Negative Feedback Regulation of Auxin Signaling by ATHB8/ACL5-BUD2 Transcription Module

The role of auxin as main regulator of vascular differentiation is well established, and a direct correlation between the rate of xylem differentiation and the amount of auxin reaching the （pro）cambial cells has been proposed. It has been suggested that thermospermine produced by ACAULIS5 （ACL5） and BUSHY AND DWARF2 （BUD2） is one of the factors downstream to auxin contributing to the regulation of this process in Arabidopsis. Here, we provide an in-depth characterization of the mechanism through which ACL5 modulates xylem differentiation. We show that an increased level of ACL5 slows down xylem differentiation by negatively affecting the expression of homeodomain-leucine zipper （HD- ZIP） III and key auxin signaling genes. This mechanism involves the positive regulation of thermospermine biosynthesis by the HD-ZIP III protein ARABIDOPSIS THALIANA HOMEOBOX8 tightly controlling the expression of ACL5 and BUD2. In addition, we show that the HD-ZIP III protein REVOLUTA contributes to the increased leaf vascularization and long hypocotyl phenotype of acl5 likely by a direct regulation of auxin signaling genes such as LIKE AUXIN RESISTANT2 （LAX2） and LAX3. We propose that proper formation and differentiation of xylem depend on a balance between positive and negative feedback loops operating through HD-ZIP III genes.

In this paper we present a research-through-design study where we employed text-to-text,text-to-image,and image-to-image generative tools for a conceptual architecture project for the eVolo skyscraper competition.We trained these algorithms on a dataset that we collected and curated,consisting of texts about and images of architecture.We describe our design process,present the final proposal,reflect on the usefulness of such tools for early-stage design,and discuss implications for future research and practice.By analysing the results from training the text-to-text generators we could establish a specific design brief that informed the final concept.The results from the image-to-image generator gave an overview of the shape grammars of previous submissions.All results were intriguing and can assist creativity and in this way,the tools were useful for gaining insight into historical architectural data,helped shape a specific design brief,and provoked new ideas.By reflecting on our design process,we argue that the use of language when employing such tools takes a new role and that three layers of language intertwined in our work:architectural discourse,programming languages,and annotations.We present a map that unfolds how these layers came together as a contribution to making machine learning more explainable for creatives.

The sea bass Dicentrarchus labrax as a marine model species in immunology:Insights from basic and applied research

This review summarizes the current knowledge on immune defence activities of the European sea bass Dicentrarchus labrax by reporting the consistent amount of work done on this economically-important species.A draft genome sequence is available for this species,together with whole transcriptomes from lymphoid and non-lymphoid tissues.Available full-length coding sequences of many immunoregulatory and immune-related genes allow for targeted quantitative PCR analysis,nowadays needed for-omics data verification,ex vivo and in vitro.The first anti-T cells monoclonal antibody teleost-wise was obtained in sea bass,followed by several monoclonal and polyclonal markers of lymphocyte populations,namely T cells(pan-T,CD3ε,TcRγ,CD45),and B cells(IgM,IgT,IgD).The combined use of molecular and biochemical tools enabled investigations on innate and acquired immune responses of sea bass in unstimulated/stimulated fish,along the development and under variable environmental conditions and food regimes.An overview of sea bass viral and bacterial pathogens and available vaccines against these microorganisms is also provided.The knowledge accumulated in the past 25 years validates the European sea bass as a reference marine model in the field of fish immunology.

Herpes Simplex Virus Encephalitis Complicated by Acute Ischemic Stroke

Introduction: Herpes simplex virus is the most common etiology for life-threatening sporadic encephalitis worldwide. Antiviral therapy with acyclovir has been shown to reduce mortality and should be started promptly in patients with clinically suspected viral encephalitis before serological confirmation of the diagnosis. Despite antiviral treatment, it is associated with significant mortality and a wide range of neurologic sequelae or neuropsychiatric disorders. Clinical presentation includes fever, headache, altered mental status, and focal or generalized seizures. In some cases, it can present with focal neurological deficits, such as an acute stroke. The aim of this study is to identify rare complications of HSVE. Presentation: We present a case of a 71-year-old female patient with herpes virus encephalitis and an ischemic cerebral accident. The findings of CT scan of the brain revealed an extensive right temporal hypodensity. CSF findings include an elevated protein level, normal glucose level and pleocytosis with lymphocyte predominance. The lumbar tap confirmed the presence of herpes simplex virus type 1 with polymerase chain reaction (PCR) in the CSF. Neurological manifestations include focal neurological deficit with left-sided hemiparesis and coma. After 40 days of complex therapy, an improvement in the mental state was observed. Conclusion: There are varying degrees of neurologic sequelae among survivors in children and adults despite the antiviral treatment. Herpes simplex encephalitis has significant morbidity and high mortality due to the lack of prophylactic treatment and preventable strategies.

Detecting Malicious Uniform Resource Locators Using an Applied Intelligence Framework

The potential of text analytics is revealed by Machine Learning(ML)and Natural Language Processing(NLP)techniques.In this paper,we propose an NLP framework that is applied to multiple datasets to detect malicious Uniform Resource Locators(URLs).Three categories of features,both ML and Deep Learning(DL)algorithms and a ranking schema are included in the proposed framework.We apply frequency and prediction-based embeddings,such as hash vectorizer,Term Frequency-Inverse Dense Frequency(TF-IDF)and predictors,word to vector-word2vec(continuous bag of words,skip-gram)from Google,to extract features from text.Further,we apply more state-of-the-art methods to create vectorized features,such as GloVe.Additionally,feature engineering that is specific to URL structure is deployed to detect scams and other threats.For framework assessment,four ranking indicators are weighted:computational time and performance as accuracy,F1 score and type error II.For the computational time,we propose a new metric-Feature Building Time(FBT)as the cutting-edge feature builders(like doc2vec or GloVe)require more time.By applying the proposed assessment step,the skip-gram algorithm of word2vec surpasses other feature builders in performance.Additionally,eXtreme Gradient Boost(XGB)outperforms other classifiers.With this setup,we attain an accuracy of 99.5%and an F1 score of 0.99.

Aicardi-Goutieres syndrome(AGS)is a systemic inflammatory disorder caused by mutations in any one of the nine different genes,whose deficiency provokes a type I(interferon)IFN response probably central to pathogenesis.^(1) ADAR1,one of the genes mutated in AGS(AGS6),encodes for an enzyme that belongs to the ADAR family(ADAR1,ADAR2,and ADAR3)that catalyzes the conversion of adenosine to inosine within double-stranded RNAs(dsRNAs)(RNA editing A-to-1).

Background Hepatic encephalopathy(HE)is highly prevalent in patients with liver diseases.The pathophysiology of HE is centered on the synergic role of hyperammonemia and systemic inflammation.However,some data suggest altered functioning of the blood–brain barrier(BBB).Assessing BBB function is challenging in clinical practice and at the bedside.Protein-S-100 Beta(PS100-Beta)could be a useful peripheral marker of BBB permeability in HE.This study aimed to assess plasmatic PS100-Beta levels in a prospective cohort of patients admitted to the intensive care unit(ICU)with decompensated cirrhosis with and without overt HE.Methods We retrospectively evaluated a prospective cohort of cirrhotic patients admitted to the ICU from October 2013 to September 2015 that had an available plasmatic PS100-Beta measurement.Patients with previous neurological impairment or limitation of intensive or resuscitative measures were excluded.Overt HE was defined as West-Haven grades 2 to 4.The patients were compared to a control cohort of outpatient clinic cirrhotic and non-cirrhotic patients explored for isolated elevation of liver enzymes.After ICU discharge,the patients were followed for at least 3 months for the occurrence of overt HE.Adverse outcomes(liver transplantation or death)were collected.The ability of PS100-Beta–in combination with other factors–to predict overt HE was evaluated in a multivariate analysis using logistic regression.Likelihood ratios were used to determine the effects and calculate odds ratios(OR).Survival analysis was performed by using the Kaplan–Meier method and survival between groups was compared using a Log-rank test.Results A total of 194 ICU patients and 207 outpatients were included in the study.Increased levels of plasmatic PS100-Beta were detected in the ICU decompensated cirrhotic patients compared with the outpatients([0.15±0.01]mg/L vs.[0.08±0]mg/L,P<0.001).ICU patients with overt HE had higher levels of PS100-Beta([0.19±0.03]mg/L)compared with the ICU patients without overt HE([0.13±0.01]mg/L)(

Bone remodeling:an operational process ensuring survival and bone mechanical competence

Bone remodeling replaces old and damaged bone with new bone through a sequence of cellular events occurring on the same surface without any change in bone shape. It was initially thought that the basic multicellular unit(BMU) responsible for bone remodeling consists of osteoclasts and osteoblasts functioning through a hierarchical sequence of events organized into distinct stages. However, recent discoveries have indicated that all bone cells participate in BMU formation by interacting both simultaneously and at different differentiation stages with their progenitors, other cells, and bone matrix constituents. Therefore,bone remodeling is currently considered a physiological outcome of continuous cellular operational processes optimized to confer a survival advantage. Bone remodeling defines the primary activities that BMUs need to perform to renew successfully bone structural units. Hence, this review summarizes the current understanding of bone remodeling and future research directions with the aim of providing a clinically relevant biological background with which to identify targets for therapeutic strategies in osteoporosis.

Liver cancer is the sixth most commonly diagnosed cancer worldwide,with hepatocellular carcinoma(HCC)comprising most cases.Besides hepatitis B and C viral infections,heavy alcohol use,and nonalcoholic steatohepatitis(NASH)-associated advanced fibrosis/cirrhosis,several other risk factors for HCC have been identified(i.***.old age,obesity,insulin resistance,type 2 diabetes).These might in fact partially explain the occurrence of HCC in non-cirrhotic patients without viral infection.HCC surveillance through effective screening programs is still an unmet need for many nonalcoholic fatty liver disease(NAFLD)patients,and identification of pre-cirrhotic individuals who progress to HCC represents a substantial challenge in clinical practice at the moment.Patients with NASHcirrhosis should undergo systematic HCC surveillance,while this might be considered in patients with advanced fibrosis based on individual risk assessment.In this context,interventions that potentially prevent NAFLD/NASH-associated HCC are needed.This paper provided an overview of evidence related to lifestyle changes(i.***.weight loss,physical exercise,adherence to healthy dietary patterns,intake of certain dietary components,etc.)and pharmacological interventions that might play a protective role by targeting the underlying causative factors and pathogenetic mechanisms.However,well-designed prospective studies specifically dedicated to NAFLD/NASH patients are still needed to clarify the relationship with HCC risk.
