Golgi-localized MORN1 promotes lipid droplet abundance and enhances tolerance to multiple stresses in Arabidopsis

Lipid droplet(LD)in vegetative tissues has recently been implicated in environmental responses in plants,but its regulation and its function in stress tolerance are not well ***,we identified a Membrane Occupation and Recognition Nexus 1(MORN1)gene as a contributor to natural variations of stress tolerance through genome-wide association study in Arabidopsis *** of its loss-of-function mutant and natural variants revealed that the MORN1 gene is a positive regulator of plant growth,disease resistance,cold tolerance,and heat *** MORN1 protein is associated with the Golgi and is also partly associated with *** truncations that disrupt these associations abolished the biological function of the MORN1 ***,the MORN1 gene is a positive regulator of LD abundance,and its role in LD number regulation and stress tolerance is highly ***,this study identifies MORN1 as a positive regulator of LD abundance and a contributor to natural variations of stress *** implicates a potential involvement of Golgi in LD biogenesis and strongly suggests a contribution of LD to diverse processes of plant growth and stress responses.

Stigma exsertion is an important trait for outcrossing ability in *** exsertion rate(SER)of male sterile lines(MSLs)is a key factor affecting F1-seed production in hybrid *** this study,seven QTLs for SER were detected on five chromosomes using a set of single-segment substitution lines(SSSLs)derived from *** of the QTLs were mapped in the estimated intervals of 92.5–333.0 ***-5 was located in a substitution segment of 92.5 *** ER-1 b and qS ER-8 b were respectively limited to 333.0 kb and 107.5 kb by secondary substitution ***-1 b and qSER-3 had bigger additive effects of 11.5%and 11.9%,respectively,while the other five QTLs had smaller additive effects from 5.7%to 8.6%.Open reading frames were identified in the regions of qS ER-5 and qSER-8 b in *** and *** *** mapping of the QTLs laid a foundation for the cloning of genes,and QTLs for SER will be used to develop MSLs with strong ability of outcrossing.

Rice(Oryza sativa L.)is grown nearly worldwide and provides the staple food for more than half of the global population(Luo et al,2017).The genomes of several cultivated rice varieties including Nipponbare(NPB)(Kawahara et al,2013;Sakai et al,2013),IR64(Tanaka et al,2020),93-11(Zhang et al,2018)and R498(Du et al,2017)at chromosome level,and Minghui 63 and Zhenshan 97(Zhang et al,2016)at scaffold level have been assembled,annotated and released,among which the R498 and NPB genomes are widely used as reference genomes in rice ***,there are thousands of rice cultivars,landraces and wild rice varieties in the world with dramatically different genetic backgrounds,and the genomes of native rice varieties in South China,which is one of the major rice production areas in China,have not been de novo *** 74(HJX74)is an indica rice variety bred in South China Agricultural University,Guangdong Province with widely environmental adaptability and high yield(***/variety/varis/***).HJX74 exhibits significant phenotypic and genetic differences from those varieties whose whole genomes have been properly sequenced and assembled(Fig.1).

Grain size is an important determinant of grain weight and yield in *** several genes related to grain size have been identified,natural variations in these genes that affect grain size are poorly ***,we describe the grain length QTL GL10,encoding MADS56,which positively regulates grain length and grain weight.A natural allelic variation of NIL-gl10,containing an~1.0-kb deletion in the first exon that abolishes its transcription,results in shorter grain length,lower grain weight and delayed flowering in gl10 *** knockout of GL10 in the HJX74 background leads to grain phenotypes similar to that of NIL-gl10,while overexpression of GL10 results in increased grain length and weight and earlier heading ***10 regulates grain length by promoting greater longitudinal cell growth in the grain ***,GL10 participates in the regulation of gibberellic acid(GA)signaling pathway genes in young panicle *** of GL10 haplotypes shows obvious divergence between the japonica and indica *** findings reveal an allelic variation of GL10 that may explain differences in grain length among modern cultivars and could be used to breed rice varieties with optimized grain shape.

Flowers are the core reproductive organ of plants, and flowering is essential for cross-pollination. Diurnal flower-opening time is thus a key trait influencing reproductive isolation, hybrid breeding, and thermostability in plants. However, the molecular mechanisms controlling this trait remain unknown. Here, we report that rice Diurnal Flower Opening Time 1 (DFOT1) modulates pectin methylesterase (PME) activity to regulate pectin methylesterification levels of the lodicule cell walls, which affect lodicule swelling to control diurnal flower-opening time. DFOT1 is specifically expressed in the lodicules, and its expression gradually increases with the approach to flowering but decreases with flowering. Importantly, a knockout of DFOT1 showed earlier diurnal flower opening. We demonstrate that DFOT1 interacts directly with multiple PMEs to promote their activity. Knockout of PME40 also resulted in early diurnal flower opening, whereas overexpression of PME42 delayed diurnal flower opening. Lower PME activity was observed to be associated with higher levels of pectin methylesterification and the softening of cell walls in lodicules, which contribute to the absorption of water by lodicules and cause them to swell, thus promoting early diurnal flower opening. Higher PME activity had the opposite effect. Collectively, our work uncovers a molecular mechanism underlying the regulation of diurnal flower-opening time in rice, which would help reduce the costs of hybrid breeding and improve the heat tolerance of flowering plants by avoiding higher temperatures at anthesis.

O. meridionalis indigenous to Australia is an AA genome wild species in Oryza,it has been considered an important germplasm resource for improvement of cultivated rice. In this study,a total of 99 chromosomal single-segment substitution lines(SSSLs) were developed by using O. meridionalis as a donor and an elite indica cultivar,Huajingxian 74 as recipient,through successive backcrossing and SSR marker-based genotyping. The 99 substituted segments were distributed on 12 chromosomes with the estimated total lengths of 1580.16 cM and 15.11 c M mean length of every segment. The 837.94 c M coverage length of substituted segments covered 54.98 percent of rice genome. Fourteen quantitative traits were evaluated in SSSLs for two seasons,including plant vegetative growth traits,i.e.,heading date,plant height,flag leaf length,flag leaf width;Grain yield related traits,i.e.,panicle numbers per plant,panicle length,spikelet numbers per panicle,grain density,number of primary branches per panicle,seed-setting rate,100-grain weight,grain length,grain width,and ratio of grain length to width. A total of 161 putative QTLs detected for thirteen traits except panicle numbers per plant. Among them,32 QTLs related to nine traits were identified both in the late season of 2014 and the early season of 2015,in which 10 QTLs had the positive additive effects. The additive effect of the QTLs ranged from 0.10 to 33.55,and the contribution of the additive effect to traits were from 0.06-20.92 percent. The present study demonstrated that the SSSLs offer both opportunities and a good germplasm platform for identification and transformation beneficial genes of O. meridionalis,as well as a precious long-time in-vivo conservation reservoir for O. meridionalis.
