以文献分析和经验总结为主要研究方法,结合高中英语阅读教学现状,指出部分学生存在学习深度不足的问题,并提出在高中英语阅读教学中应用深度学习理念有助于让学生设计更加科学的学习目标,投入更多的精力参与学习活动,最终取得理想的学习成果。结合教学案例,指出教师可以在阅读教学中通过找出深度学习目标、补充深度学习资源、设计深度学习任务、组织学生展开自评的教学过程自然应用深度学习理念,达成英语课程育人总目标。

Neuroendocrine carcinoma(NEC)of the gallbladder(GB-NEC)is a rare but extremely malignant subtype of gallbladder cancer(GBC).The genetic and molecular signatures of GB-NEC are poorly understood;thus,molecular targeting is currently unavailable.Inthe present study,we applied whole-exome sequencing(WES)technology to detect gene mutations and predicted somatic singlenucleotide variants(SNVs)in 15 cases of GB-NEC and 22 cases of general GBC.in 15 GB-NECs,the C>T mutation was predominantamong the 6 types of SNVs.TP53 showed the highest mutation frequency(73%,11/15).Compared with neuroendocrine carcinomasof other organs,signifcantly mutated genes(SMGs)in GB-NECs were more similar to those in pulmonary large-cell euroendocrinecarcinomas(LCNECs),with drver roles for TP53 and RB1.Iin the COSMIC database of cancer-related genes,211 genes were mutated.Strikingly,RB1(4/15,27%)and NAB2(3/15,20%)mutations were found specifically in GB-NECs;in contrast,mutations in 29 genes,including ERB82 and ERBB3,were identified exclusively in GBC.Mutations in RB1 and NAB2 were significanty related to downregulation of the RB1 and NAB2 proteins,respectively,according to immunohistochemical(IHC)data(p values=0.0453 and0.0303).Clinically actionable genes indicated 23 mutated genes,including ALK,BRCA1,and BRCA2.Iin addition,potential somaticSNVs predicted by ISowN and SomVarlUS constituted 6 primary coSMIC mutation signatures(1,3,30,6,7,and 13)in GB-NEC.Genes carrying somatic SNVs were enriched mainly in oncogenic signaling pathways involving the Notch,WNT,Hippo,and RTK-RASpathways.In summary,we have systematically identified the mutation landscape of GB-NEC,and these findings may providemechanistic insights into the specifc pathogenesis of this deadly disease.

随着中国环境的日益恶化,企业经营活动与自然环境的冲突是政府、企业和学术界关注的热点话题。实现绿色制造和绿色产品生产供给是企业面临的一项重要议题。绿色管理作为一种新的管理模式,在落实新发展理念、推动绿色发展过程中起着重要作用。根据现有文献梳理了绿色管理演变过程和绿色管理的障碍因素,在此基础上提出了绿色管理对策。对于企业而言,增强企业领导的绿色承诺、实施绿色实践和绿色人力资源管理是企业实现绿色管理的重要举措。同时,只有实施绿色管理,企业才可能实现可持续性发展。

Screening and characterization of proteases produced by deep-sea cold seep bacteria

Fifty protease-producing strains were screened from sediment of deep-sea cold seep,and divided into four diff erent categories:Bacillus,Pseudoalteromonas,Vibrio,and Alteromonas according to the sequences of 16s rRNA.Their abilities to produce protease,amylase,and lipase were determined,and a Bacillus strain gcc-1 displayed very strong alkaline protease activity and stability under different thermal and acidic conditions.The purifi cation of the protease produced by strain gcc-1 was carried out by precipitation with ammonium sulfate,and sequentially chromatographed by anion exchange column and gel filtration.The purifi ed protease showed a single band at the molecular weight of 28 kDa by SDS-PAGE.The characterization results show that the purified protease exhibited a considerable activity and stability in a wide thermal range of 10-80℃and a wide acidic range of pH 6.5-11.5,and displayed highest activity at 40℃ and pH 8.5.Notably,the protease still maintained high activity even at low to 10℃.Furthermore,the protease exhibited good stability in presence of diff erent surfactants,organic solvents,oxidizing agent H_(2)O_(2),and commercial detergents.Therefore,the protease produced by gcc-1 is a cold active and high stable enzyme,and has a promising potential in laundry detergent as an additive.

The F3′5′H gene family plays an important role in regulating anthocyanin biosynthesis,abiotic stress,and hormone signaling.In this study,14 F3′5′H genes were identified from the blueberry genome.The chromosomal distribution,physicochemical properties,F3′5′H domain,conserved motifs,cis-acting elements,and intron/exon compositions were analyzed.The functional prediction analysis of these VcF3′5′Hs indicated that their biological functions included light response and other secondary metabolites.The results of qRT-PCR showed that VcF3′5′Hs(especially VcF3′5′H4)were highly expressed at the ripening stage.Subcellular localization revealed that VcF3′5′H4 may be located in the endoplasmic reticulum.Co-expression analysis showed that the VcF3′5′H gene family was related to anthocyanin.This research provides an overview of the blueberry F3′5′H family and helps verify the role of these genes in regulating anthocyanin biosynthesis.

Changes in the Economic Structure and Trends in China’s Future Energy Demands

The scientific evaluation of trends in China's future energy demands is highly important.Using provincial-level panel data from 1995 to 2015,we studied the relationships between the economic aggregate,the development of energy-intensive industries and energy demand from the perspective of changes in the proportion of energy-intensive industries in the national economy.We find that economic aggregate affects energy demand through energy-intensive industries and that changes in the economic structure are the key factor for change in energy demand.This means that China's future energy demand will be much lower than that contained in forecasts that did not consider this factor.Comprehensively promoting green-tech development and strengthening the regulation of energy-extensive industries will be one of the key options for realizing China's objective of controlling total energy consumption.
