Eicosanoid pathway in colorectal cancer:Recent updates

Enzymatic metabolism of the 20 C polyunsaturated fatty acid(PUFA) arachidonic acid(AA) occurs via the cyclooxygenase(COX) and lipoxygenase(LOX) pathways, and leads to the production of various bioactive lipids termed eicosanoids. These eicosanoids have a variety of functions, including stimulation of homeostatic responses in the cardiovascular system, induction and resolution of inflammation, and modulation of immune responses against diseases associated with chronic inflammation, such as cancer. Because chronic inflammation is essential for the development of colorectal cancer(CRC), it is not surprising that many eicosanoids are implicated in CRC. Oftentimes, these autacoids work in an antagonistic and highly temporal manner in inflammation; therefore, inhibition of the pro-inflammatory COX-2 or 5-LOX enzymes may subsequently inhibit the formation of their essential products, or shunt substrates from one pathway to another, leading to undesirable side-effects. A better understanding of these different enzymes and their products is essential not only for understanding the importance of eicosanoids, but also for designing more effective drugs that solely target the inflammatory molecules found in both chronic inflammation and cancer. In this review, we have evaluated the cancer promoting and anti-cancer roles of different eicosanoids in CRC, and highlighted the most recent literature which describes how those molecules affect not only tumor tissue, but also the tumor microenvironment. Additionally, we have attempted to delineate the roles that eicosanoids with opposing functions play in neoplastic transformation in CRC through their effects on proliferation, apoptosis, motility, metastasis, and angiogenesis.

Solifenacin is a muscarinic receptor antagonist that has been used to treat overactive bladder since *** has a great affinity for the detrusor M3 receptor,which must be stimulated for bladder muscle contraction,and demonstrates the most selective profile to the bladder of the muscarinic receptor *** is thought that urinary antimuscarinic agents,due to their passage to the central nervous system and lipophilic properties,may cause central nervous system symptoms in some rare cases.A case report of a 42-year old male patient who had an acute psychotic attack as a result of solifenacin treatment for overactive bladder is presented in this article.

The design and application of morphing systems are ongoing issues compelling the aviation *** Clean Sky-program represents the most significant aeronautical research ever launched in Europe on advanced technologies for greening next-generation *** primary purpose of the program is to develop new concepts aimed at decreasing the effects of aviation on the environment,increasing reliability,and promoting eco-friendly *** ambitions are pursued through research on enabling technologies fostering noise and gas emissions reduction,mainly by improving aircraft aerodynamic *** the Clean Sky framework,a multimodal morphing flap device was designed based on tight industrial requirements and tailored for large civil aircraft *** flap is deployed in one unique setting,and its cross section is morphed differently in take-off and landing to get the necessary extra lift for the specific flight ***,during the cruise,the tip of the flap is deflected for load control and induced drag *** manufacturing the first flap prototype,a high-speed(Ma=0.3),large-scale test campaign(geometric scale factor 1:3)was deemed necessary to validate the performance improvements brought by this novel system at the aircraft *** the other hand,the geometrical scaling of the flap prototype was considered impracticable due to the unscalability of the embedded mechanisms and actuators for shape ***,a new architecture was conceived for the flap model to comply with the scaled dimensions requirements,withstand the relevant loads expected during the wind tunnel tests and emulate the shape transition capabilities of the true-scale *** strategies were developed to effectively morph the model during wind tunnel tests while ensuring the robustness of each morphed configuration and maintaining adequate stiffness levels to prevent undesirable deviations from the intended aerodynamic ***,

In the wake of sustainable construction entering the mainstream,many developing countries are either currently pursuing green building guidelines or are planning to pursue them in the near *** research attempts to formulate an implementation strategy for the rapid adoption of these guidelines for the developing *** implementation strategy is based on the detailed review of the major green building guidelines globally and contex-tual information of Indian society collected through a survey *** proposed strategy shows that it is important for green building initiatives to identify:the organizations that accelerate the adoption of green building guidelines in a society,the incentives and barriers associated with the green building guidelines,and the necessary motivations for the adopter *** informed approach in the form of this implementation strategy might potentially contribute to the acceleration of green building guidelines’adoption in developing countries.

A protective action of melatonin(MELAT)on docetaxel(DCT)-induced inflammation,apoptosis,and reactive free oxygen radical(fROS)generation values via blocking of TRPM2 calcium-permeable channel was investigated in different cells except for laryngo-tracheal epithelial(LT-Epi)***,the protective action ofMELAT on DCT-induced oxidative toxicity and inflammation in LT-Epi tissue and cells of mice were investigated in the current *** treatment ameliorated DCTinduced mitochondrial ROS in the LT-Epi cells by reducing the generation of fROS(cytosolic and mitochondrial),lipid peroxidation,and depolarization of the mitochondrial membrane,while increasing reduced glutathione(GSH),GSH peroxidase,and total antioxidant *** addition,DCT-induced increases of cytokine(IL-1β,IL-6,and TNF-α)generations were also diminished in the LT-Epi tissue by MELAT ***,MELAT treatment increased viability and count of the cells followed by decreasing levels of cell death,caspase-3,*** TRPM2 activity was also reduced by MELAT and TRPM2 channel blocker(ACA)*** conclusion,MELAT modulated the increase of DCT-induced LT-Epi cell death by inhibiting mitochondrial oxidative stress and TRPM2 channel ***,DCTcaused side cell death,oxidant,and inflammatory actions in the LT-Epi were diminished via the treatment of MELAT.

AIM:To detect retinal microvascular variations in obstructive sleep apnea syndrome ***:This prospective,observational case-control study included healthy controls and patients with mild,moderate,and severe obstructive sleep apnea *** parameters,foveal avascular area,and flow areas in macula-centered,6.00×6.00 mm2 scan size optical coherence tomography angiography images were ***:The control group had the highest whole image,parafoveal,and perifoveal vessel density among the groups in both superficial and the deep capillary plexus(all P<0.05).Rapid eye movement sleep apnoea-hypopnoea index was reversely correlated with whole(Rho=-0.195,P=0.034),parafoveal(Rho=-0.242,P=0.008),perifoveal(Rho=-0.187,P=0.045)vessel density in the superficial capillary plexus,and whole(Rho=-0.186,P=0.046),parafoveal(Rho=-0.260,P=0.004),perifoveal(Rho=-0.189,P=0.043)vessel density in the deep capillary plexus,though the mean and non-rapid eye movement sleep apnoeahypopnoea index related with only parafoveal vessel density in the superficial capillary plexus(Rho=-0.213,P=0.020;Rho=-0.191,P=0.038)and the deep capillary plexus(Rho=-0.254,P=0.005;Rho=-0.194,P=0.035).CONCLUSION:This study shows decreased vessel density and its reverse correlation with the apnoea-hypopnoea index in patients with obstructive sleep apnea syndrome.

The lattice parameters,bulk modulus,rst derivative of the bulk modulus,electronic band structures,phonon dispersion curves and phonon density of states calculations for Li_(2)AlGa and Li_(2)AlIn Heusler alloys are performed and compared in this study using density functional theory within the generalized gradient *** lattice parameters display a good agreement with the *** electronic band structures of both Heusler alloys show that they are in semi-metallic *** dispersion curves and the phonon density of states graphs are also obtained in order to study the lattice dynamics of these Heusler *** is noticed that Li_(2)AlGa and Li_(2)AlIn Heusler alloys are dynamically stable in the ground state.

This study was conducted to clarify the toxic effects of sertraline （SRT） on the reproductive system of male rats and to elucidate the underlying mechanisms. Rats were treated orally with SRT at doses of 5, 10, and 20 mg kg-1 for 28 consecutive days. At the end of the treatment period, sperm concentration, sperm motility, and sperm morphology were investigated by computer-assisted sperm analysis system whereas sperm DNA damage was detected by comet assay. The oxidative status of the testes was investigated, and a histopathological examination was conducted. Serum testosterone, follicle-stimulating hormone （FSH）, and luteinizing hormone （LH） levels were measured to determine the effects of SRT on the spermatogenesis process. One-way ANOVA, post-hoc Dunnett＇s T3 test for the sperm comet assay, and post-hoc Tukey＇s test for the others were performed for statistical analysis. The results showed that SRT caused an increase in sperm DNA damage and induced histopathological lesions in all groups treated with SRT. There was abnormal sperm morphology and increased malondialdehyde （MDA） in the 10 mg kg-1 treatment group. More dramatic changes were observed in the 20 mg kg-1 treatment group. Decreased sperm count was accompanied by a significant increase in abnormal sperm morphology, DNA damage, and degeneration in cellular-tubular structures. Serum LH and testosterone levels were elevated in the 20 mg kg-~ treatment group. Decreased glutathione （GSH） and increased MDA were signs of enhanced oxidative stress （OS）. In conclusion, SRT induced testicular toxicity in a dose-dependent manner and OS is suggested as a crucial mechanism.
