Macrophages-regulating nanomedicines for sepsis therapy

Sepsis is the leading cause of death in intensive care unit(ICU), which is caused by deregulated immune responses to pathogens infection. Clinically, sepsis treatment is limited to antibiotics and supportive care, while there still lacks of specific molecular therapy. As a type of immune dysfunction disease,macrophages have been recognized as the key immune cells precipitating in the whole process of sepsis,which is activated into M1-like to trigger various inflammatory responses at early stage whereas polarized into M2-like to cause immunosuppression in later stage. Therefore, great attention has been paid on the design of nanomedicines to regulate the functions of macrophages for etiological treatment of sepsis, by virtue of the unique advantages of nano-drug delivery systems, such as enhanced drug bioavailability, targetability, reduced side-effects. This critical review aims to summarize the recent progress of macrophages-regulating nanoparticles for sepsis therapy. First, the essential roles of macrophages in the development and progression of sepsis have been introduced, including the positive roles of macrophages to combat infections and dysfunction of macrophages to cause body damages. We then focus our main attention to discuss the nanomedicines with different therapeutic mechanisms corresponding to each stage of sepsis, such as infection blockage, inflammation inhibition, immune functions recovery, as well as multifunctional nanomedicines. Finally, a few limitations of current nanomedicines are highlighted,and future perspective are speculated for potential clinical translation, which might pave the way for the development of macrophages-centered nanomedicines for more effective sepsis therapy.

Vibration Serviceability of Large-Span Steel–Concrete Composite Beam with Precast Hollow Core Slabs Under Walking Impact

A large-span steel–concrete composite beam with precast hollow core slabs(CBHCSs)is a relatively new floor structure that can be applied to various long-span structures.However,human-induced vibrations may present serviceability issues in such structures.To alleviate vibrations,both the walking forces excited by humans and the associated floor responses must be elucidated.In this study,150 load–time histories of walking,excited by 25 test participants,are obtained using a force measuring plate.The dynamic loading factors and phase angles in the Fourier series functions for one-step walking are determined.Subsequently,walking tests are performed on seven CBHCS specimens to capture the essential dynamic properties of mode shapes,natural frequencies,damping ratios,and acceleration time histories.The CBHCS floor system generally exhibits a high frequency(>10 Hz)and low damping(damping ratio<2%).Sensitivity studies using the finite element method are conducted to investigate the vibration performance of the CBHCS floor system,where the floor thickness,steel beam type,contact time,and human weight are considered.Finally,analytical expressions derived for the fundamental frequency and peak acceleration agree well with the experimental results and are hence proposed for practical use.

产业政策的本质目标是通过政策引导实现要素优化配置,进而推动产业技术进步和价值链升级。因而,从资源配置视角反思和挖掘不同类型产业政策失败与成功的道理具有重要理论价值和现实意义。基于此,本文基于竞争性与选择性的分类原则,从资源配置视角考察了不同类型产业政策对制造业价值链升级的差异化影响。研究表明:选择性产业政策显著抑制了制造业价值链升级,而竞争性产业政策显著推进了制造业价值链升级;资源配置机制的检验结果显示,选择性产业政策主要通过恶化资本错配而非劳动力错配进一步抑制制造业价值链升级,竞争性税收优惠政策与竞争性政府补贴政策将通过改善资本错配与劳动力错配间接推进制造业价值链升级,而竞争性低息贷款政策只通过改善资本错配间接推进制造业价值链升级,劳动力错配机制显著为负。基于以上结论,本文认为中国当前应转向实施以竞争性产业政策为主体的产业政策体系,并注重金融供给侧改革与劳动力素质的提升。

Xiaoaiping injection affects the invasionand metastasis of hepatocellular carcinomaby controlling AFP expression

Objective Xiaoaiping (XAP) is a traditional Chinese medicine that is a commonly used as an anticancerdrug in clinical practice owing to its high efficiency and low toxicity. Specifically, XAP can effectively inhibitthe growth of hepatocellular carcinoma (HCC). Alpha-fetoprotein (AFP) is a key HCC diagnostic marker andis closely related to certain malignant cytological behaviors of HCC. However, whether AFP expression andXAP treatment are related to the invasion and metastasis of HCC remains unclear. In the present study, weaimed to evaluate the effects and underlying mechanism of XAP on the invasion and metastasis of HCC..Methods Using a cell scratch assay, Transwell technology, and western blotting we detected the differentinvasion and metastatic abilities of Hep3B cells in XAP treatment and blank control groups. This allowedcomparison of the invasion and metastatic abilities of Hep3B cells with differing levels of AFP expression.AFP mRNA sequencing technology was used to analyze the mechanism of tumor invasion and metastasisassociated with AFP and XAP treatment.Results Cell invasion and metastasis abilities in the XAP group were significantly lower than those in thecontrol group (P < 0.05). Additionally, compared to the control group, the expression of AFP significantlydecreased after XAP treatment (P < 0.05). The ability of Hep3B cells to invade and metastasize waspromoted when AFP expression was up-regulated, whereas it was inhibited when AFP was silenced. XAPinjection and AFP regulate the invasion and metastatic ability of HCC by affecting matrix metalloproteinases(MMPs).Conclusion XAP injection inhibits the invasion and metastatic ability of HCC by influencing the expressionof AFP;additionally, this inhibition of AFP is achieved by affecting MMPs.

Embelin alleviates weaned piglets intestinal inflammation and barrier dysfunction via PCAF/NF-κB signaling pathway in intestinal epithelial cells

Background:Intestinal barrier plays key roles in maintaining intestinal homeostasis.Inflammation damage can severely destroy the intestinal integrity of mammals.This study was conducted to investigate the protective effects of embelin and its molecular mechanisms on intestinal inflammation in a porcine model.One hundred sixty 21-day-old castrated weaned pigs(Duroc×Landrace×Yorkshire,average initial body weight was 7.05 d to four groups and fed with a basal diet o±0.28 kg,equal numbers of castrated males and females)were allotter a basal diet containing 200,400,or 600 mg embelin/kg for 28 d.The growth performance,intestinal inflammatory cytokines,morphology of jejunum and ileum,tight junctions in the intestinal mucosa of piglets were tested.IPEC-1 cells with overexpression of P300/CBP associating factor(PCAF)were treated with embelin,the activity of PCAF and acetylation of nuclear factor-κB(NF-κB)were analyzed to determine the effect of embelin on PCAF/NF-κB pathway in vitro.Results:The results showed that embelin decreased(P<0.05)serum D-lactate and diamine oxidase(DAO)levels,and enhanced the expression of ZO-1,occludin and claudin-1 protein in jejunum and ileum.Moreover,the expression levels of critical inflammation molecules(interleukin-1β,interleukin-6,tumor necrosis factor-α,and NF-κB)were downregulated(P<0.05)by embelin in jejunal and ileal mucosa.Meanwhile,the activity of PCAF were down-regulated(P<0.05)by embelin.Importantly,transfection of PCAF siRNAs to IPEC-1 cell decreased NF-κB activities;embelin treatment downregulated(P<0.05)the acetylation and activities of NF-κB by 31.7%-74.6%in IPEC-1 cells with overexpression of PCAF.Conclusions:These results suggested that embelin ameliorates intestinal inflammation in weaned pigs,which might be mediated by suppressing the PCAF/NF-κB signaling pathway.

Numerical analysis of matrix swelling and its effect on microstructure of digital coal and its associated permeability during CO_(2)-ECBM process based on X-ray CT data

Matrix swelling effect will cause the change of microstructure of coal reservoir and its permeability,which is the key factor affecting the engineering effect of CO_(2)-ECBM technology.The Sihe and Yuwu collieries are taken as research objects.Firstly,visualization reconstruction of coal reservoir is realized.Secondly,the evolution of the pore/fracture structures under different swelling contents is discussed.Then,the influence of matrix phase with different swelling contents on permeability is discussed.Finally,the mechanism of swelling effect during the CO_(2)-ECBM process is further discussed.The results show that the intra-matrix pores and matrix-edge fractures are the focus of this study,and the contacting area between matrix and pore/fracture is the core area of matrix swelling.The number of matrix particles decreases with the increase of size,and the distribution of which is isolated with small size and interconnected with large size.The swelling effect of matrix particles with larger size has a great influence on the pore/fracture structures.The number of connected pores/fractures is limited and only interconnected in a certain direction.With the increase of matrix swelling content,the number,porosity,width,fractal dimension,surface area and volume of pores/fractures decrease,and their negative contribution to absolute permeability increases from 0.368% to 0.633% and 0.868%-1.404%,respectively.With the increase of swelling content,the number of intra-matrix pores gradually decreases and the pore radius becomes shorter during the CO_(2)-ECBM process.The matrix continuously expands to the connected fractures,and the width of connected fractures gradually shorten.Under the influence of matrix swelling,the bending degree of fluid flow increases gradually,so the resistance of fluid migration increases and the permeability gradually decreases.This study shows that the matrix swelling effect is the key factor affecting CBM recovery,and the application of this effect in CO_(2)-ECBM process can be discussed.

Epidemiological study of community- and hospital-acquired intraabdominal infections

Purpose: To investigate and analyze the clinical and etiological characteristics of community-acquired intraabdominal infections (CIAIs) and hospital-acquired or nosocomial intraabdominal infections (NIAIs) in a comprehensive hospital, to understand the characteristics, pathogen composition, and drug resistance of CIAIs as well as NIAIs, and to provide a reference for clinical treatment. Methods: We collected the clinical data of patients with intraabdominal infections admitted to our hospital from June 2013 to June 2014. In vitro drug sensitivity tests were conducted to separate pathogens, and the data were analyzed using the WHONET 5.4 software and SPSS 13.0 software. Results: A total of 221 patients were enrolled in the study, including 144 with CIAIs (55 mild-moderate and 89 severe) and 77 with NIAIs. We isolated 322 pathogenic strains, including 234 strains of gram-negative bacteria, 82 strains of gram-positive bacteria, and 6 strains of fungi. Based on clinical features, NIAIs and severe CIAIs presented significantly higher values in age, length of hospital stay, mortality, and the incidence of severe intra-abdominal infection than mild-moderate CIAIs ( p < 0.05). There was no significant difference in the prognosis between NIAIs and severe CIAIs. Primary diseases leading to CIAIs and NIAIs mostly were hepatobiliary diseases and gastrointestinal diseases respectively. Bacteria isolated from various types of IAIs mainly were Enterobacteriaceae ; mild-moderate CIAIs mostly were mono-infection of gram-negative bacteria; NIAIs mostly were mixed infections of gram-negative and gram-positive bacteria; and severe CIAIs were from either type of infection. The rate of Extended Spectrum β-Lactamase-producing Escherichia coli and Klebsiella pneumoniae was much higher in NIAIs than in CIAIs ( p < 0.05). The antimicrobial drug sensitivity of gram-negative bacteria isolated from NIAIs was significantly lower than that of CIAIs. Flow response hysteresis of throat regulation process of a two-dimensional mixed-compression supersonic inlet

The variable geometry supersonic inlet tends to decrease the throat area to reduce the Mach number upstream of the terminal shock,so as to reduce the flow loss.However,excessive Internal Contraction Ratio(ICR)exposes the inlet to a greater risk of unstart,which inevitably results in a process of increasing the throat area to aid the inlet restart.In the above throat regulation process,the inlet undergoes the start,unstart,and restart states in turn.In order to reveal the flow structure and mechanism of this process,a two-dimensional unsteady numerical simulation combined with a dynamic mesh technique were employed.The shock-on-lip Mach number of the studied inlet is 4.0 and the flight angle of attack is+6°.Analysis was focused on the state with a freestream Mach number of 3.0.The results clearly show that the flow response hysteresis appears,and restart is only realized when the throat area is obviously increased as compared to that of unstart due to the historical unstart flow structure.In addition,three typical flow fields were analyzed,and it is found that the separation ahead of the inlet was the key factor affecting the hysteresis.Finally,unstart and restart boundaries of the inlet were discussed,and the factors influencing its deviation from the typical boundaries of dual-solution area were analyzed.The newly predicted unstart and restart boundaries are much closer to the CFD results.

Fermi surface gapping in the hidden order state of PrFe_(4)P_(12);by point-contact spectroscopy

We have applied point-contact spectroscopy(PCS) to investigate the nonmagnetic hidden order(HO) state of the skutterudite compound PrFe_(4)P_(12) with its transition temperature THO~6.5 K. Its point-contact conductance curves exhibit a reproducible gap structure in the HO state below THOand its temperature dependent gap magnitude follows a BCS-like mean-field behavior.A Fano-like conductance shape is observed to emerge below the coherence temperature of PrFe_(4)P_(12). In a magnetic field, the gap feature is gradually suppressed and evolves into a pronounced Fano peak, signaling the heavy fermion state and vividly demonstrating the competition between HO and the formation of heavy fermion states. Our results strongly suggest the itinerant characteristic of f electrons in PrFe_(4)P_(12), which constrains theoretical models to explain the HO mechanism.

Comprehensive Evaluation of Electric Power Prediction Models Based on D-S Evidence Theory Combined with Multiple Accuracy Indicators

A comprehensive evaluation method of electric power prediction models using multiple accuracy indicators is proposed.To obtain the preferred models,this paper selects a number of accuracy indicators that can reflect the accuracy of single-point prediction and the correlation of predicted data,and carries out a comprehensive evaluation.First,according to Dempster-Shafer(D-S)evidence theory,a new accuracy indicator based on the relative error(RE)is proposed to solve the problem that RE is inconsistent with other indicators in the quantity of evaluation values and cannot be adopted at the same time.Next,a new dimensionless method is proposed,which combines the efficiency coefficient method with the extreme value method to unify the accuracy indicator into a dimensionless positive indicator,to avoid the conflict between pieces of evidence caused by the minimum value of zero.On this basis,the evidence fusion is used to obtain the comprehensive evaluation value of each model.Then,the principle and the process of consistency checking of the proposed method using the entropy method and the linear combination formula are described.Finally,the effectiveness and the superiority of the proposed method are validated by an illustrative instance.
