Coiled-coil domain-containing 38 is required for acrosome biogenesis and fibrous sheath assembly in mice

During spermiogenesis,haploid spermatids undergo dramatic morphological changes to form slender sperm flagella and cap-like acrosomes,which are required for successful fertilization.Severe deformities in flagella cause a male infertility syndrome,multiple morphological abnormalities of the flagella(MMAF),while acrosomal hypoplasia in some cases leads to sub-optimal embryonic developmental potential.However,evidence regarding the occurrence of acrosomal hypoplasia in MMAF is limited.Here,we report the generation of base-edited mice knocked out for coiled-coil domain-containing 38(Ccdc38)via inducing a nonsense mutation and find that the males are infertile.The Ccdc38-KO sperm display acrosomal hypoplasia and typical MMAF phenotypes.We find that the acrosomal membrane is loosely anchored to the nucleus and fibrous sheaths are disorganized in Ccdc38-KO sperm.Further analyses reveal that Ccdc38 knockout causes a decreased level of TEKT3,a protein associated with acrosome biogenesis,in testes and an aberrant distribution of TEKT3 in sperm.We finally show that intracytoplasmic sperm injection overcomes Ccdc38-related infertility.Our study thus reveals a previously unknown role for CCDC38 in acrosome biogenesis and provides additional evidence for the occurrence of acrosomal hypoplasia in MMAF.

Due to their excellent advantages such as low toxicity,superior optoelectronic properties,low-temperature fabrication,and costeffectiveness,Sn-based perovskites have become the most promising alternatives for high performance lead-free perovskite solar cells.However,the character of Sn ^(2+)is easily oxidized toSn ^(4+),causing unnecessary p-type self-doping and high leakage current.More seriously,trap-induced non-radiative recombination from rapid crystallization causes into large energy loss with a low open circuit voltage.Therefore,the Sn-based solar cells have efficiency far behind the Pb-based solar cells.Herein,the polymer poly(ethylene glycol)diacrylate(PEGDA)is used to control crystal growth and passivate the defects in FA_(0.85)PEA_(0.15)SnI_(3) thin film.This Sn-perovskite layer shows compact crystal with large grain size and reduced defects.Optimized perovskite thin film is further processed to fabricate the inverted solar cell with device structure of ITO(indium tin oxide)/PEDOT:PSS(Poly(3,4-ethylenedioxythiophene)/poly(styrenesulfonate))/FA_(0.85)PEA_(0.15)SnI_(3)/ICBA(indene-C60 bisadduct)/BCP(bathocuproine)/Ag,which shows the power conversion efficiency(PCE)of 11.45%with voltage of 0.82 V.Moreover,corresponding perovskite solar cells exhibit an enhanced stability due to PEGDA induced compressive strain in perovskite.This work could shed light on one of successful attempts to improve Sn-based solar cell efficiency for sustainable energy conversion.

研究不同海拔地区植物共有种叶片化学计量的变化及其影响因素和适应策略,对于理解全球环境变化中的生物地球化学循环具有重要意义。在本研究中,我们测量了中国西北部祁连山5个海拔高度(海拔2400-3200 m,间隔200 m)的土壤有机碳和养分浓度,以及植物共有种的叶片化学计量,以期能更好地了解山区植物共有种如何对海拔变化表现出适应性反应及未来潜在的环境变化如何影响其叶片功能。研究结果表明,随海拔高度的增加,植物共有种叶片C:N:P化学计量的变化不同。在不同的海拔梯度上,年平均气温(MAT)、土壤全磷(STP)、年平均降水量(MAP)、土壤含水量(SWC)和土壤硝态氮(NO3-N)是影响植物共有种叶片元素浓度的主要因素。而叶片化学计量比主要由MAT、MAP和土壤全氮(STN)决定。MAT和MAP对植物共有种叶片元素浓度和叶化学计量比均有显著影响。研究区植物生长主要受到磷的限制。研究结果不仅凸显了植物的可塑性生存策略,还有助于加深我们对植物叶片化学计量学的理解,建立单个植物物种和植物群落与共有种之间的联系。

Bi-Level Programming for the Optimal Nonlinear Distance-Based Transit Fare Structure Incorporating Principal-Agent Game

The urban transit fare structure and level can largely affect passengers’travel behavior and route choices.The commonly used transit fare policies in the present transit network would lead to the unbalanced transit assignment and improper transit resources distribution.In order to distribute transit passenger flow evenly and efficiently,this paper introduces a new distance-based fare pattern with Euclidean distance.A bi-level programming model is developed for determining the optimal distance-based fare pattern,with the path-based stochastic transit assignment(STA)problem with elastic demand being proposed at the lower level.The upper-level intends to address a principal-agent game between transport authorities and transit enterprises pursing maximization of social welfare and financial interest,respectively.A genetic algorithm(GA)is implemented to solve the bi-level model,which is verified by a numerical example to illustrate that the proposed nonlinear distance-based fare pattern presents a better financial performance and distribution effect than other fare structures.

The RNA N^(6)-methyladenosine demethylase ALKBH9B modulates ABA responses in Arabidopsis

The mRNA modification N^(6)-methyladenosine(m^(6)A)plays vital roles in plant development and biotic and abiotic stress responses.The RNA m^(6)A demethylase ALKBH9 B can remove m^(6)A in alfalfa mosaic virus RNA and plays roles in alfalfa mosaic virus infection in Arabidopsis.However,it is unknown whether ALKBH9 B also exhibits demethylation activity and has a biological role in endogenous plant mRNA.We demonstrated here that mRNA m^(6)A modification is induced by the phytohormone abscisic acid(ABA)and that ALKBH9 B has m^(6)A demethylation activity on endogenous mRNA.Knocking out ALKBH9 B led to hypersensitivity to ABA treatment during seed germination and early seedling development.We further showed that ALKBH9 B removes the m^(6)A modification in the ABA INSENSITIVE 1(ABI1)and BRI1-EMS-SUPPRESSOR 1(BES1)transcripts following ABA treatment,affecting the stability of these mRNAs.Furthermore,we determined that ALKBH9 B acts genetically upstream of the transcription factors ABI3 and ABI5,and its regulatory function in ABA responses depended on ABI3 and ABI5.Our findings reveal the important roles of the m^(6)A modification in ABA responses and highlight the role of ALKBH9 Bmediated m^(6)A demethylation in regulating ABA responses post-transcriptionally.

N6-Methyladenosine(m^(6)A)is the most abundant internal chemical modification in eukaryotic mRNA and plays important roles in gene expression regulation,including transcriptional and post-transcriptional regulation.m^(6)A is a reversible modification that is installed,removed,and recognized by methyltransferases(writers),demethylases(erasers),and m^(6)A-binding proteins(readers),respectively.Recently,the breadth of research on m^(6)A in plants has expanded,and the vital roles of m^(6)A in plant development,biotic and abiotic stress responses,and crop trait improvement have been investigated.In this review,we discuss recent developments in research on m^(6)A and highlight the detection methods,distribution,regulatory proteins,and molecular and biological functions of m^(6)A in plants.We also offer some perspectives on future investigations,providing direction for subsequent research on m^(6)A in plants.

Premature ovarian insufficiency(POI) is a heterogeneous female disorder characterized by the loss of ovarian function before the age of 40.It represents a significant detriment to female fertility.However,the known POI-causative genes currently account for only a fraction of cases.To elucidate the genetic factors underlying POI,we conducted whole-exome sequencing on a family with three fertile POI patients and identified a deleterious missense variant in RNF111.In a subsequent replication study involving 1,030 POI patients,this variant was not only confirmed but also accompanied by the discovery of three additional predicted deleterious RNF111 variants.These variants collectively account for eight cases,representing 0.78%of the study cohort.A further study involving 500 patients with diminished ovarian reserve also identified two additional RNF111 variants.Notably,RNF111 encodes an E3 ubiquitin ligase with a regulatory role in the TGF-β/BMP signaling pathway.Our analysis revealed that RNF111/RNF111 is predominantly expressed in the oocytes of mice,monkeys,and humans.To further investigate the functional implications of RNF111 variants,we generated two mouse models:one with a heterozygous missense mutation(Rnf111+/M) and another with a heterozygous null mutation(Rnf111+/-).Both mouse models exhibited impaired female fertility,characterized by reduced litter sizes and small ovarian reserve.Additionally,RNA-seq and quantitative proteomics analysis unveiled that Rnf111 haploinsufficiency led to dysregulation in female gonad development and negative regulation of the BMP signaling pathway within mouse ovaries.In conclusion,our findings strongly suggest that monoallelic deleterious variants in RNF111 can impair female fertility and induce POI in both humans and mice.

创新生态系统是加快形成区域高质量发展经济格局的重要支撑,加速释放的数字生产力为区域创新带来了巨大红利。基于2013—2019年的省级面板数据,使用多重中介模型、调节模型、门槛回归模型及空间模型,结合生态位理论和协同理论等,在创新生态系统视角下探究数字技术与区域创新绩效的关系,得出以下结论:(1)创新生态系统视角下数字技术能够显著提高区域创新绩效,且对技术子系统绩效的影响最大,其次是商业子系统和科学子系统。(2)数字技术影响区域创新绩效的路径中,各子系统的生态位适宜度和子系统间协同不仅存在并行中介作用,也存在链式中介作用。(3)技术子系统和商业子系统的生态位宽度能够正向调节数字技术和区域创新绩效的关系。(4)数字技术发展强化了创新极化的格局。本文拓展了数字技术驱动区域创新相关研究的理论视角,也为各区域通过数字技术塑造多维生态优势提供了参考与启示。

Background:The resurgence of seasonal influenza virus circulation has been seen in 2021-2022 after the tempo-rary suppression in 2020-2021.Neuraminidase inhibitors(NAIs)are widely applied in the clinical treatment of influenza A despite several limitations.Objective:To access the efficacy of Xu’s influenza decoction(XID)in combination therapy with oseltamivir for the treatment of influenza A.Methods:In this retrospective cohort study,the eligible participants were diagnosed with influenza A between June 1,2018,and May 30,2022,in the First Affiliated Hospital of Zhejiang Chinese Medical University.According to whether Xu’s influenza decoction was applied,patients were divided into two groups:treated with or without XID.Propensity score matching(PSM)was used to further adjust the covariates between groups.The primary outcome was to compare time to defervescence via K-M curves,Breslow tests,and Cox regression analysis.In Cox proportional hazards model,a univariate analysis was performed to obtain preliminary results,while a further multivariate analysis was conducted to study the independent factors that influence defervescence.Subgroup analysis was conducted according to body temperature and time from onset to admission.The secondary outcome consisted of routine blood and C-reactive protein(CRP),length of stay,and medical costs.Results:A total of 336 patients with influenza A were enrolled in this study(i.e.,163 patients in the XID+oseltamivir group;173 patients in the oseltamivir group).After 1:1 matching via PSM,230 patients meeting the criteria were included in the analysis,with 115 in each arm.The XID+oseltamivir group had shorter time to defervescence(36 h vs 44 h,P=0.011),shorter length of stay(3 days vs 4 days,P=0.018),and higher defervescence possibility(HR=1.384,95%CI:1.054-1.818).Subgroup analysis indicated that for patients during non-window period(≥48 h)with medium-grade fever(38.1℃-39℃),the XID+oseltamivir combination therapy reduced time to defervescence(P=0.04995/0.004)with a higher defervescence pos
