Aim： To investigate the associations of autosomal and X-chromosome homologs of the RNA-binding-motif （RNAbinding-motif on the Y chromosome, RBMY） gene with non-obstructive azoospermia （NOA）, as genetic factors for NOA may map to chromosomes other than the Y chromosome. Methods： Genomic DNA was extracted using a salting-out procedure after treatment of peripheral blood leukocytes with proteinase K from Japanese patients with NOA （n = 67） and normal fertile volunteers （n = 105）. The DNA were analyzed for RBMX by expressed sequence tag （EST） deletion and for the like sequence on chromosome 9 （RBMXL9） by microsatellite polymorphism. Results： We examined six ESTs in and around RBMX and found a deletion of SHGC31764 in one patient with NOA and a deletion of DXS7491 in one other patient with NOA. No deletions were detected in control subjects. The association study with nine microsatellite markers near RBMXL9 revealed that D9S319 was less prevalent in patients than in control subjects, whereas D9S1853 was detected more frequently in patients than that in control subjects. Conclusion： We provide evidence that deletions in or around RBMX may be involved in NOA. In addition, analyses of markers in the vicinity of RBMXL9 on chromosome 9 suggest the possibility that variants of this gene may be associated with NOA. Although further studies are necessary, this is the first report of the association between RBMX and RBMXL9 with NOA. （Asian J Andro12006 Mar; 8： 213-218）

Biochemically curative surgery for gastrinoma in multiple endocrine neoplasia type 1 patients

AIM： To search for the optimal surgery for gastrinoma and duodenopancreatic neuroendocrine tumors in patients with multiple endocrine neoplasia type 1. METHODS： Sixteen patients with genetically confirmed multiple endocrine neoplasia type 1 （MEN 1） and Zollinger-Ellison syndrome （ZES） underwent resection of both gastrinomas and duodenopancreatic neuroendocrine tumors （NETs） between 1991 and 2009. For localization of gastrinoma, selective arterial secretagogue injection test （SASI test） with secretin or calcium solution was performed as well as somatostatin receptor scintigraphy （SRS） and other imaging methods such as computed tomography （CT） or magnetic resonance imaging （MRI）. The modus of surgery for gastrinoma has been changed over time, searching for the optimal surgery： pancreaticoduodenectomy （PD） was first performed guided by localization with the SAST test, then local resection of duodenal gastrinomas with dissection of regional lymph nodes （LR）, and recently pancreas-preserving total duodenectomy （PPTD） has been performed for multiple duodenal gastrinomas. RESULTS： Among various types of preoperative localizing methods for gastrinoma, the SASI test was the most useful method. Imaging methods such as SRS or CT made it essentially impossible to differentiate functioning gastrinoma among various kinds of NETs. However, recent imaging methods including SRS or CT were useful for detecting both distant metastases and ectopic NETs; therefore they are indispensable for staging of NETs. Biochemical cure of gastrinoma was achieved in 14 of 16 patients （87.5%）; that is, 100% in 3 patients who underwent PD, 100% in 6 patients who underwent LR （although in 2 patients （33.3%） second LR was performed for recurrence of duodenal gastri- noma）, and 71.4% in 7 patients who underwent PPTD. Experimental Confirmation on the Calibration Curves for Preston＇s Method

The Preston＇s method is considered as one of the most commonly employed methods to measure the wall shear stress. However, it is only possible to determine the wall shear stress from measured pressure differences of the Preston tube and undisturbed static pressure, combined with calibration curves, which depend on the Preston tube diameter, fluid density, and viscosity. Since its invention, no significant advancement in theory has been made, and calibration curves proposed by Preston, Patel and Bechert are still in use. In the present study, a need to measure surface shear stress over a circular cylinder prompted us to develop our original Preston tube system. The developed system has been calibrated by measuring the wall shear stress in the fully developed turbulent flow regime in a circular pipe. The present results generally confirm the previously reported calibration curves. A slight modification of the coefficients in the calibration equation shows further improvement.

Serum Cholesterol Levels at the Onset of Bloodstream Infection Have Prognostic Value

Objective: The aims of this study were to investigate the changes in serum cholesterol levels at the onset of bloodstream infection (BSI) and to determine whether serum cholesterol levels were associated with patients’ clinical backgrounds and the prognosis of BSI;Methods: A retrospective chart review was done to collect demographic information and the subjects’ medical history, invasive procedures, and medications;Patients and Methods: From April 2003 to March 2006, all patients aged ≥20 years with positive blood cultures in the University of Tokyo Hospital (a tertiary teaching hospital with 1200 beds) were enrolled;Results: Average cholesterol levels before the onset of BSI were 166.5 ± 46.5 mg/dL, and, at the onset of BSI, they decreased to 134.4 ± 45.0 mg/dL (p < 0.001). Thirty-day survivors had higher cholesterol levels both before and at the onset of BSI than non-survivors. Cholesterol levels at the onset of BSI were associated with the organisms of BSI. Patients with lower cholesterol levels tended to have higher 30-day mortality rates and longer medical treatment than patients with higher cholesterol levels (13% vs 3%, p = 0.12;24.3 days vs 18.4 days, p = 0.15);Conclusion: Cholesterol levels at the onset of BSI could be used as a prognostic marker in patients with BSI.

Immunolocalization of a Normal Wood Specific Pectin Methylesterase (CoPME) and Quantification of PME Gene Expression in Differentiating Xylem of <i>Chamaecyparis obtusa</i>

The transverse section of compression wood tracheids has a circular shape and intercellular spaces. The cause has not been determined yet;however, we hypothesized that peeling of the cell wall adhesion would cause cellular intervals, resulting in circularity of the transverse section of tracheids. Homogalacturonan, a type of pectin, functions in cell wall adhesion. Further, pectin methylesterase (PME) is involved in functionalization of homogalacturonan. We quantitated PME gene expression levels in differentiating xylem cells using different degrees of compression wood samples and examined the correlation with circularity of the transverse section of tracheids in each sample. We found that lower gene expression level of the sample corresponded with increasing circularity of the transverse section of tracheids. It is considered that the transverse section of compression wood tracheids becomes circular by suppression of PME gene expression during differentiation. Further, we observed the normal wood specific pectin methylesterase (CoPME) localization in differentiating xylem tracheids by immunolabeling. Labels localized at the entire perimeter of the compound middle lamella in normal wood, whereas sparse labeling was found in compression wood. It suggests that cell walls adhere at sites of CoPME function in differentiating xylem tracheids, but there is inadequate adhesion between cell walls where CoPME does not function. At the end of the expansion zone, the volume of the cell decreases due to a decrease in the turgor pressure of the tracheid. Further, due to moisture shrinkage of the tracheid, the adhesion begins to peel off in places of inadequate adhesion between cell walls, resulting in cell gaps and, thereby, generating a circular cell shape of cell wall formation in compression wood.

A circuit was devised for removing the influence of Rn and Tn on an ionization chamber gas monitor for β rays. The circuit subtracted the current due to Rn and Tn from the gross current. A monitor installed the circuit canceled the concentration variation of Rn and Tn caused by ventilation condition. The background level of the gas monitor compensated the influence of Rn and Tn was reduced by 10%. The minimum detectable concentrations of the compensated monitor were lower than the legal concentration limits in exhaust gas of 8 radioisotopes used in life science research and nuclear medicine: 3H, 14C, 24Na, 51Cr, 67Ga, 75Se, , 89Sr, 99mTc, and 3 main radioisotopes discharged from the nuclear power plants: 85Kr , 41Ar and 133Xe.

Zaxinone is an apocarotenoid regulatory metabolite required for normal rice growth and *** addition,zaxinone has a large application potential in agriculture,due to its growth-promoting activity and capability to alleviate infestation by the root parasitic plant Striga through decreasing strigolactone(SL)***,zaxinone is poorly accessible to the scientific community because of its laborious organic synthesis that impedes its further investigation and *** this study,we developed easy-to-synthesize and highly efficient mimics of zaxinone(MiZax).We performed a structure-activity relationship study using a series of apocarotenoids distinguished from zaxinone by different structural *** the obtained results,we designed several phenyl-based compounds synthesized with a high-yield through a simple *** tests showed that MiZax3 and MiZax5 exert zaxinone activity in rescuing root growth of a zaxinone-deficient rice mutant,promoting growth,and reducing SL content in roots and root exudates of wild-type ***,these compounds were at least as efficient as zaxinone in suppressing transcript level of SL biosynthesis genes and in alleviating Striga infestation under greenhouse conditions,and did not negatively impact *** together,MiZax are a promising tool for elucidating zaxinone biology and investigating rice development,and suitable candidates for combating Striga and increasing crop growth.

Salivary secretion impairment after preoperative irradiation and surgery in patients with oral squamous cell carcinoma

Purpose: This study was designed to determine the influence of preoperative irradiation and surgical operation on salivary secretion impairment in patients with oral squamous cell carcinoma (OSCC)。Methods: A total of 42 patients with OSCC were studied between 2004 and 2008。Salivary secretion was evaluated by Saxson's test before and 3-4 weeks after preoperative 30 Gy of irradiation, and at various intervals after surgery。Results: After preoperative irradiation, salivary secretion was 40.8% compared with pretreatment salivary secretion, then 27.8% 6 months after surgery。In several patients whose data we can get by Saxson's test more than 6 months after surgery, salivary secretion markedly increased between 12 and 24 months after surgery。Conclusions: In-patient with OSCC received preoperative irradiation and surgery, salivary secretion was about 30% 6 months after operation, then likely to recover significantly between 12 and 24 months after surgery。
