Our study related to the renal toxicity of Wistar rats induced by solutions of amphotericin B prepared under extreme conditions of pH (5.4 and 10.8). The results obtained show that with pH 5.4 of stock solution, urea and creatinin rate blood is not disturbed. These means that the renal function is not deteriorated by the amphotericin B. Furthers, treatment of animals infected by the yeast Candida albicans, with the solution of amphotericin B prepared at pH 5.4 and injected at 0.5 mg of AmB/Kg every 24 hours, seems to be effective.

The aim of our work is the study of nutrition diabetic women, and the study of the health status of their newborns in West of Algeria. Our study was performed on 165 diabetic mothers aged 20 to 45 years and 161 newborns admitted to the hospital specialized in obstetrics and gynecology of Sidi Bel Abbes. A food survey and anthropometric study were achieved in our population, clinical and socioeconomic data were recorded in an identification card. Our results show that the average age of women with diabetes is 33.26 ± 5.15 ans. The socio-economic survey shows that 47.32% of women have an average monthly income;their education level is low with only 8.06% having a higher level. Anthropometric parameters show that 48.08% of elderly diabetic women aged 30 to 40 years are overweight BMI > 25. Dietary survey indicates that women experienced iron, calcium and vitamin B9 deficiency. The results of clinical data have shown the existence of gestational diabetes with a rate of 50.90%;macrosomia is dominant in their newborns with a rate of 22.26%. We noted the presence of hypoglycemia in 18.01% and inutero death in 12.42%.

Background: The prevalence of overweight and obesity associated with oxidative stress and immune abnormalities is continuously increasing. Antioxidant supplementations might counteract potential damage caused by ROS to cellular tissues. Objective: To determine the role of vitamins on immune improvement during obesity, we investigated in vitro effects of vitamins C, E, and NADH on mitogen-stimulated proliferation, Th1- and Th2-type cytokine production, and oxidant/antioxidant status of lymphocytes isolated from obese patients. Methods: Peripheral blood lymphocytes were isolated using a density gradient of Histopaque. They were in vitro cultured and stimulated by Con A in the presence or absence of vitamins. Cell proliferation was determined by MTT assay and interleukin-2, interleukin-4 and interferon-γ (INFγ) secretions. Cell oxidant/antioxidant balance was studied by assaying glutathione (GSH), malondialdehyde (MDA), carbonyl protein levels, catalase activity and micronucli frequency. Results: Obesity is associated with enhanced oxidative stress response. Indeed, vitamin C, E and NADH improved significantly lymphocyte proliferation and diminished cellular oxidative stress. Conclusion: Treatments of lymphocytes with vitamins had beneficial effects on lymphocyte proliferation, cytokines secretions and redox status, generating an anti-inflammatory profile and should be considered in therapeutic approaches for normalizing immune cell function in obesity.

The objective of this work was to determine the sodium metabisulphite (NaMBS) subchronic toxicity used as a food additive in Algeria. Three groups of female Wistar rats were treated with 0.25%, 1% and 4% of NaMBS in their drinking water for 90 days. An immunization protocol was conducted during the experiment. Mortality, comportmental and weight modifications, and food and water consumption were recorded. At the end of the experiment, the control and experiment rats were killed, and their blood and organs were removed. Immunoglobulin levels were evaluated;biochemical and hematological parameters were investigated. Our results showed that the administration of NaMBS at 1% and 4% had significant effects on body weight, food and water consumed. There was an increase in biochemical parameters (calcium, urea, creatinine, uric acid, transaminases) and decrease immunoglobulin levels. The hematology revealed a decrease in red blood cells and hemoglobin, as well as leucocytosis. Physiological study showed enlarged spleen, kidney, liver and stomach. In light of our results, we can conclude that subchronic intake of NaMBS 1% and 4% seems to alter immune function, biochemical, hematological and physiological para-meters in Wistar rats.

Background: Dietary fatty acids have important homeostatic functions in regulating the immune response and may exert beneficial effects on immune alterations during obesity. Objective: To assess the in vitro effects of oil fatty acids, different oils (olive, linseed, Nigel, sunflower) were tested on T-lymphocyte proliferation, Th1- and Th2-type cytokine production, and intracellular oxidant/antioxidant status in obese patients. Methods: Peripheral blood lymphocytes were isolated using Histopaque and were in vitro cultured and stimulated by Con A in the presence or absence of the oils. Cell proliferation, interleukin-2, interleukin-4 and interferon-γ (INFγ) secretions and intracellular oxidative status (glutathione (GSH), malondialdehyde (MDA), carbonyl protein levels, catalase activity and micronuclei frequency) were investigated. Results: Abnormalities in lymphocyte function and intracellular oxidative stress were observed in obesity. Linseed oil induced a reduction in T-lymphocyte proliferation and IL-2 production while Nigel oil increased them in both obese and control groups. In addition, Nigel oil enhanced IFNγ and IL-4 secretion. Olive and sunflower oils had no effect on lymphocyte proliferation and cytokine secretion in both groups. Linseed and Nigel oils induced an increase in T cell GSH concentrations and catalase activity with a concomitant decrease in MDA, carbonyl protein contents and micronuclei frequency especially in obese patients. Conclusion: Linseed and Nigel oils had beneficial effects on lymphocyte proliferation, cytokines secretions and redox status, while olive and sunflower oils had no effects on immune cell function in obesity.

Molecular Weight Determination of a Protease Extracted from <i>Mucor pusillus</i>: Comparison Methods

Mucor pepsin, a protease used in milk coagulation, is purified by ion-exchange and by molecular exclusion on Sephadex G100. The molecular weight (MW) is determined by polyacrylamide gel electrophoresis under denaturing conditions in presence of sodium dodecyl sulfate (SDS) and by molecular exclusion chromatography. Approximate evaluation of molecular mass was conducted by elution of known MW proteins (BSA: 67 kDa, pepsin: 35 kDa and trypsin: 23.8 kDa) on Sephadex G-100 under the same conditions as the experimental sample. The electrophoretic profile shows that the active fraction studied appears as a single homogeneous band (monomeric form). According to the curve calibration, the molecular mass of the coagulant fraction is about 48 kDa. For Mucor, the observed MW value seems to be enigmatic. However, this result is confirmed by a proteomic analysis with close MW values obtained using conventional techniques. The protease studied by the Scafold ver. Software 2.0 and the analysis of the protein similarities indicate a MW of 46 kDa and the protease sequence of 427 amino acids.

Risk Factors for Overweight and Obesity among Children Ages 0 to 3.5 Living in the Miramichi Public Health Region of New-Brunswick, Canada

Obesity, a major risk factor in numerous pathologies, poses a public health problem. The objective of this study was to assess the prevalence of the risk of overweight, overweight and obesity, as well as to identify and analyze the risk factors for weight gain among children in Miramichi in New Brunswick’s Horizon Health Network (HHN). This descriptive cross-sectional study was done between 2009 and 2014. The study population was composed of 335 children (185 boys and 150 girls) ages 0 to 42 months and their parents. Overweight and obesity were determined according to World Health Organisation (WHO) criteria adapted for Canada. A logistic regression analysis was performed to determine the risk factors associated with overweight and obesity. The prevalence of risk for overweight is 21% at birth as opposed to 55% at 42 months (both sexes together), and the prevalence of risk for overweight including obesity affects 11.8% of children, that is, 12.7% of boys as opposed to 10.94% of girls (p < 0.0001). The prevalence among boys is 1.2 times that among girls. This study also reveals that at 42 months, the average prevalence of obesity is 6.5% (8% for boys and 5% for girls). The mothers of overweight children have a higher post-pregnancy BMI (32.78 ± 4.16 kg/m2) than do the mothers of children who are a healthy weight (26.17 ± 7.90 kg/m2) (p < 0.0001). Moreover, 29.7% of children are overweight when both parents are overweight compared to 14.97% when both parents are a healthy weight (p < 0.05). This means that children are twice as likely to be overweight when both parents are overweight compared to children whose parents are a healthy weight. Otherwise, only 17% of the 335 children assessed in this study were breastfed and started on solid foods in accordance with the WHO recommendations. Background: A number of epidemiological studies around the world have shown that the occurred and the high prevalence of some chronic diseases such as diabetes, obesity, cardiovascular disease, etc. are mainly caused by a high calorie and unbalanced diet associate with reduced physical activity. In Algeria, as in many emerging countries to improve the standard of living of the population and changes in lifestyle, more westernized characterized by abundant food and sedentary lifestyle especially among young people contribute to the increased incidence of these chronic diseases. Objectives: This work consists of a dietary survey conducted among adolescent boys and girls in Tlemcen (Algeria). Subject and Methods: This is a descriptive cross-sectional study of dietary intake of adolescent aged 10 - 17 years and anthropometric parameters: weight, height, body mass index. Results: Increased fat intake was associated with increased body mass index in both sexes, increased obesity and abdominal obesity in girls, boys are more active than girls and we observed decreased intake of sugar, the nutritient adequacy ratio of folate, niacin, vitamin B6, iron, magnesium. Conclusion: The early prevention of obesity in children and adolescents is essential in the prevention of chronic diseases in adults. We recommended adolescent by healthy diets towards helps regulate their body weight and reduces the risk of cardiovascular disease and some types of cancer and especially the girls by lifestyle more active.
