A lateral flow immunoassay(LFA)biosensor that allows the sensitive and accurate identification of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and other common respiratory viruses remains highly desired in the face of the coronavirus disease 2019 ***,we propose a multiplex LFA method for the on-site,rapid,and highly sensitive screening of multiple respiratory viruses,using a multilayered film-likeﬂuorescent tag as the performance enhancement and signal amplification *** film-like three-dimensional(3D)tag was prepared through the layer-by-layer assembly of highly photostable CdSe@ZnS-COOH quantum dots(QDs)onto the surfaces of monolayer graphene oxide nanosheets,which can provide larger reaction interfaces and specific active surface areas,higher QD loads,and better luminescence and dispersibility than traditional spherical fluorescent microspheres for LFA *** constructedﬂuorescent LFA biosensor can simultaneously and sensitively quantify SARS-CoV-2,influenza A virus,and human adenovirus with low detection limits(8 pg/mL,488 copies/mL,and 471 copies/mL),short assay time(15 min),good reproducibility,and high ***,our proposed assay has great potential for the early diagnosis of respiratory virus infections given its robustness when validated in real saliva samples.

旨在研究全价粮添加红景天苷对犬血液和肝转录组学的影响,以期为犬的肝养护及肝疾病预防工作提供试验数据支持。选取10只成年中华田园犬(平均体重8.67±1.00 kg),随机分为对照组(n=5)和试验组(n=5),分别饲喂对照犬粮和红景天苷全价粮。通过血液检查来评估红景天苷全价粮的饲喂效果和安全性,并通过转录组学技术探究红景天苷全价粮对犬潜在的调节机制,试验期为8周。血液检查结果显示,红景天苷全价粮显著降低了犬血清谷丙转氨酶的浓度(P0.05),但相比于对照犬粮,采食红景天苷全价粮的犬血液指标波动小,能保持血液指标的稳定。转录组学分析结果显示,差异表达基因显著富集在钙信号通路、PI3K-Akt信号通路和MAPK信号通路等。针对表型的基因集富集分析结果表明,红景天苷全价粮在脂代谢、糖代谢、炎症、氧化应激和纤维化方面起调节作用,其中对于糖脂代谢的调节更显著,相关信号通路在试验组显著上调。综上所述,红景天苷全价粮具有降低犬血清谷丙转氨酶浓度的作用,能够稳定犬的血液指标浓度,调节了犬的肝糖脂代谢,这可能主要通过调节钙信号通路、PI3K-Akt信号通路和MAPK信号通路来实现。

To screen specific antitumor drugs targeting telomerase catalytic subunit (hEST2), 12 different hEST2 antisense oligonucleotides were designed based on hEST2 mRNA second structure and transfected into tumor cell lines by the lipofectin-mediated method. Cell growth activity was evaluated by MTT assay. HEST212 was picked out and its specificity, antitumor tree and continuous effect were analyzed. The results showed that hEST212 had promising anti-tumor activity in vitro, hEST2 can be used as a pratical target and an antisense drug candidate for cancer.

Dear Editor,During the COVID-19 pandemic,several SARS-CoV-2 variants such as Alpha,Delta,and Omicron successively became dominant *** infection of SARS-CoV-2 is triggered by the binding of spike protein to the cell-surface receptor angiotensinconverting enzyme 2(ACE2),which then fuses with cell membrane or lysosomal membrane after endocytosis.1 Extensive cell fusion and syncytia formation are a signature feature of severe SARS-CoV-2 and may play an important role in COVID-19 *** has shown that SARS-CoV-2 spikemediated cell fusion leads to the formation of abnormal and multinucleated cells in the lungs of patients.2 Accordingly,the fusion ability of SARS-CoV-2 spike protein may be a leading indicator of viral infectivity and disease severity in SARS-CoV-2 patients.

Prostate cancer(PCa)is the second most common malignancy among men *** Fu-Zheng-Yi-Liu(FZYL)Formula has been widely utilized in the treatment of *** study investigates whether the FZYL Formula can inhibit PCa by tar-geting the TAMs/CCL5 *** conducted in vitro co-cultures and in vivo co-injections of PCa cells and TAMs to mimic their *** showed that the FZYL Formula significantly reduced the proliferation,colony formation,subpopulations of PCSCs,and sphere-formation efficacy of PCa cells,even in the presence of TAM ***,the Formula markedly decreased the migration,invasion,and epithelial-mesenchymal transition(EMT)of PCa cells induced by *** FZYL Formula also reversed M2 phenotype polarization in TAMs and dose-dependently reduced their CCL5 expression and secretion,with minimal cytotoxicity *** studies confirmed that the TAMs/CCL5 axis is a critical target of the FZYL Formula,as the addition of exogen-ous CCL5 partially reversed the formula’s inhibitory effects on PCSCs self-renewal in the co-culture ***,the Formula also significantly inhibited the growth of PCa xenografts,bone metastasis,and PCSCs activity in vivo by targeting the TAMs/CCL5 ***,this study not only elucidates the immunomodulatory mechanism of the FZYL Formula in PCa therapy but also highlights the TAMs/CCL5 axis as a promising therapeutic target.
