An atlas of immune cell transcriptomes in human immunodeficiency virus-infected immunological non-responders identified marker genes that control viral replication

Background:Previous studies have examined the bulk transcriptome of peripheral blood immune cells in acquired immunodeficiency syndrome patients experiencing immunological non-responsiveness.This study aimed to investigate the characteristics of specific immune cell subtypes in acquired immunodeficiency syndrome patients who exhibit immunological non-responsiveness.Methods:A single-cell transcriptome sequencing of peripheral blood mononuclear cells obtained from both immunological responders(IRs)(CD4^(+)T-cell count>500)and immunological non-responders(INRs)(CD4^(+)T-cell count<300)was conducted.The transcriptomic profiles were used to identify distinct cell subpopulations,marker genes,and differentially expressed genes aiming to uncover potential genetic factors associated with immunological non-responsiveness.Results:Among the cellular subpopulations analyzed,the ratios of monocytes,CD16^(+)monocytes,and exhausted B cells demonstrated the most substantial differences between INRs and IRs,with fold changes of 39.79,11.08,and 2.71,respectively.In contrast,the CD4^(+)T cell ratio was significantly decreased(0.39-fold change)in INRs compared with that in IRs.Similarly,the ratios of natural killer cells and terminal effector CD8^(+)T cells were also lower(0.37-fold and 0.27-fold,respectively)in the INRs group.In addition to several well-characterized immune cell-specific markers,we identified a set of 181 marker genes that were enriched in biological pathways associated with human immunodeficiency virus(HIV)replication.Notably,ISG15,IFITM3,PLSCR1,HLA-DQB1,CCL3L1,and DDX5,which have been demonstrated to influence HIV replication through their interaction with viral proteins,emerged as significant monocyte marker genes.Furthermore,the differentially expressed genes in natural killer cells were also enriched in biological pathways associated with HIV replication.Conclusions:We generated an atlas of immune cell transcriptomes in HIV-infected IRs and INRs.Host genes associated with HIV replication were identified as mark

HIV-indeterminate Western blotting(WB)results are typically obtained in WB confirmatory assays,and the number of indeterminate samples may increase with the detection of HIV infections,which will present considerable challenges for the management of HIV/AIDS.Nucleic acid detection has been used as a laboratory test for screening suspected or indeterminate samples.However,the effectiveness of these assays for the differential diagnosis of HIV-indeterminate WB samples remained undetermined.In this study,210 subjects with HIV-indeterminate WB results were detected from 6360 positive HIV screening samples between 2015 and 2016 in southeastern China,in which HIV-indeterminate WB results accounted for 3.30%.The highest proportion of indeterminate results was observed in pregnant and lying-in women receiving physical examinations(16.67%),followed by that in voluntary blood donors(8.82%).The most common WB band patterns were p24,gpl60 and p24,and gpl60.The follow-up study revealed that the highest negative and positive conversion rates of HIV antibodies were in samples with a single p24 band(80.28%),and with gpl60 and p24 bands(86.21%),respectively.Among the Env,Gag,and Pol antibodies,samples with a Gag band showed the highest negative conversion rate(81.25%),whereas the highest positive conversion rate was observed in samples with an Env band(56.76%).In addition,quantitative and qualitative HIV nucleic acid testing exhibited the highest sensitivity(96.3%)and specificity(97.85%),respectively.Our results indicate a lower proportion of HIV indeterminate WB results in southeastern China compared to previous reports,and the follow-up re-examination of patients with HIV indeterminate results should be performed.Nucleic acid testing facilitates the identification of HIV infections.

戊己丸对吲哚美辛诱导大鼠胃溃疡的保护作用研究

目的研究戊己丸对吲哚美辛诱导的大鼠胃溃疡的保护作用,并初步探索其可能的作用机制。方法 36只SD大鼠随机分为正常组、模型组、阳性对照组(奥美拉唑,20 mg/kg)、戊己丸低、中、高(2.52、5.04、10.08 g/kg生药量)剂量组,每组6只。各组大鼠灌胃相应药物或0.5%羧甲基纤维素钠(Carboxymethyl Cellulose, CMC-Na)水溶液,1次/d,连续7 d,于末次给药1 h后,除正常组外,其余各组均给予吲哚美辛(60 mg/kg)灌胃造模;6 h后处死大鼠,收集血清和胃组织。通过改良Guth法计算胃溃疡指数和溃疡抑制率,染色观察比较各组大鼠胃组织病理学变化;使用试剂盒测定胃组织中丙二醛(Malonaldehyde, MDA)、超氧化物歧化酶(Superoxide Dismutase, SOD)和环氧合酶1(Cyclooxygenase-1,COX-1),血清中一氧化氮(Nitric Oxide, NO)、肿瘤坏死因子(Tumor Necrosis Factor-α,TNF-α)、白细胞介素-6(Interleukin-6,IL-6)和白细胞介素-10(Interleukin-10,IL-10)的含量;TUNEL染色法比较各组大鼠胃组织细胞凋亡情况;免疫组化法测定胃组织中环氧合酶2(Cyclooxygenase-2,COX-2)、NF-κB(p65)蛋白的表达水平。结果与正常组相比,模型组大鼠胃组织黏膜层、黏膜下层、肌层及浆膜部分结构受损,胃溃疡指数和胃组织细胞凋亡百分比显著升高(P<0.01);MDA、TNF-α、IL-6水平显著升高(P<0.01),COX-1、NO、IL-10和SOD水平显著降低(P<0.01);COX-2和NF-κB(p65)蛋白表达显著升高(P<0.01);与模型组相比,各给药组均能改善胃组织损伤状况;戊己丸中、高剂量组和奥美拉唑组均能显著降低(P<0.01)胃溃疡指数和升高溃疡抑制率,升高COX-1、NO、IL-10水平,降低MDA、COX-2、TNF-α、IL-6和NF-κB(p65)水平,各给药组均明显降低大鼠胃组织细胞凋亡百分比,其中戊己丸中剂量组表现出良好的作用。结论戊己丸对吲哚美辛诱导的大鼠胃溃疡有明显的保护作用,其作用机制可能与改善氧化应激抑制、炎症反应和减少细胞凋亡有关。

以种曲酶活力与原油品质为指标,探究超微粉碎和普通粉碎预处理对紫菜酱油酿造的影响,并通过单因素和响应面试验,优化制曲工艺。研究结果表明,成曲蛋白酶活力值为未添加紫菜>普通粉碎>超微粉碎,且超微粉碎条斑紫菜>超微粉碎坛紫菜;发酵180 d后,超微粉碎紫菜酱油的黏度高于普通粉碎紫菜酱油,且均高于普通酱油;紫菜酱油成曲的最佳制曲工艺为接种量0.50%、制曲时间48 h、制曲温度32℃,在该条件下,成曲中蛋白酶活力可达到2109.00 U/g。以最佳制曲工艺进行验证试验,结果表明,相比于普通粉碎预处理,超微粉碎可将坛紫菜酱油和条斑紫菜酱油中蛋白质利用率分别提高5.91%和6.05%。

At the same time, China is the biggest PGMs consumption country in the world, the mineral resource of PGMs is critical shortage, it shows the importance of recycling the secondary resource of PGMs. Sino-Platinum Metals Resource (Yimen) Co., Ltd. is the leader in recycling of PGMs from secondary resource, and has made outstanding contributions to China PGMs secondary resources recycling. This article elucidates the current situation of secondary resources recovery and development of metallurgical technology for PGMs.

Study on the Recovery of Rhodium from Spent Organic Rhodium Catalysts of Acetic Acid Industry Using Pyrometallurgical Process

A new process recycling rhodium from organic waste containing rhodium in acetic acid industry is developed. Use the special affinity of base metal sulfides (FeS, Ni2S3 , CuS, etc.) on platinum group metals, adopting high nickel matte trapping-aluminothermic activation method to recovery rhodium from incinerator residue of organic rhodium waste. The method is shorter process, lower equipment requirement, and the higher activity of rhodium black. In pyrometallurgy enrichment process,the recovery rate of rhodium reached 94.65%, the full flow of rhodium recovery rate was 92.04%.

基于GaAs pHEMT工艺的宽带6位数字移相器MMIC

基于0.15μm GaAs pHEMT (pseudomorphic High Electron Mobility Transistor)工艺,研制了一款6位数字移相器微波单片集成电路(Monolithic Microwave Integrated Circuit,MMIC).该移相器由六个基本移相位级联组成,工作频带为10~18 GHz,步进值为5.625°,移相范围为0~360°,具有64个移相态.根据最优拓扑选择理论,5.625°,11.25°,22.5°移相位采用桥T型结构,降低了移相器的插损及面积;采用开关型高低通滤波器结构实现45°,90°,180°移相位,提高了大移相位的移相精度,并有效降低了寄生调幅.实测结果表明:64态移相寄生调幅均方根误差小于0.6 dB,移相输入输出回波损耗低于-11 dB,移相均方根误差小于4.2°,基态插入损耗低于8.6 dB.芯片尺寸为3.35 mm×1.40 mm.该数字移相器具有宽频带、高移相精度、尺寸小的特点,主要用于微波相控阵T/R组件、无线通信等领域.

Protective effect of L-arginine preconditioning on ischemia and reperfusion injury associated with rat small bowel transplantation

AIM: To investigate the protective effect and possible mechanism of L-arginine preconditioning on ischemia and reperfusion injury associated with small bowel transplantation (SBT).METHODS: Male inbred Wistar rats weighting between 180 and 250 g were used as donors and recipients in thestudy. Heterotopic rat SBT was performed according to the techniques of Li and wu. During the experiment, intestinal grafts were preserved in 4 ℃ Ringer's solution for 8 h before being transplanted. Animals were divided into three groups. In group 1, donors received intravenous L-arginine (50 mg/kg, 1 mL) injection 90 min before graft harvesting. However, donors in control group were given normal saline (NS) instead. In group 3, six rats were used as sham-operated control. Specimens were taken from intestinal grafts 15 min after reperfusion. Histological grading, tissue malondialdehyde (MDA) and myeloperoxidase (MPO) levels were assessed. The graft survival of each group was monitored daily until 14 d after transplantation. RESULTS: Levels of MDA and MPO in intestine of shamoperated rats were 2.0±0.22 mmol/g and 0.66±0.105 U/g. Eight hours of cold preservation followed by 15 min of reperfusion resulted in significant increases in tissue MDA and MPO levels. Pretreatment with L-arginine before graft harvesting resulted in lower enhancement of tissue levels of MDA and MPO and the differences were significant (4.71±1.02 mmol/g vs8.02±3.49 mmol/g, 1.03±0.095 U/g vs 1.53±0.068 U/g, P＜0.05). Besides, animals in L-arginine pretreated group had better histological structures and higher 2-wk graft survival rates comparing with that in NS treated group (3.3±0.52 vs6±0.1, 0/6 vs6/6, P＜0.05or 0.01).CONCLUSION: L-arginine preconditioning attenuates ischemia and reperfusion injury in the rat SBT model,which was due to antioxidant activities partially.

Studies on Determination of Ge in Au Alloys by Potassium Iodate Potentiometric Titration Method

The paper presents a new method of determining Ge in gold alloys by potassium iodate potentiometric titration rather than by traditional distillation separation. The influences of conditions such as the reduction acidity, dosage of sodium hypophosphite and reduction time on the determination of Ge were studied. Comparison was made between the influences of end-point indication for potential method and starch method on accruracy and precision of the analysed results, stability and sensitivity of end-point, selectivity of method and so on. The possibility of reaction in an electrochemical way was discussed. Ge in the alloys such as AuGe 12 , AuGeNi 12 2 , AuAgGe 18.8 12.5 and AuAgGeNi 43.8 6 0.2 was measured, respectively, with the relative standard deviation of 0.10%~0.31% and the recoveries of added standard Ge in sample of 99.40%～100.40% when the reduction acidity was 0.40~0.80 mol/L HCl and 3.3 mol/L H 3 PO 4, 15 g sodium hypophosphite and reduction time 40 min. The new method presented is high accuracy and precision in results, good stability and sensibility in end point, easy operation and strong selectivity of determination. When it is applied to analyse actual samples, satisfactory results are achieved.
