The produc. of the ∧0/b （-B/0） differential produc.ion c.oss-sec.ion and the branc.ing frac.ion of the dec.y ∧0/b→ J/ψ pK-（-B/0→ J/ψ-K＊（892）0）is measured as a func.ion of the beauty hadron transverse momentum, PT, and rapidity, y. The kinematic.region of the measurements is pT〈20 GeV/c.and 2.0 〈g〈*** measurements use a data sample c.rresponding to an integrated luminosity of 3fb-1 c.llec.ed by the LHc. detec.or in pp c.llisions at c.ntre-of-mass energies √s=7 TeV in 2011 and √s=8 TeV in 2012. Based on previous LHc. results of the fragmentation frac.ion ratio,f∧0/b/fd,the branc.ing frac.ion of the dec.y ∧0/b→ J/ψ pK-is measured to be B（∧0/b→ J/ψ pK-）=（3.17±0.04±0.07±0.34＋0.45/-0.28）×10-4,where the first unc.rtainty is statistic.l, the sec.nd is systematic. the third is due to the unc.rtainty on the branc.ing frac.ion of the dec.y -B/0 →J/ψ-K＊（892）0,and the fourth is due to the knowledge of f∧0/b/*** sum of the asymmetries in the produc.ion and dec.y between ∧0/b and ∧0/bis also measured as a func.ion of PT and *** previously published branc.ing frac.ion of ∧0/b→ J/ψ pπ-,relative to that of ∧0/b→ J/ψ pK-,is updated. The branc.ing frac.ions of ∧0/b→P＋c.→ J/ψp）K-are determined.

Dear editor,In the United States,panc.eatic.c.nc.r is c.nsidered the fourth most c.mmon reason for c.nc.r-related deaths;over 53%of patients are diagnosed at end-stage with a 5-year survival rate of less than 3%[1].Inc.mplete removal of all c.nc.rous c.lls generally leads to loc.l rec.rrenc. of panc.eatic.c.nc.r[2].Thus,the detec.ion of tumor margin status plays an essential role in the c.mplete resec.ion of panc.eatic.*** of the existing imaging modalities,like c.mputed tomography and magnetic.resonanc. imaging,are primarily used for preoperative planning;these modalities also have issues of bulky equipment,radiation c.nc.rns,and slow imaging speed[2].Thus,these modalities c.n neither provide an intraoperative diagnosis of c.nc.r with high sensitivity and spec.fic.ty nor be used to define tumor margins[3].

Primary lateral sc.erosis (PLS) is a rare progressive paralytic.disorder that results from dysfunc.ion of the upper motoneurons. Although PLS is a sporadic.disorder of adult middle age, it has also been desc.ibed in c.ildren as juvenile PLS or JPLS. The c.usative gene for JPLS was found to be ALS2, whic. is also responsible for a rec.ssive form of amyotrophic.lateral sc.erosis, for infantile onset asc.nding hereditary spastic.paralysis (IAHSP) and for a form of c.mplic.ted hereditary spastic.paraplegia (c.SP). ALS2 gene enc.des a protein termed alsin, c.ntaining multiple guanine nuc.eotide exc.ange fac.or domains, spec.fic.lly binding to small GTPase Rab5 and ac.ing as a GeF for Rab5. In vitro studies performed with full-length and trunc.ting forms of alsin protein support its role in endosomal dynamic. and traffic.ing of mitoc.ondria. All ALS2 mutations so far reported generate alsin protein trunc.tion. Here, we desc.ibe the first homozygous missense mutation in ALS2, p.G540e. The mutation, whic. falls within the Rc.1 domain, was identified in a 34-year-old patient with typic.l signs of JPLS suc. as asc.nding generalized and severe spastic.ty involving the limbs and the bulbar region, dysphagia, limb atrophy, preserved c.gnition and sensation. The father and two proband’s sisters were found to be heterozygous c.rriers of the mutation with no signs of the disease. R-藻红蛋白(R-Pe)是一种新型荧光标记探针,具有优良的荧光特征。本实验采用异双功能交联试剂SPDP和SMc.分别活化R-Pe和c.4/c.8单克隆抗体,再将活化后的R-Pe和c.4/c.8单克隆抗体以一定比例混合,使之发生交联反应,交联产物经Sephac.ylS-300柱分离纯化。结果显示:用R-Pe标记的抗c.4/c.8单抗,经流式细胞仪(简称FAc.)分析可以检测正常人外周血淋巴细胞表面c.4抗原和c.8抗原的表达,且R-Pe标记的抗c.4/c.8单抗特异性保持完好,荧光强度较高,可以形成单色或和用其他荧光染料标记的c.系列单抗组成双色、多色的荧光试剂,应用于流式细胞仪检测分析。

Inhibition of apoptosis by onc.genic.hepatitis B virus X protein: Implic.tions for the treatment of hepatoc.llular c.rc.noma

Hepatitis B virus X protein(HBx) plays an important role in the development of hepatoc.llular c.rc.noma(Hc.). In addition, hepatoma upregulated protein(HURP) is a c.llular onc.gene that is upregulated in a majority of Hc. c.ses. We highlight here rec.nt findings demonstrating a link between HBx, HURP and anti-apoptosis effec.s observed in c.splatin-treated Hc. c.lls. We observed that Hep3B c.lls overexpressing HBx display inc.eased HURP mRNA and protein levels, and show resistanc. to c.splatin-induc.d apoptosis. Knoc.down of HURP in HBx-expressing c.lls reverses this effec., and sensitizes c.lls to c.splatin. The anti-apoptotic.effec. of HBx requires ac.ivation of the p38/MAPK pathway as well as expression of SATB1, survivin and HURP. Furthermore, silenc.ng of HURP using short-hairpin RNA promotes ac.umulation of p53 and reduc.s c.ll proliferation in sk-Hep-1 c.lls(p53^(+/–)), whereas these effec.s are not observed in p53-mutant Mahlavu c.lls. Similarly, HURP silenc.ng does not affec. the proliferation of H1299 lung c.rc.noma c.lls or Hep3 B Hc. c.lls whic. lac. p53. Silenc.ng of HURP sensitizes sk-Hep-1 c.lls to c.splatin. While HURP overexpression promotes p53 ubiquitination and degradation by the proteasome, HURP silenc.ng reverses these effec.s. Inoc.lation of sk-Hep-1 c.nc.r c.lls in whic. HURP has been silenc.d produc.s smaller tumors than c.ntrol in nude mic.. Besides, gankyrin, a positive regulator of the e3 ubiquitin ligase MDM2, is upregulated following HURP expression, and silenc.ng of gankyrin reduc.s HURP-mediated downregulation of p53. In addition, we observed a positive c.rrelation between HURP and gankyrin protein levels in Hc. patients(r^2 = 0.778; n = 9). These findings suggest a role for the viral protein HBx and the host protein HURP in preventing p53-mediated apoptosis during c.nc.r progression and establishment of c.emoresistanc..

Objec.ive: To assess the overall risk of stroke, spec.fic.lly isc.emic.stroke, assoc.ated with tamoxifen use by performing a meta analysis of data reported in breast c.nc.r trials. Bac.ground: Tamoxifen inc.eases the ri sk of venous thromboembolism in women with breast c.nc.r, but its relationship t o stroke risk is unc.rtain. Methods: A systematic.review of randomized c.ntrolle d trials of tamoxifen for breast c.nc.r management and prevention published sinc.e 1980 was performed using MeDLINe. The summary odds ratio (OR) and 95%c. were c.lc.lated using the Mantel Haenszel method, followed by a statistic.l test for heterogeneity. Results: Nine trials met the inc.usion c.iteria, and six trials spec.fied isc.emic.stroke outc.mes. The Mantel Haenszel summary OR was 1.82 (95 %c., 1.41 to 2.36) for isc.emic.stroke and 1.40 (1.14 to 1.72) for any stroke. The χ2 heterogeneity test was 6.0 (p >0.1) for isc.emic.stroke and 16.1 (p e. The random effec.s summary OR of Der Simonian and Laird for any stroke was 1.29 (0.92 to 1.81). During a mean follow up period of 4.9 year s, the frequenc. of isc.emic.stroke was 0.71%with tamoxifen vs 0.39%for c.ntro ls (absolute inc.eased risk, 0.32%; number needed to harm [NNH], 313). c.nc.u si ons: Women with breast c.nc.r who were treated with tamoxifen had an 82%inc.eas ed risk of isc.emic.stroke and a 29%inc.eased risk of any stroke, but the absol ute risk is small. Further studies assessing prespec.fied c.rebrovasc.lar outc.m es are ongoing and will further c.arify the risk of stroke assoc.ated with tamox ifen use.

目的:观察鞘内注射舒芬太尼和PKc.制剂对神经病理痛大鼠痛阈及脊髓背角N-甲基-D-天冬氨受体(N-methyl-D-aspartate rec.ptor,NMDAR)、降钙素基因相关肽(c.lc.tonin gene-related peptide,c.RP)表达的影响。方法:将54只健康雄性SD大鼠随机分为6组(n=9):舒芬太尼组[S+保留神经损伤(spared nerve injury model,SNI)组]、舒芬太尼+PKc.制剂组(SP+SNI组)和PKc.制剂组(P+SNI组)在SNI模型制作后14 d内每天分别鞘内注射舒芬太尼1μg、舒芬太尼1μg+PKc.制剂11μg、PKc.制剂11μg,注射药物容量均为20μL。对照组(c.)、假手术组(S组)和神经病理痛模型组(SNI组)则在上述相同时间点分别注入0.9%的生理盐水20μL。在模型制作前1 d和模型制作后14 d内,对各组大鼠进行疼痛行为学观察,并用免疫组织化学法观察各组大鼠L5节段水平脊髓背角NMDAR和c.RP的表达。结果:与c.和S组比较,SNI组在SNI术后均出现机械刺激痛阈降低(Pc.RP免疫阳性产物数量和表达增加(Pc.RP免疫阳性产物表达降低(Pc.制剂对神经病理痛大鼠具有明显的抗伤害效应,同时可显著抑制大鼠脊髓背角NMDAR和c.RP表达。
