Platelets are small, versatile blood cells that are critical for hemostasis/thrombosis. Local platelet accumulation is a known contributor to proinflammation in various disease states. However, the anti-inflammatory/immunosuppressive potential of platelets has been poorly explored. Here, we uncovered, unexpectedly, desialylated platelets (dPLTs) down-regulated immune responses against both platelet-associated and -independent antigen challenges. Utilizing multispectral photoacoustic tomography, we tracked dPLT trafficking to gut vasculature and an exclusive Kupffer cell-mediated dPLT clearance in the liver, a process that we identified to be synergistically dependent on platelet glycoprotein Ibα and hepatic Ashwell–Morell receptor. Mechanistically, Kupffer cell clearance of dPLT potentiated a systemic immunosuppressive state with increased anti-inflammatory cytokines and circulating CD4+ regulatory T cells, abolishable by Kupffer cell depletion. Last, in a clinically relevant model of hemophilia A, presensitization with dPLT attenuated anti-factor VIII antibody production after factor VIII ( infusion. As platelet desialylation commonly occurs in daily-aged and activated platelets, these findings open new avenues toward understanding immune homeostasis and potentiate the therapeutic potential of dPLT and engineered dPLT transfusions in controlling autoimmune and alloimmune diseases.

Vitamin D levels in subjects with or without chronic kidney disease among Veterans with diabetes in North East United States

AIM To evaluate the prevalence of vitamin D deficiency and its relation to diabetes and kidney disease in Veterans residing in the North East United States(VISN 2). METHODS In this retrospective study, we used data from the computerized patient record system at Stratton Veterans Administration Medical Center at Albany, NY(VHA) for those patients who had 25-hydroxyvitamin D levels and 1,25(OH) vitamin D levels measured between 2007 and 2010. We collected demographic information including age, sex, body mass index and race; clinical data including diabetes, hypertension and CAD; and laboratory data including calcium, creatinine and parathyroid hormone(PTH)(intact). Vitamin D deficiency is defined as a serum 25-hydroxyvitamin D level of less than 20 ng/mL(50 nmol/L), and insufficiency is defined as a serum 25-hydroxyvitamin D level of 20 to 30 ng/mL(50 to 75 nmol/L). RESULTS Data was available for approximately 68000 subjects. We identified 64144 subjects for analysis after exclusion of duplicates. Among them, 27098 had diabetes. Themean age of subjects with diabetes was 68 ± 11 with a mean body mass index(BMI) of 32 ± 7 and duration of diabetes of 5.6 ± 3.2 years. The mean 25(OH) vitamin D level among subjects with diabetes was 27 ± 11.6. There was no significant difference in 25(OH) vitamin D levels between subjects with diabetes and glomerular filtration rate(e-GFR) < 60 compared to those with e-GFR ≥ 60. As expected, subjects with e-GFR < 60 had significantly lower 1,25(OH) vitamin D levels and significantly elevated PTH-intact. Of the 64144 subjects, 580 had end-stage renal disease. Of those, 407 had diabetes and 173 did not. Vitamin D levels in both groups were in the insufficiency range and there was no significant difference irrespective of presence or absence of diabetes. Subjects with vitamin D levels less than 20 ng/mL had a higher BMI and elevated PTH, and higher HbA 1C levels compared to those with vitamin D levels more than 20 ng/mL. CONCLUSION We conclud

Extracurricular Reading for Children from Primary School Course in Bulgaria: Results of a Survey

The aim of the research is to obtain information about the current state of children’s extracurricular reading in Bulgaria—basis for further development of information skills and competencies. The study refers to the reading practices and interests of primary school students (7 - 11 years). It was implemented in the period March-June 2020 through electronic questionnaires filled out by the parents of 1169 students. The obtained data were processed with the software product SPSS, v19. A comparative analysis between the reading habits and preferences of children of different ages has been prepared. The followings are established: the thematic scope, the types of preferred readings, the motives for independent choice of reading, as well the preferred format, as the influence of the adults in the formation of the young reader. Particular attention is paid to reading in electronic format (e-books or other readings on the Internet).

Seismic Data Quality Control and Interpolation Using Principal Component Analysis

Commonly, seismic data processing procedures, such as stacking and prestack migration, require the ability to detect bad traces/shots and restore or replace them by interpolation, particularly when the seismic observations are noisy or there are malfunctioned components in the recording system. However, currently available trace/shot interpolation methods in the spatial or Fourier domain must deal with requirements such as evenly sampled traces/shots, infinite bandwidth of the signals, and linear seismic events. In this paper, we present a novel method, termed the E-S (eigenspace seismic) method, using principal component analysis (PCA) of the seismic signal to address the issue of reliable detection or interpolation of bad traces/shots. The E-S method assumes the existence of a correlation between the observed seismic entities, such as trace or shot gathers, making it possible to estimate one of these entities from all others for interpolation or seismic quality control. It first transforms a trace (or shot) gather into an eigenspace using PCA. Then in the eigenspace, it treats every trace as a point with its loading scores of PCA as its coordinates. Simple linear, bilinear, or cubic spline 1 dimensional (1D) interpolation is used to determine PCA loading scores for any arbitrary coordinate in the eigenspace, which are then used to construct an interpolated trace for the desired position in physical space. This E-S method works with either regular or irregular sampling and, unlike various other published methods, it is well-suited for band-limited seismic records with curvilinear reflection events. We developed related algorithms and applied these to processed synthetic and offshore seismic survey data with or without simulated noises to demonstrate their performance. By comparing the interpolated and observed seismic traces, we find that the E-S method can effectively assess the quality of the trace, and restore poor quality data by interpolation.
