Peanut allergy is considered to be a major health issue with global effects.To date,no effective curative approach has been applied for the therapy of the anaphylaxis resulting from the peanut allergens.The accurate and effective detection methods for the surveillance of allergens in food are still the primary strategies to avoid allergic diseases.In this study,nanobodies(Nbs)derived from the Heavy-Chain only Antibodies(HCAbs)were selected against the general peanut protein extract through the unbiased strategy to facilitate the development of the sandwich ELISA for the detection and surveillance of peanut allergen contamination.The target antigen of the selected Nb was identified as peanut allergen Ara h 3,and a cross-reaction was observed with the member of Gly 1 from the Ara h 3 family.The applicability of the self-paired Nb P43 on the establishment of the immuno-assay was verified.A sandwich ELISA against peanut allergen was developed,which reached a linear range of 0.2-10.6μg/mL,and a limit of detection of 53.13 ng/mL.

In this paper,the hydrodynamic modeling and parameter identifcation of the RobDact,a bionic underwater vehide inspired byDactylopteridae,are carried out based on computational fluid dynamics(CFD)and force measurement experiment.Firstly,thePaper briely describes the RobDact,then establishes the kinematis model and rigid body dynamics model of the RobDactaccording to the hydrodynamic force and moment equations.Through CFD simulations,the hydrodynamic force of theRobDact at diferent speeds is obtained,and then,the hydrodynamic model parameters are identified.Furthermore,themeasurement platform is developed to obtain the relationship between the thrust generated by the RobDact and the inputfluctuation parameters,Finally,combining the rigid body dynamics model and the fin'thrust mapping model,thehydrodynamic model of the RobDact at diferent motion states is constructed.

In this paper,the research on ACSR temperature of broken strand and fatigue damage after broken strand is carried out.Conduct modeling and Analysis on the conductor through AnsoftMaxwell software.The distribution of magnetic force lines in the cross section of the conductor after strand breaking and the temperature change law of the conductor with the number of broken strands are analyzed.A model based on electromagnetic theory is established to analyze the distribution of magnetic lines of force in the cross section of the conductor after strand breaking and the temperature variation law of the conductor with the number of broken strands.The finite element analysis results show that with the increase in the number of broken strands,the cross-sectional area of the conductor decreases,the magnetic line of force of the inner conductor at the broken strand becomes denser and denser,and the electromagnetic loss of the conductor becomes larger and larger.Therefore,the temperature of the conductor at the broken strand becomes higher and higher.Then,the current carrying experiment of conductor is carried out for LGJ-240/30 conductor.It is found that the temperature rise at the junction of inner and outer layers at the broken strand is particularly obvious,and the temperature of inner aluminumconductor at the broken strand also increases with the increase of broken strand.According to the analysis of experimental data,with the increase of broken strands,the antivibration ability and service life of the conductor decrease.At the same time,under certain conditions of broken strand,the fatigue life of conductor increases with the increase of temperature.

随着乌克兰危机的不断升级,欧洲诸国纷纷对俄罗斯进行制裁,只有塞尔维亚成了反俄浪潮中的“欧洲孤岛”。作为一直寻求正式加入欧盟的候选国,其支持俄罗斯的立场值得深究。同时,主流的理性选择理论提供的解释与该事件现实之间存在张力,群体情感理论则提供了一种更加贴切的分析框架。具体来说,塞尔维亚国内的政治基调情感形塑着其在乌克兰危机中立场偏好的基本方向,武契奇政府调动的政治即时情感进一步加深其国内民众的亲俄情结,以及俄罗斯和欧美对塞尔维亚的情感评价手段形成鲜明的对比,导致塞尔维亚官方在乌克兰危机中的立场偏好口头上支持欧盟,实际行动上更加倾向于俄罗斯。

Transcription activator-like effectors （TALEs） that were related to bacteria immune system have lately been employed in a promising approach of precise gene targeting. Because of the repetitive characteristics of TALEs, existing TALE assembly methods are either very complicated, time-consuming, or too tricky to be handled in common labs. Here, we reported a rapid, efficient and easy method for TALE assembly. This method takes advantage of uracil-specific excision reagent （USER）, an enzyme that can cleave DNA constructs and create long, unique single-strand DNA overhangs. Upon USER treatment, the overhangs on each individual TALE repeat unit can be rejoined hierarchically to form pentamers in a ligation-independent manner. Eventually, three pentamers are assembled into a full TALE construct by Golden Gate cloning. TALE nucleases （TALENs） generated with this method exhibit high genome-editing activity in human cells such as HEK293FT cells. Using this method, we have successfully synthesized three TALEN pairs targeting endogenous Tetl locus, and proved that all can specifically target Tetl gene, though in various degree. Comparing to other methods of TALEN assembly, this one is much less labor intensive and fairly faster, and positive clones can be obtained at high efficiency within only two days. We thus contribute to an easier approach for effective TALENs synthesis, which may highly facilitate the wide application of TALEN technology in genome editing, especially for human cells that require precise targeting.

心脑血管疾病是严重威胁人类健康的常见疾病,二者之间存在着密切而广泛的联系,从而开启了一门新的交叉学科“神经心脏病学”。目前尚缺乏神经心脏病学的确切定义,该学科主要的研究内容包括了共同因素导致的神经和心血管系统损害(包括动脉粥样硬化性心血管病、异常物质沉积和遗传代谢病、获得性以及遗传性易栓症、系统性疾病等)、神经精神系统异常导致心血管系统损害(包括急性神经系统病变和心理应激、神经调节机制、神经肌肉病等)和心血管系统异常导致神经系统损害(包括结构性心脏病、节律性心脏病、心腔内血栓以及占位、大血管病变等)。本文简要阐述了神经心脏病学的相关研究进展,指出应该加强对这一交叉学科的认识,推动学科发展,造福患者。

随着脑血管影像技术的普及, 未破裂颅内动脉瘤(UIA)临床检出率越来越高。UIA破裂风险低, 但是一旦破裂, 严重影响人类健康。UIA治疗存在较大争议, 引起临床广泛关注。中华医学会神经外科学分会、中国卒中学会脑血管外科分会、国家神经系统疾病医学中心与国家神经系统疾病临床研究中心联合制订《中国未破裂颅内动脉瘤临床管理指南(2024版)》, 该指南采用模块化形式, 突出推荐意见, 并指出当前研究不足及未来研究方向, 从UIA流行病学、人群筛查、临床影像诊断、破裂风险评估、治疗决策及选择, 术后随访及长期管理等方面提供了全面的临床管理意见。将证据来源分为中国人群和其他人群, 有助于指导国内临床实践。

Integrated computational materials engineering(ICME)has emerged to be one of the most powerful materials genome engineering(MGE)approaches in designing new materials and manufacturing processes in recent years.It has successfully deployed many new products for the electronic,automotive,and aerospace industries.This paper reviews the current status of research on first principles in the design of high-strength Mg alloys,discusses the application of crystal plasticity finite element models to the microscale slip,twinning,microstructure morphology,texture evolution,and macroscopic forming of Mg alloys,and introduces the research progress of crystal plasticity finite element models and phase field models,meta cellular automata models and first principles coupled models respectively,around the need for multi-scale coupled simulations of Mg alloys.The key technology obstacles of integrating the first principles,crystal plasticity finite element,and microstructure models for Mg alloys have been solved.This paper can provide a reference for the design of new Mg alloy compositions and the development of high-performance Mg alloys.

Photothermal therapy(PTT)is a treatment that increases the temperature of tumors to 42–48℃,or even higher for tumor ablation.PTT has sparked a lot of attention due to its ability to induce apoptosis or increase sensitivity to chemotherapy.Excessive heat not only kills the tumor cells,but also damages the surrounding healthy tissue,reducing therapeutic accuracy and increasing the possible side effects.Herein,a phase change fiber(PCF)scaffold serving as a thermal trigger in mild photothermal–chemo tumor therapy is developed to regulate temperature and control drug release.These prepared PCFs,comprised of hollow carbon fibers(HCFs)loaded with lauric acid as a phase change material(PCM),can effectively store and release any excess heat generated by irradiating with a near-infrared(NIR)laser through the reversible solid–liquid transition process of the PCM.With this feature,the optimal PTT temperature of implanted PCF-based composite scaffolds was identified for tumor therapy with minimal normal tissue damage.In addition,controlled release of chemotherapeutic drugs and heat shock protein(HSP)inhibitors from the PCF-based composite scaffolds have been shown to improve the efficacy of mild PTT.The developed PCF-based scaffold sheds light on the development of a new generation of therapeutic scaffolds for thermal therapy.

A Surrogate-Assisted Evolutionary Algorithm for Space Component Thermal Layout Optimization

In space engineering,the electronic component layout has a very important impact on the centroid stability and heat dissipation of devices.However,the expensive thermodynamic simulations in the component thermal layout optimization problems bring great challenges to the current optimization algorithms.To reduce the cost,a surrogate-assisted evolutionary algorithm with restart strategy is proposed in this work.The algorithm is consisted of the local search and global search.A restart strategy is designed to make the local search jump out of the local optimum promptly and speed up the population convergence.The proposed algorithm is compared with two state-of-the-art algorithms on the CEC2006,CEC2010,and CEC2017 benchmark problems.The experiment results show that the proposed algorithm has a high convergence speed and excellent ability to find the optimum in the expensive constrained optimization problems under the very limited computation budget.The proposed algorithm is also applied to solve an electronic component layout optimization problem.The final results demonstrate the good performance of the proposed algorithm,which is of great significance to the component layout optimization.
