Regulation of intracellular Ca2+and calcineurin by NO/PKG in proliferation of vascular smooth muscle cells

Aim:To determine whether Ca2+/calcineurin mediated the inhibitory effects ofnitric oxide/cGMP-dependent protein kinase (NO/PKG) on the proliferation ofvascular smooth muscle cells (VsMC).Methods:Proliferation and viability ofprimary VsMC from rat aorta were measured using [3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl tetrazolium bromide] (MTT) assay and acridine orange and ethidiumbromide staining,*** Ca2+was determined by Fluo-3/*** protein and its activity were assayed using immunoblotting and freeinorganic phosphate analysis,***:(e)-s-nitroso-N-acetyl-penicillamine (sNAP) and sp-8-(4-chlorophenylthio)-guanosine-3′,5′-cyclicmonophosphorothioate (sp-8-pCPT-cGMPs) decreased phenylephrine (PE)-in-duced proliferation of VsMC by 27.3% and 36.6%,respectively,but Rp-8-[(4-chlorophenyl)thio]-guanosine-3′,5′-cyclic monophosphorothioate (Rp-8-pCPT-cGMPs) increased PE-induced proliferation of ***,sp-8-pCPT-cGMPs,and Rp-8-pCPT-cGMPs did not affect the viability of *** proteinwas decreased by 63.1% and its activity was decreased by 59.7% in smooth musclecells (sMC) pretreated with verapamil (Ver) and then stimulated by *** sMCpretreated with Vet,the absorbance of cells stimulated by PE decreased by 22.0%and was further inhibited by the additional treatment of sNAP and *** sMC pretreated with cyclosporin A (CsA),the absorbance of cellsstimulated by PE decreased by 36.7%,but could not be further altered by theadditional treatment of sNAP,sp-8-pCPT-cGMPs,and ***,Ver inhibited PE-induced intracellular Ca2+variations,which could befurther inhibited by sNAP and sp-8-pCPT-cGMPs,but not by ***,the increase in calcineurin activity induced by PE was inhibited bysNAP and sp-8-pCPT-cGMPs,but was promoted by ***:NO/PKG regulates calcineurin activity via the modulation of intracel-lular Ca2+concentration,and thus partially inhibits the prolifera

Aim:To investigate whether the circadian expression of central clock genes andangiotensin Ⅱ type 1 (AT1) receptors was altered in sinoaortic-denervated (sAD)***:Male sprague-Dawley rats underwent sinoaortic denervation or asham operation at the age of 12 *** weeks after the operation,bloodpressure and heart period were measured in the conscious state in a group ofsham-operated (n=10) and sAD rats (n=9).Rest sAD and sham-operated ratswere divided into 6 groups (n=6 in each group).The suprachiasmatic nuclei (sCN)tissues were taken every 4 h throughout the day from each group for the determi-nation of the mRNA expression of clock genes (Per2 and Bmal1) and the AT1receptor by RT-PCR;the protein expression of Per2 and Bmal1 was determined byWestern ***:Blood pressure levels in the sAD rats were similar tothose of the sham-operated ***,blood pressure variabilities signifi-cantly increased in the sAD rats compared with the sham-operated *** variation of clock genes in the sCN of the sham-operated rats was char-acterized by a marked increase in the mRNA and protein expression during ***2 and Bmal1 mRNA levels were significantly lower in the sAD rats,especially during dark *** blot analysis confirmed an attenuation ofthe circadian rhythm of the 2 clock proteins in the sCN of the sAD ***1receptor mRNA expressions in the sCN were abnormally upregulated in the lightphase,changed to a 12-h cycle in the sAD ***:The circadian varia-tion of the 2 central clock genes was attenuated in the sAD *** baroreflexdysfunction also induced a disturbance in the expression of AT1 receptors in thesCN.

Background This study aimed to identify survival risk factors in Chinese children with hepatoblastoma(HB)and assess the effectiveness of the new treatment protocol proposed by the Chinese Children's Cancer Group(CCCG)in *** A multicenter,prospective study that included 399 patients with HB from January 2015 to june 2020 was *** demographics,treatment protocols,and other related information were *** regression models and Kaplan-Meier curve methods were *** The 4-year event-free survival(EFs)and overall survival(Os)were 76.9 and 93.5%,*** 4-year EFs rates for the very-low-risk,low-risk,intermediate-risk,and high-risk groups were 100%,91.6%,81.7%,and 51.0%,*** 4-year 0s was 100%,97.3%,94.4%,and 86.8%,*** regression analysis found that age,tumor rupture(R+),and extrahepatic tumor extension(E+)were independent prognostic factors.A total of 299 patients had complete remission,and 19 *** with declining alpha-fetoprotein(AFP)>75%after the first two cycles of neoadjuvant chemotherapy had a better EFs and Os than those≤75%.Conclusions The survival outcome of HB children has dramatically improved since the implementation of CCCG-HB-2016 ***≥8 years,R+,and E+were independent risk factors for *** with a declining AFP>75%after the first two cycles of neoadjuvant chemotherapy had better EFs and Os.

Accurate diagnosis of severe coronary stenosis based on resting magnetocardiography: a prospective, single-center, cross-sectional analysis

OBJECTIVE To evalu ate the role of resting magnetocardiography in identifying seve re coronary artery stenosis in patients with suspected coronary artery *** A total of 513 patients with angina symptoms were included and divided into two groups based on the extent of coronary artery disease determined by angiography:the non-severe coronary stenusis group(stenosis) and the severe coronary stenosis group(≥70% stenosis).The diagnostic model was constructed using magnetic field map(MFM) parameters,either individually or in combination with clinical *** performance of the models was evaluated using receiver operating characteristic curves,accuracy,sensitivity,specificity,positive predictive value(PPV) and ne gative predictive value(NPV).Calibration plots and decision curve analysis were performed to investigate the clinical utility and performance of the models,*** In the severe coronary stenosis group,QR_MCTDd,s_MDp,and TT_(MA)C_(50) were significantly higher than those in the non-severe coronary stenosis group(10,46±10.66 vs,5.11±6.07,P 1;7.2±8.64 vs.4.68±6.95,P=0.003;0.32±57.29 vs.0.26±57.29,P 1).While,QR_MV_(amp),R_(MA),and T_(MA) in the severe coronary stenosis group were lower(0.23±0.16 vs.0.28±0.16,P1;55.06±48.68 vs.59.24±53.01,P1;51.67±39.32 vs. 60.45±51.33,P 1).seven MFM parameters were integrated into the model,resulting in an area under the curve of 0.810(95% CI:0.765-0.855).The sensitivity,specificity,PPV,NPV,and accurecy were 71.7%,80.4%,93.3%,42.8 %,and 73.5%;*** combined model exhibited an area under the curve of 0.845(95% CI:0.798-0.892).The sensitivity,specificity,PPV,NPV,and accuracy were 84.3%,73.8%,92.6%,54.6%,and 82.1%;*** curves demonstrate d excellent agreement between the nomogram prediction and actual *** decision curve analysis showed that the c ombine d model provided greater net benefit compared to the magnetocardingraphy model.C

Morphological and functional changes in the blood-spinal cord barrier of rabbits in an experimental spinal cord presyrinx state

BACKGROUND: Presyrinx state of spinal cord can reflect the initial lesion of syringomyelia (sM). The early trials has proved that ischamia and edema are main pathological changes of presyrinx state. OBJECTIVE: To establish sM model of rabbits for investigating the relationship between changes of morphous and function of blood-spinal cord barrier and the edema degree, histological changes in presyrinx state of sM, and to explore the mechanism of the presyrinx state of sM. DEsIGN: Randomized controlled animal experiment. sETTING: department of Neurosurgery, Fourth hospital, Heibei Medical university. MATERIALs: sixty Chinese healthy white rabbits, aged 3.5-4.5 months, weighing 1.5-2.0 kg, were provided by Experimental Animal Center of Hebei Medical university [certification: (sYXK(Ji)2003-0026)]. Evan's blue (EB) and dimethylformamide (DMF) were purchased from Jingmei Biotech Co., Ltd. RM2125 paraffin section cutter (Leica Company, Japan), H-7500 transmission electron microscope (Hitachi Company, Japan), PM-20 light microscope photograph system (Olympus Company, Japan). METHODs: The experiment was carried out in the Laboratory of Neurosurgery department, second hospital of Hebei Medical university from January to june 2006. ① All the rabbits were randomly divided into two groups: model group (n =40), control group (n =20). Rabbits in two groups were divided into five subgroups once again at five time points (1st, 3rd, 7th, 14th, 21st days, n =8 and n =4 at each time point in the model group and control group, respectively). Under ketamine anesthesia, 0.6 mL Kaolin solution (250 g/L, 37 ℃) was injected into the cisterna magna of rabbits in model group, while 0.6 mL physiological saline (37 ℃) was injected into the rabbits of control group. ② On the 1st, 3rd, 7th, 14th, 21st days after kaolin injection, cervical cord samples were harvested after sacrifice of animal. Quantitative analysis on the function of blood-spinal cord barrier was performed by Evan's blue technique
