Amazonian Dark Earth and Its Black Carbon Particles Harbor Different Fungal Abundance and Diversity

Amazonian Dark Earth(ADE) is a highly fertile soil of anthropogenic origin characterized by high levels of charred black carbon(BC). It is considered a model of fertility; however, knowledge on the fungal community structure and diversity inhabiting ADE and its BC particles is scarce. Fungal community structure and diversity of ADE and its BC from four sites under different land uses(three agricultural systems and a secondary pristine forest) in the Brazilian Central Amazon were evaluated by 18S rRNA gene pyrosequencing. Fungal communities in ADE and BC were dissimilar and showed differential abundances of fungal operational taxonomic units(OTUs). Estimated fungal species richness(abundance-based coverage estimate and Chao-1 index) and diversity estimators(Shannon and simpson's reciprocal indices) were higher in ADE than in BC in all agricultural areas. No differences were observed in those parameters in ADE and BC samples from the secondary forest. Pezizomycotina fungi and OTUs assigned to Cordyceps confragosa,Acremonium vitellinum, Camarops microspora, and Hirsutella rhossiliensis were more abundant in BC particles than in ADE. These findings represent a breakthrough in our understanding of fungal communities in BC particles from ADE, and will be valuable in future studies considering biochar application in soil.

Background Probiotic supplementation has been used to alleviate abdominal pain in children and adolescents with irritable bowel syndrome(IBS),but the evidence is not ***,a systematic review and meta-analysis of randomized clinical trials(RCTs)were performed to investigate the effects of probiotic supplementation on abdominal pain in pediatric patients with *** PubMed/MEDLIne,Web of Science,Scopus,Cochrane Library,and Embase were the available databases searched to find relevant randomized clinical trials up to April *** effect size was expressed as weighted mean differenee(WMD)and 95% confidence interval(CI).Results Seven RCTs with 441 participants were included,from which the meta-analysis demonstrated that probiotic supplementation has a significant effect on reducing abdominal pain in pediatric patients with IBS(WMD=-2.36:95%CI-4.12 to-0.60;P=0.009).Although our study involved children and adolescents(≤18 years),the effects of probiotic supplementation seem to be more potent in patients under 10 years old(WMD=-2.55;95%CI-2.84 to -2.27)compared to patients aged 10-18 years(WMD=-1.70;95%CI-2.18 to -1.22).The length of supplementation longer than four weeks was more effective(WMD=-2.43;95%CI-2.76 to -2.09).Conclusion Probiotic supplementation can reduce abdominal pain in pediatric patients with IBS.

Interest in lignocellulosic biomass conversion technologies has increased recently because of their potential to reduce the dependency on non-renewable feedstocks. Residues from a variety of crops are the major source of lignocellulose, which is being produced in increasingly large quantities worldwide. The commercial exploitation of crop residues as feedstocks for biorefineries which could be used to produce a variety of goods such as biofuels, biochemicals, bioplastics, and enzymes is an attractive approach not only for adding value to residues but also for providing renewable products required by the expanding bioeconomy market. Moreover, the implementation of biorefineries in different regions has the potential to add value to the specific crop residues produced in the region. In this review, several aspects of crop residue application in biorefineries are discussed, including the role of crop residues in the bioeconomy and circular economy concepts, the main technical aspects of crop residue conversion in biorefineries, the main crop residues generated in different regions of the world and their availability, the potential value-added bioproducts that can be extracted or produced from each crop residue, and the major advantages and challenges associated with crop residue utilization in biorefineries. Despite their potential, most biomass refining technologies are not sufficiently advanced or financially viable. Several technical obstacles, especially with regard to crop residue collection, handling, and pre-treatment, prevent the implementation of biorefineries on a commercial scale. Further research is needed to resolve these scale-up-related challenges. Increased governmental incentives and bioeconomic strategies are expected to boost the biorefinery market and the cost competitiveness of biorefinery products.

AIM： To investigate the potential of bone marrow mononuclear cells （BM-MCs） in the regeneration of hepatic lesions induced by Schistosoma mansoni （***） chronic infection. METHODS： Female mice chronically infected with *** were treated with BM-MCs obtained from male green fluorescent protein （gFP） transgenic mice by intravenous or intralobular injections. Control mice received injections of saline in similar conditions, Enzyme-linked immunosorbent assay （ELISA） assay for transforming growth factor-beta （TgF-β）, polymerase chain reaction （PCR） for gFP DNA, immunofluorescence and morphometric studies were performed. RESULTS： Transplanted gFP^＋ cells migrated to granuloma areas and reduced the percentage of liver fibrosis. The presence of donor-derived cells was confirmed by Fluorescence in situ hybridization （FISH） analysis for detection of cells bearing Y chromosome and by PCR analysis for detection of gFP DNA. The levels of TgF-β, a cytokine associated with fibrosis deposition, in liver fragments of mice submitted to therapy were reduced. The number of oval cells in liver sections of ***-infected mice increased 3-4 fold after transplantation. A partial recovery in albumin expression, which is decreased upon infection with ***, was found in livers of infected mice after cellular therapy. CONCLUSION： In conclusion, transplanted BMCs migrate to and reduce the damage of chronic fibrotic liver lesions caused by ***.

The chronic inflammatory process underlying inflammatory bowel disease (IBD), comprising Crohn's disease and ulcerative colitis, derives from the interplay of several components in a genetically susceptible host. These components include environmental elements and gut microbiota a dysbiosis. For decades, immune abnormalities have been investigated as critically important in IBD pathogenesis, and attempts to develop effective therapies have predominantly targeted the immune system. nevertheless, immune events represent only one of the constituents contributing to IBD pathogenesis within the context of the complex cellular and molecular network underlying chronic intestinal inflammation. These factors need to be appreciated within the milieu of nonimmune components. Damage-associated molecular patterns (DAMPs), which are essentially endogenous stress proteins expressed or released as a result of cell or tissue damage, have been shown to act as direct proinflammatory mediators. Excessive or persistent signalling mediated by such molecules can underlie several chronic inflammatory disorders, including IBD. The release of endogenous DAMPs amplifies the inflammatory response driven by immune and non-immune cells and promotes epigenetic reprogramming in *** effects determine pathologic changes,which may sustain chronic intestinal inflammation and also underlie specific disease *** addition to highlighting the potential use of DAMPs such as calprotectin as biomarkers,research on DAMPs may reveal novel mechanistic associations in IBD pathogenesis and is expected to uncover putative therapeutic targets.

Recent changes occurred in terminus of the debris-covered Bilafond glacier in the Karakoram Range in the Himalayas, Northern Pakistan was investigated in this research. Landsat MSS, TM and ETM+ images were used for this study. Digital elevation models derived from ASTER gDEM and SRTM were also utilized. Visible, infrared and thermal infrared channels were utilized in order to get accurate glacier change maps. Three methods were tried to map this debris-covered glacier in this research. The glacier has been mapped successfully and the changes in the glacier terminus from 1978 to 2011 have been calculated. Manual, semi-automatic and thermal methods were found to give similar results. It was found that the glacier has undergone serious ablation during this period despite of the fact that many of the larger glaciers in the Hindu Kush and Karakoram mountain regions in the Upper Indus Basin were reported to be expanding. The terminus has been moved back about 600 meters during this period and there was an abrupt change in the glacier terminus during 1990-2002. We propose that debris thickness is not the only factor that influences the glacier ablation but the altitude of the debris-covered glacier as well. Many glaciers in the Karakoram region reported to be expanding were having higher altitudes compared to the study area.

Species of Staphylococcus are common in hospital infection (HI). Methicillin resistant S. aureus (MRSA) has also become a serious problem in Brazilian HI. The aim of this study was to characterize the pathogenicity of methicillin-resistant S. aureus (MRSA) and methicillin-sensitive Staphylococcus aureus (MSSA) isolated in public hospitals. The clinical isolates were obtained from intensive care unit. The MRSA and MSSA strains were genotyped by PCR for detection genes related to virulence factors. Moreover, the strains were tested for biofilm formation and cytokine induction in macrophages. Three strains of MRSA (9.68%) expressed the Sea gene, one (3.23%) Seb, 17 (54.84%) Spa and seven (22.58%) had PVL. Two MSSA strains (2.98%) expressed the Sea gene, three (4.48%) Seb, 18 (26.87%) Spa and 11 (16.42%) showed positive results for the PVL gene. There was no expression of Sec and CflA between MRSA and MSSA strains. Among MRSA and MSSA isolates, none statistical differences were observed in biofilm production. The analysis of cytokine induction in the inflammatory response of J774 macrophages by MRSA and MSSA isolates did not show statistical difference. Understanding the mechanisms of pathogenesis of S. aureus could provide important clues for both preventing and treating infection caused by these organisms.

Effects of combined pre- and post-natal protein deprivation on the myenteric plexus of the esophagus of weanling rats:A histochemical,quantitative and ultrastructural study

AIM: To evaluate the effects of protein deprivation on the myenteric plexus of the esophagus of weanling rats. METHODS: Pregnant female Wistar rats were divided into 2 groups: nourished (N),receiving normal diet,and undernourished (D),receiving a protein-deprived diet,which continued after birth. At twenty-one days of age,13 esophagi from each group were submitted to light microscopy and morphometrical analysis employing the NADH diaphorase,NADPH diaphorase and acetylcholinesterase techniques. Three other esophagi from each group were evaluated with transmission electron microscopy (TEM). RESULTS: In both the NADH- and the NADPH-reactive mounts,the neurons of the N mounts were more intensely stained,while in the D esophagi only the larger neurons were reactive. Many myenteric neurons of N were intensely reactive for AChE activity but only a few neurons of D exhibited these aspects. Ultrastructural analysis revealed that the granular reticulum of N showed large numbers of ribosomes aligned on the outer surface of its regularly arranged membrane while the ribosomes of D were disposed in clusters. The chromatin was more homogeneously scattered inside the neuron nucleus of N as well as the granular component of the nucleolus was evidently more developed in this group. Statistically significant differences between N and D groups were detected in the total estimated number of neurons stained by the NADPH technique. CONCLUSION: The morphological and quantitative data shows that feeding with protein-deprived diet in 21-d old rats induces a delay in the development of the myenteric neurons of the esophagus.

Chronic infection with viral hepatitis affects half a billion individuals worldwide and can lead to cirrhosis,cancer,and liver *** cancer is the third leading cause of cancer-associated mortality,of which hepatocellular carcinoma(HCC)represents 90%of all primary liver *** tumors like HCC are complex and have heterogeneous tumor genomic profiles contributing to complexity in diagnosis and *** infection with hepatitis B virus(HBV),hepatitis delta virus(HDV),and hepatitis C virus(HCV)are the greatest etiological risk factors for *** to the significant role of chronic viral infection in HCC development,it is important to investigate direct(viral associated)and indirect(immune-associated)mechanisms involved in the pathogenesis of *** mechanisms used by HBV,HCV,and HDV that drive hepatocarcinogenesis include persistent liver inflammation with an impaired antiviral immune response,immune and viral protein-mediated oxidative stress,and deregulation of cellular signaling pathways by viral *** integration to promote genome instability is a feature of HBV infection,and metabolic reprogramming leading to steatosis is driven by HCV *** current review aims to provide a brief overview of HBV,HCV and HDV molecular biology,and highlight specific viral-associated oncogenic mechanisms and common molecular pathways deregulated in HCC,and current as well as emerging treatments for HCC.
