steroid ***: The conjugate halved the a-2-M liver m RNA(3.3 ± 0.6 vs 6.8 ± 1.1, P < 0.01) and serum protein(201 ± 48 μg/mL vs 389 ± 67 μg/mL, P = 0.04) after LPS compared to low dose dexamethasone treated animals, while none of the free dexamethasone doses had an effect on liver m RNA or serum levels of a-2-M. Also, the conjugate reduced TNF-a(7208 ± 1977 pg/mL vs 21583 ± 7117 pg/mL, P = 0.03) and IL-6(15685 ± 3779 pg/mL vs 25715 ± 4036 pg/mL, P = 0.03) compared to the low dose dexamethasone. The high dose dexamethasone dose decreased the spleen weight(421 ± 11 mg vs 465 ± 12 mg, P < 0.05) compared to controls, an effect not seen in any other ***: Low-dose anti-CD163-dexamethasone conjugate effectively decreased the hepatic acute phase response to LPS. This indicates an anti-inflammatory potential of the conjugate in vivo.

Although there currently exist many different types of ＂smart＂ materials, those based on photoresponsive transformations are especially attractive. In this review, we focus on a relatively new class of photochromic molecules based on the photochemistry of chelate organoborates, which form intensely colored, base-stabilized *** efforts to exploit the reactivity of these systems are summarized, and future prospects in materials science discussed.

Molecular characterization and expression patterns of Nanog gene validating its involvement in the embryonic development and maintenance of spermatogonial stem cells of farmed carp,Labeo rohita

Background: The homeobox containing transcription factor Nanog plays crucial roles in embryonic development/proliferation and/or maintenance of spermatogonial stem cells(SSCs) via interacting with transcription factors such as Oct4 and Sox2 in mammals. However, knowledge of its exact mechanistic pathways remains unexploited. Very little is known about teleost Nanog. Information on the Nanog gene of farmed rohu carp(Labeo rohita) is lacking. We cloned and characterized the Nanog gene of rohu carp to understand the expression pattern in early developmental stages and also deduced the genomic organization including promoter ***: Rohu Nanog(LrNanog) cDNA comprised an open reading frame of 1,161 nucleotides bearing a structural homeodomain; whereas, the genomic structure contained four exons and three introns suggesting that it is homologous to mammalian counterparts. Phylogenetical y, it was closely related to freshwater counterparts. Protein sequence(386 AA of42.65 kDa) comparison revealed its low similarity with other vertebrate counterparts except that of the conserved homeodomain. Tissue distribution analysis revealed the existence of LrNanog transcripts only in adult gonads. The heightened abundances in the ovary and proliferating spermatogonia suggested its participations in maternal inheritance and male germ cell development. The potentiating abundances from fertilized egg onwards peaking at blastula stage vis-à-vis decreasing levels from gastrula stage onwards demonstrated its role in embryonic stem cell development. We also provided evidence of its presence in SSCs by western blotting analysis. Biochemical and histopathological changes in sheep fed different detoxified karanj(Pongamia glabra)seed cake as partial protein supplements

The study investigated the long-term effect of feeding processed solvent extracted karanj(Pongamia glabra) cake(SkC) on gross pathology and histopathological changes in some vital organs, and on the activities of serum enzymes in Jalauni lambs. Twenty-four male lambs were divided into 4 groups and allotted randomly to a soybean meal(SBM) based control(CON) and 3 treatment groups receiving concentrate mixtures, containing water washed(WW), 2.5% lime(LM) and 0.4% binder(BN) treated SkC replacing 50% nitrogen of SBM to meet the protein requirements. Blood was collected after 150 days from all the lambs and serum activities of aspartate aminotransferase(AST), alanine aminotransferase(ALT)and lactate dehydrogenase(LDH) were assayed. All lambs were sacrificed after 196 days, and vital organs screened for gross pathological lesions. The representative tissues of liver, intestine, parathyroid gland,testis, and epididymis were sampled, preserved in 10% formalin and processed to examine histopathological changes by staining with haematoxylin and eosin(HE). The serum enzyme activities of AST and ALT were similar in the treatments, but the activity of LDH was higher(P kC-fed groups than the control. The weight and size of the liver were decreased in BN group, while those of testes were decreased in both LM and BN groups. The histological sections of the testes and epididymis of lambs from LM group showed hypoplastic seminiferous tubules and empty ductules, respectively. The histological sections of the parathyroid gland in the same diet revealed smaller and non-secretory chief cells. The small intestine of lambs from BN group showed infiltration of mononuclear cells(MNC) in lamina propria with mild fibroplasia in intervillous areas. The histological section of liver from this group showed MNC infiltration in portal areas. Rapid,simple and stability-indicating determination of polyhexamethylene biguanide in liquid and gel-like dosage forms by liquid chromatography with diode-array detection

A rapid and simple method for the determination of potyhexamethylene biguanide （polyhexanide, PHMB） in liquid and gel-like pharmaceutical formulations by means of high performance liquid chromatography coupled to diode-array detection （HPLC DAD） was devel- oped. Best separation was achieved using a cyanopropyl bonded phase （Agilent Zorbax Eclipse XDB-CN column 4.6mm x 75 mm with particle size of 3.5 μm） as well as gradient elution consisting of acetonitrile/deionized water at a flow rate of 1.0 mL/min. The optimized and applied chromatographic conditions permitted separation of polyhexanide from interacting raatrix with subsequent detection at a wavelength of 235 nm with good sensitivity. The method validation was carried out with regard to the guidelines for analytical procedures demanded by the International Conference on Harmonisation （ICH）. Mean recoveries of 102% and 101% for gel-like as well as liquid preparations were obtained. Suitable repeatability as well as intermediate precision could bc achieved with limits of detection ≤0.004 mg/mL for both formulations, equivalent to ≤0.004% PHMB concerning sample preparation. Determination of PHMB was accomplished without tedious sample preparation. Interacting matrix could be eliminated by the chromatographicprocedure with excellent performance of system suitability. All analytical requirements were fulfilled permitting a reliable and precise determination of PHMB in pharmaceuticals. Further- more, the developed method was applied to stability testing of pharmaceutical preparations containing PHMB.

Dried blood spots,valid screening for viral hepatitis and human immunodeficiency virus in real-life

AIM To detect chronic hepatitis B(CHB),chronic hepatitis C(CHC) and human immunodeficiency virus(HIV) infections in dried blood spot(DBS) and compare these samples to venous blood sampling in *** We included prospective patients with known viral infections from drug treatment centers,a prison and outpatient clinics and included blood donors as negative controls. Five drops of finger capillary blood were spotted on filter paper,and a venous blood sample was obtained. The samples were analyzed for HBs Ag,antiHBc,anti-HBs,anti-HCV,and anti-HIV levels as well as subjected to a combined nucleic acid test(NAT) for HBV DNA,HCV RNA and HIV *** Samples from 404 subjects were screened(85 CHB,116 CHC,114 HIV and 99 blood donors). DBS had a sensitivity of > 96% and a specificity of > 98% for the detection of all three infections. NAT testing did not improve sensitivity,but correctly classified 95% of the anti-HCV-positive patients with chronic and past infections. Anti-HBc and anti-HBS showed low sensitivity in DBS(68% and 42%).CONCLUSION DBS sampling,combined with an automated analysis system,is a feasible screening method to diagnose chronic viral hepatitis and HIV infections outside of the health care system.
