BACKGROUND: Hypersecretion of biliary cholesterol is believed to be one of the important causes of lithogenic bile. Sterol carrier protein-2 ( SCP2 ) participates in choles- terol trafficking and metabolism and may play a key role in cholesterol gallstone formation. This study was undertaken to investigate the expression of liver SCP2 mRNA in pa- tients with cholesterol gallstone and those patients with non-cholesterol gallstone. METHODS: The expression of liver SCP2 mRNA was studi- ed in 36 patients with cholesterol gallstone and 30 patients with non-cholesterol gallstone by reverse transcription-poly- merase chain reaction (RT-PCR). RESULT: The expression of SCP2 mRNA was increased more significantly in patients with cholesterol gallstone than in patients with non-cholesterol gallstone. CONCLUSION: The SCP2 gene was overexpressed in pa- tients with cholesterol gallstone, indicating that SCP2 may be one of the important causes of cholesterol gallstone.

Vaccinations are essential for preventing and treating disease,especially cancer nanovaccines,which have gained considerable interest recently for their strong anti-tumor immune *** can prompt the immune system to generate antibodies and activate various immune cells,leading to a response against tumor tissues and reducing the negative effects and recurrence risks of traditional chemotherapy and *** enhance the flexibility and targeting of vaccines,nanovaccines utilize nanotechnology to encapsulate or carry antigens at the nanoscale level,enabling more controlled and precise drug delivery to enhance immune *** nanovaccines function by encapsulating tumor-specific antigens or tumor-associated antigens within *** small size of these nanomaterials allows for precise targeting of T cells,dendritic cells,or cancer cells,thereby eliciting a more potent anti-tumor *** this paper,we focus on the classification of carriers for cancer nanovaccines,the roles of different target cells,and clinically tested cancer nanovaccines,discussing strategies for effectively inducing cytotoxic T lymphocytes responses and optimizing antigen presentation,while also looking ahead to the translational challenges of moving from animal experiments to clinical trials.

In the field of intelligent education,the integration of artificial intelligence,especially deep learning technologies,has garnered significant *** tracing(KT)plays a pivotal role in this field by predicting students’future performance through the analysis of historical interaction data,thereby assisting educators in evaluating knowledgemastery and tailoring instructional *** knowledge tracingmethods,largely based on Recurrent Neural Networks(RNNs)and Transformer models,primarily focus on capturing long-term interaction patterns in sequential ***,these models may neglect crucial short-term dynamics and other relevant *** paper introduces a novel approach to knowledge tracing by leveraging a pure Multilayer Perceptron(MLP)*** proposeMixerKT,a knowledge tracing model based on theHyperMixer framework,which uniquely integrates global and localMixer feature *** architecture enables more effective extraction of both long-terminteraction trends and recent learning behaviors,addressing limitations in currentmodels thatmay overlook these key *** evaluations on twowidely-used datasets,ASSIS Tments2009 and Algebra2005,demonstrate that MixerKT consistently outperforms several state-of-the-art models,including DKT,SAKT,and Separated Self-Attentive Neural Knowledge Tracing(SAINT).Specifically,MixerKT achieves higher prediction accuracy,highlighting its effectiveness in capturing the nuances of learners’knowledge *** results indicate that our model provides a more comprehensive representation of student learning patterns,enhancing the ability to predict future performance with greater precision.

The combination of photodynamic therapy（PDT） and chemodynamic therapy（CDT） in tumor treatment has attracted considerable ***,tumor hypoxia and glutathione（GSH） overproduction in the tumor tissue restricted the progress of their ***,a Mn-porphyrinic metal-organic framework（Mn-TCPP） was constructed by the one-pot method and further used for immobilizing glucose oxidase（GOx） to obtain GOx@*** would rapidly exhaust endogenous glucose into hydrogen peroxide（H2O2） and gluconic acid,thus shutting off the energy supply of tumor cells for starvation ***-TCPP catalyzed H2O2to produce oxygen,regulating the hypoxic tumor microenvironment and in turn improving1O2generation under laser ***,Mn-TCPP can reduce reactive oxygen species consumption owing to the redox reaction between Mn3+and GSH,thus greatly enhancing ***,benefiting from GOx-mediated starvation therapy,Mn2+produced by Mn3+reduction can react with sufficient intracellular H2O2to generate ·OH with high cytotoxicity through a Fenton-like *** treatment by GOx@Mn-TCPP under laser irradiation in vitro,the cell viability of 4T1 and A549 tumor cells reached to 20%,reflecting GOx@Mn-TCPP could give full play to the advantages of PDT/CDT/starvation *** results in vivo demonstrated that GOx@Mn-TCPP mediated synergistic cascade therapy could significantly inhibit tumor growth and improve the therapeutic effect.

Mitochondrial Rho-GTPase 1 (MIRO1) is an outer mitochondrial membrane protein which regulates mitochondrial transport and mitophagy in mitosis. In present study, we reported the crucial roles of MIRO1 in mammalian oocyte meiosis and its potential relationship with aging. We found that MIRO1 expressed in mouse and porcine oocytes, and its expression decreased in aged mice. MIRO1 deficiency caused the failure of meiotic resumption and polar body extrusion in both mouse and porcine oocytes, which could be rescued by exogenous MIRO1supplementation. Mass spectrometry data indicated that MIRO1 associated with several cytoskeleton and cell cycle-related proteins, and MIRO1 regulated motor protein Dynein for microtubule-organizing centers (MTOCs) dynamics at germinal vesicle (GV) stage, which determined meiotic resumption. Furthermore, we found that MIRO1 regulated Aurora A and kinesin family member 11 (KIF11) for meiotic spindle assembly in oocytes. Besides, MIRO1 associated with several mitochondria-related proteins dynamic-related protein 1 (DRP1),Parkin and lysosomal-associated membrane protein 2 (LAMP2) for mitochondrial dynamics and mitophagy during oocyte meiosis. Taken together, our results suggested that MIRO1 played pivotal roles in meiotic resumption, spindle assembly and mitochondrial function in mouse and porcine oocytes, and its insufficiency might contribute to the oocyte maturation defects during aging.

Surveillance recommendations for gastric cancer（GC） in current guidelines focused on advanced precancerous lesions and were based on precise diagnosis of severity/extent of baseline lesions. We aimed to develop a less endoscopy-related equipment-dependent risk-stratification tool, and assessed whether mild-precursor-lesion patients can be safely exempt from surveillance. In the multicenter communitybased cohort, 75,051 participants receiving baseline endoscopy were enrolled during 2015–2017 and followed-up until 2021. Cumulative incidence rates（CIRs） of GC for precancerous-conditions were calculated by Kaplan-Meier method and compared by Log-rank tests. Mixedeffects Cox regression models were used to detect potential factors for progression towards GC. A risk score was calculated as counts of selected factors. An independent cohort, including 26,586 participants was used for external validation. During a median follow-up of 6.25 years, CIRs of GC were 0.302%, 0.436%, and 4.756% for normal group, non-neoplastic（atrophic gastritis/intestinal metaplasia） and neoplastic lesions（low-grade/high-grade dysplasia）, respectively(Ptrend<0.001). Four predictors, including male, ≥60 years, smoking, and limited vegetable consumption, were selected for risk-stratification. High-risk patients（≥3 risk factors） with non-neoplastic lesions showed higher GC risks（adjusted HR=7.73, 95%CI: 4.29–13.92）, and their four-year CIR reached the one-year CIR of neoplastic lesions. Further categorizing non-neoplastic lesions by histological grade, both patients with moderate-to-severe lesions（aHR=3.07, 95%CI: 1.67–5.64） and high-risk patients with mild lesions（aHR=7.29, 95%CI: 3.58–14.86） showed higher risks. Consistent trends were observed in validation cohort. High-risk mild-precursor-lesion patients should receive surveillance within 3–5 years after baseline screening. Our study provides evidence on supplementing current guideline recommendations.

目的:探讨肠型白塞氏病的临床特点、病理学特征及鉴别诊断要点.方法:收集肠型白塞氏病确诊患者3例,进行临床资料复习和组织病理学检查.患者均以消化系症状入院,同时患者伴有白塞氏病的主要临床症状.结果:患者溃疡病变多发生于肠系膜附着的对侧,溃疡呈圆形,较深;镜下见肠黏膜组织溃疡形成,周围黏膜下淋巴组织增生显著,主要表现以静脉为主的血管炎症.治疗上,糖皮质激素及免疫抑制剂是基础用药,其预后不令人满意.结论:肠型白塞氏病早期诊断有赖于对病史及临床症状的综合分析.肠型白塞氏病无特异性治疗方法,除非合并严重并发症,一般不主张手术治疗.

As a potential renewable and clean energy source,the distribution and exploration prospects of natural hydrogen have garnered widespread ***,the limitations in understanding the formation mechanisms and accumulation patterns of underground natural hydrogen have significantly constrained its efficient exploration and *** study reports H2content and isotopes in natural gas from deep formations in sedimentary basins through geochemical analysis of 58 conventional gas samples from the Sinian to Triassic strata in the central Sichuan *** discovered that H2content in deep natural gas in central Sichuan generally exceeds 0.1%,with a maximum of 1.6%,and hydrogen isotopes (δ2HH2) range from-850‰to-586‰.Analysis of the geological conditions,hydrogen generation mechanisms,and isotope fractionation characteristics in the study area confirms that microbial fermentation or in-situ organic matter cracking are not effective pathways for deep *** with helium content and isotope analysis of the natural gas,it is inferred that the volcanic basement in the central Sichuan area is the potential source rock for natural H2in the Sinian to Triassic ***,H2originates from the radiolysis of water and hydrothermal reactions of Fe（II） in the basement ***,a diffusion fractionation model reveals significant isotope fractionation during the migration and accumulation of deep H2into sedimentary reservoirs,which lead to anomalously light δ2HH2in deep natural gas in central *** indicates that the underground natural hydrogen accumulation is in a long-term dynamic process,with simultaneous H2accumulation and dissipation in the gas *** research not only elucidates the formation mechanism of deep H2but also provides insights for the resource potential of natural hydrogen in sedimentary basins.
