Unlike chemosynthetic drugs designed for specific molecular and disease targets,active small-molecule natural products typically have a wide range of bioactivities and multiple targets,necessitating extensive screening and *** address this issue,we propose a strategy for the direct in situ microdynamic examination of potential drug candidates to rapidly identify their effects and mechanisms of *** a proof-of-concept,we investigated the behavior of mussel oligosaccharide(MOS-1)by tracking the subcellular dynamics of fluorescently labeled MOS-1 in cultured *** recorded the entire dynamic process of the localization of fluorescein isothiocyanate(FITC)-MOS-1 to the lysosomes and visualized the distribution of the drug within the ***,lysosomes containing FITC-MOS-1 actively recruited lipid droplets,leading to fusion events and increased cellular lipid *** drug behaviors confirmed MOS-1 is a candidate for the treatment of lipid-related ***,in a high-fat HepG2 cell model and in high-fat diet-fed apolipoprotein E(ApoE)^(-/-)mice,MOS-1 significantly promoted triglyceride degradation,reduced lipid droplet accumulation,lowered serum triglyceride levels,and mitigated liver damage and ***,our work supports the prioritization of in situ visual monitoring of drug location and distribution in subcellular compartments during the drug development phase,as this methodology contributes to the rapid identification of drug ***,this methodology is significant for the screening and development of selective small-molecule drugs,and is expected to expedite the identification of candidate molecules with medicinal effects.

Production of Functional Materials Derived from Regenerated Silk Fibroin by Utilizing 3D Printing and Biomimetic Enzyme-induced Mineralization

Critical-sized bone defects, commonly encountered in clinical orthopedic surgery, present a significant challenge. One of the promising solutions is to prepare synthetic bone substitute materials with precise structural control, mechanical compatibility, and enhanced osteogenic induction performance, nevertheless the successful preparation of such materials remains difficult. In this study, a two-step technique,integrating an extrusion-based printing process with biomimetic mineralization induced by alkaline phosphatase(ALP), was developed. Initially,a pre-cured hydrogel of regenerated silk fibroin(RSF) with a small quantity of hydroxypropyl cellulose(HPC) and ALP was prepared through heating the mixed aqueous solution. This pre-cured hydrogel demonstrated thixotropic property and could be directly extruded into predetermined structures through a 3D-printer. Subsequently, the 3D-printed RSF-based materials with ALP underwent biomimetic in situ mineralization in calcium glycerophosphate(Ca-GP) mineralizing solution, utilizing the polymer chains of RSF as templates and ALP as a trigger for cleaving phosphate bonds of Ca-GP. The resulting 3D-printed RSF-mineral composites including hydrogel and sponge possessed adjustable compression modulus of megapascal grade and variable hydroxyapatite content, which could be controlled by manipulating the duration of the mineralization ***, these 3D-printed RSF-mineral composites demonstrated non-cytotoxicity towards rat bone marrow mesenchymal stem cells. Therefore, they may hold great potential for applications involving the replacement of tissues characterized by osteoinductivity and intricate structures.

Zinc-promoted umpolung thiolation of alkyl electrophiles with masked sulfur transfer reagents in the absence of nickel or copper catalysis is described. This protocol proceeds via a SET process of Zn to electrophilic sulfur reagent followed by insertion of Zn into disulfide and nucleophilic thiolation, providing straightforward access to a wide range of alkyl sulfides with broad substrate scope. A neutral TMEDA-ligated four-coordinated zinc thiolate with tetrahedra geometry was synthesized, isolated and fully characterized by NMR, IR and X-ray analysis. More importantly, the chemical reactivity of this active intermediate has been investigated, enabling the construction of C-Se, C-Te, Sb-S and Bi-S bonds to prepare valuable sulfur-containing molecules and beyond.

CPT: a pre-trained unbalanced transformer for both Chinese language understanding and generation

In this paper, we take the advantage of previous pre-trained models(PTMs) and propose a novel Chinese pre-trained unbalanced transformer(CPT). Different from previous Chinese PTMs, CPT is designed to utilize the shared knowledge between natural language understanding(NLU) and natural language generation(NLG) to boost the performance. CPT consists of three parts: a shared encoder, an understanding decoder, and a generation decoder. Two specific decoders with a shared encoder are pretrained with masked language modeling(MLM) and denoising auto-encoding(DAE) tasks, *** the partially shared architecture and multi-task pre-training, CPT can(1) learn specific knowledge of both NLU or NLG tasks with two decoders and(2) be fine-tuned flexibly that fully exploits the potential of the model. Moreover, the unbalanced transformer saves the computational and storage cost, which makes CPT competitive and greatly accelerates the inference of text generation. Experimental results on a wide range of Chinese NLU and NLG tasks show the effectiveness of CPT.

Superhydrophobic surface(SHS) has been well developed, as SHS renders the property of minimizing the water/solid contact interface. Water droplets deposited onto SHS with contact angles exceeding 150°, allow them to retain spherical shapes, and the low adhesion of SHS facilitates easy droplet collection when tilting the substrate. These characteristics make SHS suitable for a wide range of applications. One particularly promising application is the fabrication of microsphere and supraparticle materials. SHS offers a distinct advantage as a universal platform capable of providing customized services for a variety of microspheres and supraparticles. In this review, an overview of the strategies for fabricating microspheres and supraparticles with the aid of SHS, including cross-linking process, polymer melting,and droplet template evaporation methods, is first presented. Then, the applications of microspheres and supraparticles formed onto SHS are discussed in detail, for example, fabricating photonic devices with controllable structures and tunable structural colors, acting as catalysts with emerging or synergetic properties, being integrated into the biomedical field to construct the devices with different medicinal purposes, being utilized for inducing protein crystallization and detecting trace amounts of analytes. Finally,the perspective on future developments involved with this research field is given, along with some obstacles and opportunities.

The influences of canopy temperature measuring on the derived crop water stress index

Crop water stress index(CWSI)is widely used for efficient irrigation *** canopy temperature(T_(c))measurement is necessary to derive a reliable *** objective of this research was to investigate the influences of atmospheric conditions,settled height,view angle of infrared thermography,and investigating time of temperature measuring on the performance of the *** irrigation treatments were used to create different soil water conditions during the 2020-2021 and 2021-2022 winter wheat-growing *** CWSI was calculated using the CWSI-E(an empirical approach)and CWSI-T(a theoretical approach)based on the T_(c).Weather conditions were recorded continuously throughout the experimental *** results showed that atmospheric conditions influenced the estimation of the CWSI;when the vapor pressure deficit(VPD)was>2000 Pa,the estimated CWSI was related to soil water *** height of the installed infrared thermograph influenced the T_(c)values,and the differences among the T_(c)values measured at height of 3,5,and 10 m was smaller in the afternoon than in the ***,the lens of the thermometer facing south recorded a higher T_(c)than those facing east or north,especially at a low height,indicating that the direction of the thermometer had a significant influence on T_(c).There was a large variation in CWSI derived at different times of the day,and the midday measurements(12:00-15:00)were the most reliable for estimating *** linear relationships were found between the transpiration rate and CWSI-E(R^(2)of 0.3646-0.5725)and CWSI-T(R^(2)of 0.5407-0.7213).The relations between fraction of available soil water(FASW)with CWSI-T was higher than that with CWSI-E,indicating CWSI-T was more accurate for predicting crop water *** addition,The R^(2)between CWSI-T and FASW at 14:00 was higher than that at other times,indicating that 14:00 was the optimal time for using the CWSI for crop water status *** hi
