This study wa. conducted to cla.ify the toxic effects of sertra.ine （SRT） on the reproductive system of ma.e ra.s a.d to elucida.e the underlying mecha.isms. Ra.s were trea.ed ora.ly with SRT a. doses of 5, 10, a.d 20 mg kg-1 for 28 consecutive da.s. a. the end of the trea.ment period, sperm concentra.ion, sperm motility, a.d sperm morphology were investiga.ed by computer-a.sisted sperm a.a.ysis system wherea. sperm DNa.da.a.e wa. detected by comet a.sa.. The oxida.ive sta.us of the testes wa. investiga.ed, a.d a.histopa.hologica. exa.ina.ion wa. conducted. Serum testosterone, follicle-stimula.ing hormone （FSH）, a.d luteinizing hormone （LH） levels were mea.ured to determine the effects of SRT on the sperma.ogenesis process. One-wa. a.OVa. post-hoc Dunnett＇s T3 test for the sperm comet a.sa., a.d post-hoc Tukey＇s test for the others were performed for sta.istica. a.a.ysis. The results showed tha. SRT ca.sed a. increa.e in sperm DNa.da.a.e a.d induced histopa.hologica. lesions in a.l groups trea.ed with SRT. There wa. a.norma. sperm morphology a.d increa.ed ma.ondia.dehyde （MDa. in the 10 mg kg-1 trea.ment group. More dra.a.ic cha.ges were observed in the 20 mg kg-1 trea.ment group. Decrea.ed sperm count wa. a.compa.ied by a.significa.t increa.e in a.norma. sperm morphology, DNa.da.a.e, a.d degenera.ion in cellula.-tubula. structures. Serum LH a.d testosterone levels were eleva.ed in the 20 mg kg-~ trea.ment group. Decrea.ed gluta.hione （GSH） a.d increa.ed MDa.were signs of enha.ced oxida.ive stress （OS）. In conclusion, SRT induced testicula. toxicity in a.dose-dependent ma.ner a.d OS is suggested a. a.crucia. mecha.ism.

Objective: To investiga.e the effects of a.ra.orin, a.lichen seconda.y meta.olite, on SPC212 ma.igna.t mesothelioma.cells in vitro. Methods: SPC212 ma.igna.t mesothelioma.cell line wa. used. 3-(4,5-dimethylthia.ol-2-yl)-2,5-diphenyltetra.olium bromide a.sa. wa. used to eva.ua.e cytotoxic effects of a.ra.orin a.d cispla.in a. 24, 48 a.d 72 h. Hema.oxylin-eosin sta.ning a.d 4’,6-dia.idino-2-phenylindole, dihydrochloride sta.ning were used for determining cell a.d nucleus morphology, respectively. Wound hea.ing a.sa. wa. used for investiga.ing cell migra.ion. The xCELLigence rea.-time cell a.a.ysis system wa. used for determining cell ***: a.ra.orin a. 5-450 μM decrea.ed cell via.ility a. 24, 48 a.d 72 h. IC50 va.ues of a.ra.orin were 300.94, 292.6 a.d 278.02 μM a. 24, 48 a.d 72 h, respectively;mea.while, the IC50 va.ues of cispla.in were 128.00, 34.37 a.d 17.05 μM a. 24, 48 a.d 72 h, respectively. Furthermore, a.ra.orin disrupted cell a.d nuclea. morphology with increa.ing concentra.ions. a.ra.orin significa.tly reduced cell migra.ion by 38%, 37% a.d 35% a. 300, 250 a.d 200 μM, respectively(Pa.ra.orin a. 160-450 μM decrea.ed cell prolifera.ion a. 72 h(Pa.ra.orin ha. cytotoxic, a.tiprolifera.ive, a.optotic a.d cell migra.ion inhibitory effects on SPC212 ma.igna.t mesothelioma.ca.cer cells.
