Targeted regulation of neuroinflammation via nanobiosignaler for repairing the central nerve system injuries

Neuroinflammation,commonly associated with various central nervous system(CNS)diseases such as postoperative cognitive dysfunction(POCD),is primarily mediated by the disruption of biological signals in microglia.However,the effective treatment of CNS diseases remains an ongoing challenge as biological signals show limited microglia-targeting effect.In this study,taking advantage of the highly expressed lipoprotein receptor-related protein-1(LRP1)on the microglia,a nanobiosignal delivery system modified by LRP1 high-affinity peptide ligand RAP12(RAP:receptor-associated protein)was constructed to specifically regulate neuroinflammation via targeting microglia.The uptake of the RAP12 modified-nanobiosignaler by microglia increased significantly,indicating its microglia-targeting ability.Both in vitro/vivo studies proved that the“nanobiosignaler”significantly reduced the secretion of pro-inflammatory cytokines,induced specific M2(anti-inflammatory type)microglia differentiation,and remarkably alleviated cognitive function impairment in the mice model when compared with unmodified groups.It was indicated that the“nanobiosignaler”could target microglia to deliver the biological signal and inhibit the excessive activation of microglia.Overall,the cell-targeted biological signal transmission system inspired by“nanobiosignaler”has broad application prospects in the future.

The global pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)virus has necessitated rapid,easy-to-use,and accurate diagnostic methods to monitor the virus infection.Herein,a ratiometric fluorescence enzyme-linked immunosorbent assay(ELISA)was developed using Si-fluorescein isothiocyanate nanoparticles(FITC NPs)for detecting SARSCoV-2 nucleocapsid(N)protein.Si-FITC NPs were prepared by a one-pot hydrothermal method using 3-aminopropyl triethoxysilane(APTES)-FITC as the Si source.This method did not need post-modification and avoided the reduction in quantum yield and stability.The p-nitrophenyl(pNP)produced by the alkaline phosphatase(ALP)-mediated hydrolysis of pnitrophenyl phosphate(pNPP)could quench Si fluorescence in Si-FITC NPs via the inner filter effect.In ELISA,an immunocomplex was formed by the recognition of capture antibody/N protein/reporter antibody.ALP-linked secondary antibody bound to the reporter antibody and induced pNPP hydrolysis to specifically quench Si fluorescence in Si-FITC NPs.The change in fluorescence intensity ratio could be used for detecting N protein,with a wide linearity range(0.01-10.0 and 50-300 ng/mL)and low detection limit(0.002 ng/mL).The concentration of spiked SARS-CoV-2 N protein could be determined accurately in human serum.Moreover,this proposed method can accurately distinguish coronavirus disease 2019(COVID-19)and non-COVID-19 patient samples.Therefore,this simple,sensitive,and accurate method can be applied for the early diagnosis of SARS-CoV-2 virus infection.

What is already known about this topic?Kashin-Beck disease(KBD)is a chronic and degenerative osteoarthropathy characterized by cartilage degeneration.It is an endemic disease that is highly prevalent among the Chinese population and poses a significant health risk.What is added by this report?This is the first national report on the economic burden of KBD in China.According to the data from 2021,KBD has caused significant disease and economic burdens.The most substantial reduction in healthy life expectancy was observed among patients with degree II severity and those aged 60 years and older,resulting in a total indirect economic burden of 112.74 million Chinese Yuan(CNY).What are the implications for public health practice?The results of this study will contribute to informing the development of tailored prevention and control strategies by the government.These strategies will include targeted policies and recommendations for appropriate healthcare and financial subsidies,which will be based on the demographic characteristics of the endemic areas.

αSetup-PCTL:An Adaptive Setup-Based Two-Level Preconditioner for Sequence of Linear Systems of Three-Temperature Energy Equations

The iterative solution of the sequence of linear systems arising from threetemperature(3-T)energy equations is an essential component in the numerical simulation of radiative hydrodynamic(RHD)problem.However,due to the complicated application features of the RHD problems,solving 3-T linear systems with classical preconditioned iterative techniques is challenging.To address this difficulty,a physicalvariable based coarsening two-level(PCTL)preconditioner has been proposed by dividing the fully coupled system into four individual easier-to-solve subsystems.Despite its nearly optimal complexity and robustness,the PCTL algorithm suffers from poor efficiency because of the overhead associatedwith the construction of setup phase and the solution of subsystems.Furthermore,the PCTL algorithm employs a fixed strategy for solving the sequence of 3-T linear systems,which completely ignores the dynamically and slowly changing features of these linear systems.To address these problems and to efficiently solve the sequence of 3-T linear systems,we propose an adaptive two-level preconditioner based on the PCTL algorithm,referred to as αSetup-PCTL.The adaptive strategies of the αSetup-PCTL algorithm are inspired by those of αSetup-AMG algorithm,which is an adaptive-setup-based AMG solver for sequence of sparse linear systems.The proposed αSetup-PCTL algorithm could adaptively employ the appropriate strategies for each linear system,and thus increase the overall efficiency.Numerical results demonstrate that,for 36 linear systems,the αSetup-PCTL algorithm achieves an average speedup of 2.2,and a maximum speedup of 4.2 when compared to the PCTL algorithm.

目的分析超声引导下细针穿刺细胞学(US-FNAC)在不同大小甲状腺结节诊断中的应用。方法选取2021年1月至2022年6月于该院就诊的80例甲状腺结节患者纳入研究,均行US-FNAC检查,按照结节直径大小分为大于20 mm组、大于10~20 mm组、大于5~10 mm组及小于或等于5 mm组,比较各组患者Bethesda分类,以病理学结果为“金标准”,分析US-FNAC的诊断效能。结果在96个甲状腺结节中,大于20 mm组有23个甲状腺结节,占比23.96%;大于10~20 mm组有35个甲状腺结节,占比36.46%;大于5~10 mm组有27个甲状腺结节,占比28.13%;小于或等于5 mm组有11个甲状腺结节,占比11.46%。对照病理学诊断结果,Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ、Ⅵ类甲状腺结节的恶性率分别为37.50%、26.32%、40.00%、16.67%、71.43%、100.00%,其中,乳头状癌46个(47.92%)、髓样癌2个(2.08%)、滤泡癌9个(9.38%)、未分化癌1个(1.04%);US-FNAC对甲状腺结节大于20 mm组及大于10~20 mm组的诊断准确度均较高,其诊断灵敏度可达100.00%;US-FNAC对甲状腺结节小于或等于5 mm组的诊断准确度相对较低。结论 US-FNAC可用于不同大小甲状腺结节的临床诊断,其对直径大于20 mm及大于10~20 mm甲状腺结节的诊断准确度更高。

共享用工:“狭义借调”中的三方合同构成

"共享用工"是多元化用工体制的重要组成部分,涉及劳动者、用人单位(借出单位)、用工单位(借入单位)三方合同关系,与"借调"和"劳务派遣"在形式上相类似,但又存在诸多不同。从概念的内涵上分析,我国的"共享用工"属于"狭义借调"。三方合同的法律构成诸学说大多以传统的劳资两元主体结构为理论前提,无法有效解释当前多元复杂的劳动合同关系,故而需要从三方合同性质出发,重新认识劳动力与生产资料相结合产生劳动关系理论、使用从属关系(人的从属性)理论及雇主指挥管理权构成理论。在三方合同关系中,共享用工与借调、劳务派遣具有不同的法律属性,共享员工只与用人单位具有一重劳动关系,一般不与用工单位具有默示或双重的劳动关系。共享用工中的雇主责任主要由用人单位承担,用工单位可以行使用人单位让渡的部分指挥命令权,并基于部分使用从属性单独承担或与用人单位共同承担一定的雇主责任。共享用工的实施要由劳资双方达成合意,且需具备生产经营的必要性、人选确定的合理性及合同变更的合法性等条件,并通过严格履行劳动合同变更程序、明确两个企业之间的法律责任与管理权限来防范伪装的共享用工等违法行为。

区别于民事雇佣关系的劳动关系,被认为包含“从属性”之特征,学界主要从人格从属性、经济从属性和组织从属性等方面加以研究,而作为本源性从属和间接性从属的技术从属性一直未受重视。技术要素是形成大工业时代工厂劳动的基本动因,也是雇主在劳动过程中将劳资直接矛盾转移为机器与劳动者间接对立之手段。随着人工智能的发展和算法的运用,本来意义上自主灵活的用工形态受到了技术要素的影响,雇主管控效能得以提高,形成了雇主算法权力。雇主算法权力是劳动管理权的技术构成和范围扩张,其形成和运行有特殊性,由此产生了劳资之间的技术势差。需以劳动关系平衡作为雇主算法权力行使之界限,通过制定系统规则,对雇主算法权力的溢出和滥用加以限制和矫正。算法权力构成应防止劳动的技术异化,算法系统设计应做到透明、知情及可释,算法机制运行应避免劳动歧视、直接决定劳动者重大利益及破坏劳动团结。

目的掌握2020年全国大骨节病病情动态,为控制和消除大骨节病评估提供依据。方法按照《大骨节病监测方案(2019版)》的要求,在全国13个病区省份的所有病区县(市、区、旗)全部病区村进行基本资料收集和儿童病情监测。首先对病区村内全部7~12周岁儿童进行大骨节病临床检查,对临床检查结果疑似大骨节病的儿童拍摄双手X线片。按照《大骨节病诊断》标准(WS/T 207-2010)临床检查及X线复核检查均为阳性者,诊断为大骨节病病例。结果在监测的827986名7~12周岁儿童中,共有703名儿童具有类似大骨节病的临床体征,判断为大骨节病疑似病例。对大骨节病疑似病例的X线复核检查结果显示,703名儿童X线表现均为正常,未有大骨节病X线阳性改变者,未检出儿童大骨节病病例。结论2020年全国未检出儿童大骨节病病例,全国儿童大骨节病病情持续处于消除水平。

Characteristics of parallel-hole and pinhole collimators for nuclear medicine imaging

In this paper,the spatial resolution and geometry sensitivity of parallel- and pin-hole collimators in compact gamma camera are presented,and quantitatively compared by GEANT4 code in Monte-Carlo library.The results show that the geometry sensitivity for pinhole collimator rapidly drops with increasing the gamma source-to- collimator distance(SCD);and for parallel-hole collimator,mildly.Meanwhile,the spatial resolution for pin-hole collimator mildly deteriorates;and for parallel-hole collimator,severely.The pin-hole collimator for close imaging objects has higher geometry sensitivity than parallel-hole collimator.Our findings are helpful for setting compact gamma camera collimators in nuclear medicine imaging.
