Phytochemical studies and anti-ulcerative colitis effect of moringa oleifera seeds and Egyptian propolis methanol extracts in a rat model

Objective: To analyze the phytochemical constituents, and to explore potential protective effect of the methanol extract of moringa oleifera(m. oleifera) seeds and Egyptian propolis, each alone or concurrently administered on acetic acid-induced ulcerative colitis in rats. methods: Eight groups of 5 rats each were used: normal control group with distilled water, model group, two groups with m. oleifera seeds(100 and 200 mg/kg), two groups with propolis(50 and 100 mg/kg), one group with concurrent administration of both, and one group with prednisolone(reference drug). macro-and microscopic picture, ulcer index and lesion scores, oxidative markers, inflammatory mediators, in vitro activity of the inflammatory enzymes and 1, 1-diphenyl-2-picrylhydrazyl free radicals scavenging activity were evaluated. The phytochemical constituents of both extracts were explored by GC-mS analysis. Results: Both treatments modulated the macro-and microscopic picture, decreased the ulcerative index, lesion score, oxidative markers and inflammatory mediators, and inhibited the COX-1 and COX-2 enzymes. Propolis appeared to be powerful free radicals scavenger. A powerful synergistic effect of both treatments in modulating the course of the disease was reported. GCmS analysis of methanol extract of m. oleifera seeds and propolis revealed the presence of 50 and 34 compounds, respectively. Conclusions: m. oleifera seeds and propolis methanol extracts have modulated the course of acetic acid-induced ulcerative colitis. moreover, both treatments induce a good synergistic effect against the disease. Isolation of the active constituents is recommended.

Phytochemical, antioxidant and hepatoprotective effects of different fractions of moringa oleifera leaves methanol extract against liver injury in animal model

Objective: To evaluate the potential antioxidant and hepatoprotective effects of n-hexane, dichloromethane(DCm), ethyl acetate(EtOAc), n-butanol and aqueous fractions of moringa oleifera(m. oleifera) leaves methanol extract against carbon tetrachloride(CCl_4)-induced liver injury in rats. methods: These fractions were prepared from the m. oleifera leaves methanol extract by solubilization in water and partitioning in n-hexane, EtOAc, DCm and n-butanol. Their phyto-components were identified by GC-mS analysis. The in vitro antioxidant effect of these fractions was carried out by assessment of 1,1-diphenyl-2-picrylhydrazyl scavenging activity. A total of 40 Sprague Dawley rats were allocated into 8 equal groups: group 1 given olive oil(1 m L/kg ***.), group 2 injected with CCl_4, group 3 to 7 administered with n-hexane, DCm, EtOAc, n-butanol and aqueous fractions, respectively after CCl_4, group 8 administered with silymarin after CCl_4. The activities of aspartate aminotransferase, alanine aminotransferase, and the levels of total cholesterol, triglycerides, glucose, total proteins and albumin in serum were determined spectrophotometrically. Glutathione reduced, lipid peroxide by-products levels, glutathione-s-transferase and catalase enzyme activities in the liver homogenate were determined by spectrophotometer. Liver specimens were also examined for histopathological alterations under light microscope. Results: The GCmS analysis of different fractions of the m. oleifera leaves methanol extract revealed that n-hexane, DCm, EtOAc, n-butanol, and aqueous fractions contained 17, 22, 23, 19 and 32 compounds, respectively. The percent and the molecular structure of each component in each fraction were identified. The n-butanol and EtOAc fractions exhibited the strongest in vitro antioxidant activity against 1,1-diphenyl-2-picrylhydrazyl. Objective: To evaluate the effect of moringa oleifera leaf ethanol extract as an adjunct treatment on lead acetate induced hepatonephrotoxicity in rabbits. methods: Thirty-six male New Zealand White rabbits were assigned into two main groups. The first group(14 rabbits) served as normal control. The second group(22 rabbits) was administered orally with lead acetate at a dose of 40 mg/kg/day, 5 days/week for 8 weeks. At the 4 th and the 8 th week of treatment, 6 animals(3 animals at each period) of the second group were sacrificed while the remaining animals(16 rabbits) were assigned randomly into 2 subgroups(8 rabbits each): treated and non-treated. The first subgroup was orally given 1 m L phosphate-buffered saline for further 4 weeks while the second subgroup was administered orally with moringa oleifera leaf ethanol extract at a dose of 400 mg/kg/day for the same period. Blood samples were collected to determine hematological and serum biochemical indices. Tissue specimens were collected from the liver and kidney for evaluation of the oxidant/antioxidant markers and for histopathological ***: Lead acetate exposure decreased the mean body weight gain, hematocrit, hemoglobin, mean corpuscular volume, and lymphocytes count. moreover, it markedly increased counts of monocytes and platelets, serum enzyme activity, levels of creatinine, total cholesterol, triglycerides, and low-density lipoprotein cholesterol. malondialdehyde level was markedly increased while the reduced glutathione content was significantly decreased in liver tissue of lead intoxicated-rabbits. Histopathological alterations were also noticed in the liver and kidney of lead intoxicated rabbits. moringa oleifera leaf ethanol extract significantly improved hematological and serum biochemical parameters and histopathological structure of the liver and kidney. Effect of Egyptian propolis on cryptosporidiosis in immunosuppressed rats with special emphasis on oocysts shedding,leukogram,protein profile and ileum histopathology

Objective:To investigate the activity of Egyptian propolis extracts(ethanol and water)on cryptosporidiosis in experimentally infected dexamethasone-immunosuppressed ***:A total of 180 male rats(190-220)g BWt were randomly divided into 9 equal groups(G1-G9).Groups of rats were kept as(G1):normal control,(G2-G9):immunosuppressed with dexamethasone and(G3-G9):infected with Cryptosporidium *** from(G4to G9)were given orally ethanol and water extract of propolis(at a dose of 50 mg/kg BWt)and nitazoxanide(standard anti-cryptosporidial drug at a dose of 100 mg/kg BWt)to infected rats with different *** pellets were collected from all groups to monitor oocysts shedding from the 2nd to the 15th day post *** the end of the experiment,blood was collected from all groups for determination of leukogram and serum *** specimens were also examined ***:The highest reduction of oocysts shedding in faecal samples was 88%in rats prophylactically treated with propolis ethanol extract at the 4th dpi,and in rats prophylactically treated with water extract of propolis,was91%at the 6th *** was a marked increase in neutrophils count and α_2-andβ-globulins levels in infected rats treated with both extracts,while a significant decrease was detected in lymphocytes compared to the infected non treated group.β-globulin level markedly increased in the rats administered *** changes were observed in the ileum of rats infected with ***:Egyptian propolis extracts have an activity on cryptosporidiosis in ***,propolis modulated the immunity in dexamethasone-immunosuppressed rats.

Assessment the Female Sexual Function among Libyan Women at Tripoli, Libya, 2024

Background: The sexual function is an essential aspect of life for women irrespective of age because it is closely correlated with overall wellbeing and relationship satisfaction. Female Sexual Dysfunction (FSD) is a common public health issue that is defined as one or more problems of female sexual desire, arousal, orgasm and/or sexual pain/discomfort that leads to significant distress. Aim: This study aimed to assess the female sexual function among Libyan women. methods and materials: This descriptive cross-sectional study was carried out at Tripoli University Hospital over six months duration between January and June 2024. It included 314 female participants who attended the hospital during that period. The data underwent analysis and prescription via a computerized program of SPSS version 24. Results: The most frequent age range was between 26 to 35 years accounting for 54.8% (172). The mean duration of marriage was 7.42 years ± 7.228 SD. The majority of participants were multipara accounting for 79.9% (251). Based on the classification of female sexual dysfunction severity, 41.7% (131) of participants had mild to moderate severity followed by 32.2% (101) had mild severity with an overall female sexual dysfunction rate of 81.2% (255). Based on female sexual function characteristics, 28.3% (89) had abnormal desire, 22% (69) had abnormal arousal, 55.4% (174) had abnormal lubrication, 24.5% (77) had undetected orgasm, 23.2% (73) had inappropriate satisfaction and 19.1% (60) had pain. Conclusion: The overall rate of female sexual dysfunction among Libyan women was (81.2%) which is considered high but with low severity, (41.7%) had mild to moderate dysfunction followed by (32.2%) with mild dysfunction. Component Analysis and Antifungal Activity of the Compounds Extracted from Four Brown Seaweeds with Different Solvents at Different Seasons

The polar and nonpolar extracts of Colpomenia sinuosa, Padina pavonia, Cystoseira barbata and Sargassum vulgare collected during spring, summer and autumn were evaluated for their antifungal activities. The phytochemicals of unexplored seaweeds were analyzed by gas chromatography and mass spectrometric （GC/mS）. The algal extracts were tested for their antifungal activities against Aspergillus niger, A. flavus, Penicillium parasiticus, Candida utilis and Fusarium solani. Phytochemicals were extracted from the four seaweeds with various solvents including methanol, ethanol, acetone, chloroform and dimethyl ether. Among the various extracts, methanolic extract showed the highest inhibition activity on all fungal species. Seasonal variation in antifungal activity was studied, while methanol extracts showed the best antifungal activity in spring. Cystoseira barbata was the most effective seaweed, having anatifungal activity throughout the whole year. The UV-Vis phytochemical procedure and GC-mS analysis of the methanol extracts from tested species indicated the existence of different constituents. In conclusions, the compounds with antifungal activity were identified as indoles, terpenes, acetogenins, phenols, and volatile halogenated hydrocarbons.

Suppression of meloidogyne javanica by antagonistic and plant growth-promoting rhizobacteria

Four rhizobacteria selected out of over 500 isolates from rhizosphere of the vegetables in China were further studied for suppression of the root-knot nematode and soil-borne fungal pathogens in laboratory and greenhouse in Belgium. They were identified as Brevibacillus brevis or Bacillus subtilis by Biolog test and partial 16s rDNA sequence comparison. They not only inhibited the radial growth of the root-infecting fungi Rhizoctonia solani SX-6, Pythium aphanidermatum ZJP-1 and Fusarium oxysporum ***. cucumerinum ZJF-2 in vitro, but also exhibited strong nematicidal activity by killing the second stage larvae of meloidogyne javanica to varying degrees in the greenhouse. The toxic principles of bacterium B7 that showed the highest juvenile mortality were partially characterized. The active factors were heat stability and resistance to extreme pH values. B7 used either as seed dressing or soil drench significantly reduced the nematode populations in the rhizosphere and enhanced the growth of mungbean plants over the controls in the presence or absence of R. solani.

OBJECTIVE:To investigate the efficacy of Fig fruit powder and olive on hepatic,renal and splenic injury induced by 2-nitropropane(2-NP)in mice,especially if they were used in ***:A total of 40 adult BALB/c male mice weighting 25-30 g/*** were categorized into five groups(8 each).Group 1 as negative *** 2 as positive control group intraperitoneally injected with 2-NP(100 mg/kg b.w.)3 times/weekly for eight *** 3 injected with 2-NP and were orally supplemented with Fig(300 mg/kg).Group 4 injected with 2-NP and were orally supplemented with olive(100 mg/kg).Group 5 injected with 2-NP and were orally supplemented with mixture of Fig and olive(3∶1 respectively).RESULTS:Histopathological observation of liver in mice treated with 2-NP showed cellular degeneration,pyknosis,and congestion of the portal *** kidney there were disorganization of the cortical tissues,cellular necrosis and plenty of inflammatory lymphocytic *** elevations in liver function parameters(alanine aminotransferase and aspartate aminotransferase),m RNA expression levels of tumor necrosis factor-α,nicotinamide adenine dinucleotide phosphate oxidase and cyclooxygenase were detected as anti-inflammatory markers and 5-lipoxygenase,interleukin-1βand interleukin-6 as inflammatory biomarkers for liver and spleen,also significant elevations was detected in lipid peroxidation *** levels of antioxidants,glutathione,glutathione peroxidase,catalase and superoxide dismutase were significantly ***:our findings indicated that Fig fruit powder and olive protected against hepatic,renal and splenic injury induced with 2-NP in mice,especially if they were used in combination.

Synergetic effect of Egyptian propolis in immunization of BALB/c mice against bovine cysticercosis

Objective: To evaluate the synergetic effect of an ethanolic extract of Egyptian propolis in immunization of BALB/c mice with Taenia saginata(T. saginata) crude antigen against bovine cysticercosis, with reference to its effects on liver and kidney ***: Sixty female mice BALB/c strain weighing 20 to 25 g and 6-8 weeks old were randomly allocated into six groups of ten mice each. mice in groups 1 and 2(G1 and G2) were immunized intraperitoneally with 100 μg of T. saginata crude antigen in 100 μL phosphate buffer saline emulsified in Freund's adjuvant. Besides, the mice in G2 were administered with propolis extract simultaneously with immunization. Control mice were either administered with propolis extract(G3) or injected with the same volume of phosphate buffer saline emulsified in Freund's adjuvant(G4). The mice in G5 were non-immunized infected control while, those in G6 were non-immunized non-infected control. Two weeks after the last immunization, each mouse was challenged intraperitoneally with 5 000 oncospheres except those of G6. Ethanolic extract of propolis was prepared at a dose 50 mg/kg body ***: After 24 weeks of challenge, the mice in G2 showed the highest level of protection(100%), with no cyst being detected rather than mice in G1(33.3% protection). Additionally,the ELISA results, in this study, showed higher antibody titer in G2 with reduction the alteration in liver and kidney functions compared to ***: Egyptian propolis could increase the level of protection against experimental challenge infection with T. saginata eggs when administered simultaneously with immunization. Furthermore, it could enhance the production of antibodies to immunized antigen and decrease the alteration in liver and kidney functions.

Sesamol Alleviates the Cytotoxic Effect of Cyclophosphamide on Normal Human Lung WI-38 Cells via Suppressing RAGE/NF-κB/Autophagy Signaling

Cyclophosphamide(CYL)is a chemotherapeutic medication commonly used in managing various malignancies like breast cancer or ***,CYL has been documented to induce lung *** of CYL toxicity is through oxidative stress and the release of damage-associated molecular patterns(DAmPs).Sesamol(SES)is a natural antioxidant isolated from Sesamum indicum and its effect against CYL-induced lung toxicity is not studied *** study aims to inves-tigate whether SES could prevent any deleterious effects induced by CYL on lung using normal human lung cells,WI-38 cell line,without suppressing its *** were pretreated with SES and/or CYL for 24 h,then cell viability was estimated by mTS and trypan blue *** mode of cell death was determined by AO/EB ***,caspase-3 level,oxidative stress,and inflammatory markers were evaluated by colorimetric and ELISA ***-PCR was performed to evaluate RAGE,NF-κB,and Beclin-1 ***-treated WI-38 cells developed a significantly increased cell death with enhanced oxidative and RAGE/NF-κb/Autophagy signaling,which were all attenuated after pretreatment with ***,we concluded that SES offered a protective role against CYL-induced lung injury via suppressing oxidative stress and RAGE/NF-κB/Autophagy signaling,which is a natural safe therapeutic option against CYL toxicities.
