Mesoporous silica nanoparticles with chiral pattern topological structure function as“antiskid tires”on the intestinal mucosa to facilitate oral drugs delivery

The weak adhesion between nanocarriers and the intestinal mucosa was one of the main reasons caused the failure in oral delivery.Inspired by the“antiskid tires”with complex chiral patterns,mesoporous silica nanoparticles AT-R@CMSN exhibiting geometrical chiral structure were designed to improve the surface/interface roughness in nanoscale,and employed as the hosting system for insoluble drugs nimesulide(NMS)and ibuprofen(IBU).Once performing the delivery tasks,AT-R@CMSN with rigid skeleton protected the loaded drug and reduced the irritation of drug on gastrointestinal tract(GIT),while their porous structure deprived drug crystal and improved drug release.More importantly,AT-R@CMSN functioned as“antiskid tire”to produce higher friction on intestinal mucosa and substantively influencedmultiple biological processes,including“contact”,“adhesion”,“retention”,“permeation”and“uptake”,compared to the achiral S@MSN,thereby improving the oral adsorption effectiveness of such drug delivery systems.By engineering AT-R@CMSN to overcome the stability,solubility and permeability bottlenecks of drugs,orally administered NMS or IBU loaded AT-R@CMSN could achieve higher relative bioavailability(705.95%and 444.42%,respectively)and stronger anti-inflammation effect.In addition,AT-R@CMSN displayed favorable biocompatibility and biodegradability.Undoubtedly,the present finding helped to understand the oral adsorption process of nanocarriers,and provided novel insights into the rational design of nanocarriers.

Mechanical intestinal obstruction due to isolated diffuse venous malformations in the gastrointestinal tract: A case report and review of literature

BACKGROUND Isolated gastrointestinal venous malformations(GIVMs)are extremely rare congenital developmental abnormalities of the venous vasculature.Because of their asymptomatic nature,the diagnosis is often quite challenging.However,as symptomatic GIVMs have nonspecific clinical manifestations,misdiagnosis is very common.Here,we report a case of isolated diffuse GIVMs inducing mechanical intestinal obstruction.A literature review was also conducted to summarize clinical features,diagnostic points,treatment selections and differential diagnosis in order that doctors may have a comprehensive understanding of this disease.CASE SUMMARY A 50-year-old man presented with recurrent painless gastrointestinal bleeding for two months and failure to pass flatus and defecate with nausea and vomiting for ten days.Digital rectal examination found bright red blood and soft nodular masses 3 cm above the anal verge.Computed tomography showed that part of the descending colon and rectosigmoid colon was thickened with phleboliths in the intestinal wall.Colonoscopy exhibited bluish and reddish multinodular submucosal masses and flat submucosal serpentine vessels.Endoscopic ultrasonography showed anechoic cystic spaces within intestinal wall.The lesions were initially thought to be isolated VMs involving part of the descending colon and rectosigmoid colon.Laparoscopic subtotal proctocolectomy,pullthrough transection and coloanal anastomosis and ileostomy were performed.Histopathology revealed intact mucosa and dilated,thin-walled blood vessels in the submucosa,muscularis,and serosa involving the entire colorectum.The patient recovered with complete symptomatic relief during the 52-mo follow-up period.CONCLUSION The diagnosis of isolated GIVMs is challenging.The information presented here is significant for the diagnosis and management of symptoms.

Genetic risk factors have been shown to contribute to the development of sexual dysfunction.However,the role of methylenetetrahydrofolate reductase(MTHFR)gene variants in the risk of erectile dysfunction(ED)remains unclear.In this study,we recruited 1254 participants who underwent ED assessed by the International Index of Erectile Function-5.The MTHFR c.677C>T variant was also measured by fluorescence polymerase chain reaction(PCR).No significant difference in the genotypic frequency of the MTHFR C677T polymorphism(CC,CT,and TT)was observed between men from the ED and non-ED groups.In addition,on binary logistic regression analysis,both crude and adjusted models showed that the risk of ED was not significantly associated with the C677T polymorphism.Interestingly,a significantly higher frequency of the 677TT polymorphism was found in severe and moderate ED(P=O.02).The positive correlation between the MTHFR 677TT polymorphism and severe ED was confirmed by logistic regression analysis,even after adjusting for potential confounders(odds ratio[OR]=2.46,95%confidence interval[CI]:1.15-5.50,P=0.02).These findings suggest a positive correlation between the MTHFR 677TT polymorphism and the risk of severe ED.Identification of MTHFR gene polymorphisms may provide complementary information for ED patients during routineclinicaldiagnosis.

Efficient micrometer-scale thick-film perovskite solar cells with superior stability

It is essential to enhance the thickness of the absorber layer for perovskite solar cells(PSCs) to improve device performance and reduce industry refinement.However,thick perovskite films(> 1 μm) are difficult to be fabricated by employing traditional solvents,such as N,N-dimethylformamide(DMF),dimethyl sulfoxide(DMSO).Besides,it is a challenge to fabricate thick-film perovskite owing to the deteriorated surface morphology and serious defect density.Herein,a simple method was reported to deposit uniform pinhole-free perovskite films with a thickness of more than 2 μm utilizing the methylamine acetate(MAAc) ionic liquid as the solvent.Combined with methylammonium chloride(MACl) as an additive,thick-film perovskite with～ 2μm in grain size and few grain boundaries(GBs) was prepared,which dramatically improved the perovskite crystal quality and enhanced carrier transport performance.The final PSCs exhibited a power conversion efficiency(PCE) of 20.16%.The device showed improved stability with 95% of its initial efficiency in a nitrogen environment over 5000 h.This work provides an alternative strategy to produce extremely efficient and stable thick-film PSCs.It can be believed that this device has great potential in the application of large areas and laminated PSCs.

粮食价格、劳动力转移与农民收入增长 --基于中国省际面板数据的实证分析

基于2004~2017年中国大陆31省(市、区)数据,通过面板数据模型实证分析粮食价格、劳动力转移对农民收入影响的结构效应、区域异质性和门槛效应。结果显示:(1)粮食价格和劳动力转移能促进农民收入增长。其中粮食价格能显著促进农民经营性和工资性收入,劳动力转移能显著促进工资性收入,但对经营性收入有显著抑制作用。(2)东中西部地区的粮食价格能显著促进农民增收,其中东部作用最强,西部最弱,中西部地区劳动力转移能显著促进农民增收,而东部作用不明显。(3)粮食价格和劳动力转移对农民收入的促进作用存在门槛效应,近似地表现为倒“U”型关系的左半支。据此,提出建立健全粮食价格稳定长效机制,加快实行城乡统筹就业政策,重视农业生产机械化和技术创新等建议。

目的:探讨干扰素-γ(IFN-γ)刺激后人巨噬细胞内上调的转录组基因表达谱及其相应的细胞内信号调控机制。方法:运用RNA-seq对IFN-γ刺激前后的人巨噬细胞系U937差异基因表达谱进行测序和比较,从中筛选出显著上调的基因,利用实时荧光定量PCR技术(qPCR)分别在U937和THP1两个巨噬细胞株内进行验证;在IFN-γ刺激U937细胞的同时,分别加入JAK/STAT3、MAPK/ERK和PI3K/AKT通路抑制剂,检测其对上调基因的影响,从中找出关键的调控机制。结果:巨噬细胞经IFN-γ刺激后,RNA-seq筛选结合qPCR验证结果显示:趋化因子CXCL9、CXCL10、CXCL11,APOL家族包括APOL1、APOL2、APOL3、APOL4、APOL6以及GBP家族GBP1、GBP2、GBP3、GBP4、GBP5均显著上调。JAK/STAT3通路抑制剂抑制了IFN-γ诱导的APOL1、APOL4、GBP1、GBP4和GBP5基因上调表达,MAPK/ERK通路抑制剂抑制了IFN-γ诱导的CXCL10基因上调表达,PI3K/AKT通路抑制剂抑制了IFN-γ诱导的APOL1、APOL4、APOL6、GBP1和GBP5基因上调表达,三种信号通路抑制剂均能抑制IFN-γ诱导的CXCL9基因上调表达,三种通路抑制剂对IFN-γ诱导的APOL3基因上调均无抑制作用。结论:IFN-γ刺激后巨噬细胞内APOL和GBP部分家族分子表现为显著上调,PI3K/AKT、JAK/STAT3和MAPK/ERK通路对其表达分别具有一定的正向调控作用。

B细胞淋巴瘤/白血病是恶性淋巴瘤中最常见的亚型,其复发和难治常常是导致治疗失败的主要原因。长期以来,手术、放疗、化疗和姑息治疗等作为传统的抗肿瘤治疗模式,挽救了许多患者的生命,但每年仍然有大量患者因肿瘤不治而死亡。B细胞淋巴瘤/白血病,尤其是表达CD20的成熟B细胞淋巴瘤/白血病,在利妥昔单抗问世之后,其治愈率有了大幅度的上升,然而仍然有近20%-40%患者因复发和难治而去世。近5年来,随着可特异性识别B细胞表面CD19的嵌合抗原受体的T细胞(CAR-T)的发展,尤其是CD19 CAR-T细胞免疫治疗在针对复发和难治B细胞淋巴瘤的临床试验中取得了非常显著的疗效,CAR-T细胞免疫治疗便逐渐受到广大研究者和临床学家的重视,包括我国在内的全球多家医疗机构纷纷注册和开展了针对B细胞淋巴瘤/白血病的临床试验。本文就CD19 CAR-T细胞在治疗B细胞淋巴瘤/白血病过程中的发展历史,在前进行中的主要的临床试验和已经证实的主要潜在不良反应作一综述。

Inhibitory　Effects　of　Chlorpheniramine　and　Astemizolum　on　Contraction　of　Isolated　Rat　and　Rabbit　Uteri　Induced　by　Oxytocin　an

The contraction of isolated rat and rabbit uteri induced by oxytocin and PGF2α was markedly inhibited by chlorpheniramine (Chl) and astemizolum (Ast) , both of which also decreased the resting tension of uteri, and their spontaneous contraction. The inhibitory effects of both drugs were dose-dependent. At high concentrations , Chl 7. 4 × 10-4 mol/L and Ast 10-4 mol/L could counteract the contraction of the uteri induced by Oxy and PGF2α, and their spontaneous contraction as well. They decreased the resting tension to the lower level. The mechanism of their non-special relaxed action on uteri could not be completely explained only by their H1-receptor blocking action. Whether they act by blocking calcium channel or by inhibiting calmodulin (CaM) remains to be further explored.

Photonic quantum computation plays an important role and offers unique advantages.Two decades after the milestone work of Knill-Laflamme-Milburn,various architectures of photonic processors have been proposed,and quantum advantage over classical computers has also been demonstrated.It is now the opportune time to apply this technology to real-world applications.However,at current technology level,this aim is restricted by either programmability in bulk optics or loss in integrated optics for the existing architectures of processors,for which the resource cost is also a problem.Here we present a von-Neumann-like architecture based on temporal-mode encoding and looped structure on table,which is capable of multimode-universal programmability,resource-efficiency,phasestability and software-scalability.In order to illustrate these merits,we execute two different programs with varying resource requirements on the same processor,to investigate quantum signature of chaos from two aspects:the signature behaviors exhibited in phase space(13 modes),and the Fermi golden rule which has not been experimentally studied in quantitative way before(26 modes).The maximal program contains an optical interferometer network with 1694 freely-adjustable phases.Considering current state-of-the-art,our architecture stands as the most promising candidate for real-world applications.
