Association of Lysosome Associated Protein Transmembrane 4 Beta Gene Polymorphism with the Risk of Pancreatic Cancer

Objective： Lysosome associated protein transmembrane 4 beta （LAPTM4B） was originally identified as a gene in human hepatocellular carcinoma （HCC）. It was successfully cloned by fluorescence differential display, rapid amplification of cDNA ends （RACE） and reverse transcription polymerase chain reaction （RT-PCR）. Previous study showed that the novel gene played an important role in the occurrence, development, migration and prognosis of tumors. Pancreatic cancer is an aggressive malignancy with the majority of patients dying within one year after diagnosis. This study tries to find out the relationship between lysosome associated protein transmembrane 4 beta gene polymorphism and the susceptibility of pancreatic cancer. Methods： A case-control study was conducted in China, including 58 pancreatic cancer cases and 156 healthy controls. Human genomic DNA was used as the template, polymerase chain reaction （PCR） was used to detect the distribution of LAPTM4B genotype. Analyses Odds ratio （OR） and corresponding 95% confidence interval （95%CI） with logistic regression were performed. Results： Two alleles of LAPTM4B generated three kinds of genotypes in population, ＊1/1, ＊1/2, and ＊2/2. The genotype frequency of ＊1/1, ＊1/2 and ＊2/2 in the pancreatic cancer group were 41.4%, 44.8% and 13.8% respectively, which were not significantly different from those of healthy group （47.4%, 42.9%, 9.6%）（P=0.773, P=0.291）. Also the ＊2 allele frequency of LAPTM4B among pancreatic cancer had no significantly difference with the controls （P=0.354）. When compared to the ＊1 allele, the people with ＊2 allele had no increased risk of pancreatic cancer. Conclusion： The gene polymorphism of LAPTM4B may not influence the susceptibility of pancreatic cancer.

This study involved the investigation of the effects of the continuous cooling process conditions on the crystallization and liberation characteristics of anosovite in Ti-bearing titanomagnetite smelting slag. The samples were heated until melting and then the temperature was held at 1650℃ for nearly 0.5 h;subsequently, the samples were cooled at different cooling rates to different temperatures and water-quenched after being held for different times at these temperatures. Last, the obtained crystallized samples were used to analyze the crystallization and liberation characteristics. It was found that, during the continuous cooling process, anosovite particles were found to initially precipitate in the slag at a relatively high crystallization temperature, showing the characteristics of euhedral crystal. The precipitation and growth of anosovite grain is strong and the morphology of anosovite was basically not affected by the continuous cooling conditions. From the morphology perspective, the formed anosovite is an excellent Ti-rich phase to be selective separated. The formation of spinel and diopside is negative for the liberation and selective separation of the anosovite phase. The crystallization diagrams of TiO2-MgO-CaO-SiO2-Al2O3-FeO slag undergoing different continuous cooling processes were constructed to help to determine the optimal continuous cooling–quenching condition for selective separation of anosovite. Moreover, the addition of B2O3 can enlarge the range of the optimal continuous cooling–quenching conditions for selective separation of anosovite.

目的基于美国食品药品管理局不良事件报告系统(FAERS)数据库,对抗体药物偶联物(ADC)相关血液系统不良事件进行数据挖掘,为临床安全使用ADC药物提供参考。方法下载FAERS数据库2011年第3季度至2022年第4季度数据,经过去重、名称标准化等数据清洗,提取ADC相关血液系统不良事件,采用报告比值比法和信息成分法进行信号检测。结果共提取到以8种ADC药物为首要怀疑药品的不良事件报告101 610份,ADC相关血液系统不良事件报告5 768份,其中粒细胞缺乏症涉及3 423例,信号强度从强到弱的药品依次为戈沙妥珠单抗(SG)、奥星-吉妥珠单抗(GO)、维布妥昔单抗(BV)、维汀-珀拉妥珠单抗(PV)、维汀-恩弗妥单抗(EV)、德曲妥珠单抗(TD)、奥星-艾诺妥珠单抗(IO)和恩美曲妥珠单抗(TDM-1);造血细胞缺少症涉及2 327例,信号强度从强到弱的药品依次为IO、SG、BV、EV、PV、TD、TDM-1和GO。ADC药物相关血液系统不良事件临床结局为死亡的病例包括BV179例(16.84%),TDM-1 102例(13.01%),TD 88例(27.08%),GO 12例(16.90%),IO 8例(11.59%),EV 54例(24.32%),PV 22例(27.16%),SG 84例(21.05%)。不良事件发生时间分析显示,TD、IO和SG用药首日事件发生数占总例数的40%以上;TD、GO、IO、EV、PV和SG发生血液系统不良事件中位时间在1个给药疗程(21 d)内。结论应重视ADC药物相关血液系统不良事件风险,临床用药过程中应密切监测血细胞计数变化,出现异常应及时诊治。

With the increasing demand for energy conservation and emission reduction,more attentions have been paid to the intelligentization,greenization and low carbonization during the transformation and upgrading of steelmaking plants.Ladle furnace(LF)refining is one of the key procedures in steelmaking process and has been widely used in steelmaking plants for its high equipment matching degree,low equipment investment and outstanding refining performance.According to the main tasks of LF refining process,the modeling methods of temperature prediction model,slag-making model,alloying model,argon blowing model and model of inclusions behavior were systematically reviewed,and the advantages and disadvantages of each modeling method were summarized.In addition,the technical framework for the future has also been proposed based on existing works,including classification of raw materials,graphic representation of knowledge,introduction,upgradation and management of device/equipment,customization of steelmaking,modeling of refining process,synergy of models,intelligentization of decision-making,automation of control,and digitization of processes and operations,aiming to provide a reference for the modeling and intelligent development of LF refining process.

目的探究后巩膜葡萄肿对高度近视白内障患者术后人工晶状体稳定性的影响。方法57例(63眼)行白内障超声乳化联合人工晶状体植入术的白内障伴高度近视患者根据是否存在后巩膜葡萄肿分研究组(26例共28眼,合并后巩膜葡萄肿)和对照组(31例共35眼,单纯白内障伴高度近视)。对比两组术前眼轴,术后3个月眼轴、中轴区晶状体后囊膜至视网膜距离、前房深度、角膜曲率、最佳矫正视力(BCAV)、平均绝对屈光误差(MAE)、人工晶状体囊袋偏心量及倾斜角。结果研究组术前及术后3个月眼轴长度、术后3个月BCAV、裸眼视力数值(logMAR)、MAE、水平及垂直方向的倾斜角、前房深度均高于对照组(P<0.01或0.05)。结论高度近视合并巩膜葡萄肿患者行白内障手术时应关注人工晶状体的稳定性。

BACKGROUND Patients with type 2 diabetes mellitus(T2DM)have large fluctuations in blood glucose(BG),abnormal metabolic function and low immunity to varying degrees,which increases the risk of malignant tumor diseases and affects the efficacy of tumor chemotherapy.Controlling hyperglycemia may have important therapeutic implications for cancer patients.AIM To clarify the influence of BG fluctuations on chemotherapy efficacy and safety in T2DM patients complicated with lung carcinoma(LC).METHODS The clinical data of 60 T2DM+LC patients who presented to the First Affiliated Hospital of Ningbo University between January 2019 and January 2021 were retrospectively analyzed.All patients underwent chemotherapy and were grouped as a control group(CG;normal BG fluctuation with a mean fluctuation<3.9 mmol/L)and an observation group(OG;high BG fluctuation with a mean fluctuation≥3.9 mmol/L)based on their BG fluctuations,with 30 cases each.BGrelated indices,tumor markers,serum inflammatory cytokines and adverse reactions were comparatively analyzed.Pearson correlation analysis was performed to analyze the correlation between BG fluctuations and tumor markers.RESULTS The fasting blood glucose and 2-hour postprandial blood glucose levels in the OG were notably elevated compared with those in the CG,together with markedly higher mean amplitude of glycemic excursions(MAGE),mean of daily differences,largest amplitude of glycemic excursions and standard deviation of blood glucose(P<0.05).In addition,the OG exhibited evidently higher levels of carbohydrate antigen 19-9,carbohydrate antigen 125,carcinoembryonic antigen,neuron-specific enolase,cytokeratin 19,tumor necrosis factor-α,interleukin-6,and highsensitivity C-reactive protein than the CG(P<0.05).Pearson analysis revealed a positive association of MAGE with serum tumor markers.The incidence of adverse reactions was significantly higher in the OG than in the CG(P<0.05).CONCLUSION The greater the BG fluctuation in LC patients after chemotherapy,the more unfavorable the therapeuti

AIM:To evaluate the role of genetic factors in the pathogenesis of idiopathic infant cholestasis.METHODS:We performed a case-control study,in-cluding 78 infants with idiopathic infant cholestasis and 113 healthy infants as controls.Genomic DNA was extracted from peripheral venous blood leukocytes us-ing phenol chloroform methodology.Polymerase chain reaction was used to amplify the multidrug resistance protein 3(MDR3)R652G fragment,and products were sequenced using the ABI 3100 Sequencer.RESULTS:The R652G single nucleotide polymorphism(SNP)was significantly more frequent in healthy infants(allele frequency 8.0%)than in patients(allele frequency 2.60%)(P < 0.05),odds ratio,0.29;95% confidence interval,0.12-0.84.The conjugated bilirubin in patients with the AG genotype was significantly lower than in those with the AA genotype(44.70 ± 6.15 μmol/L vs 95.52 ± 5.93 μmol/L,P < 0.05).CONCLUSION:MDR3 R652G is negatively correlated with idiopathic infant cholestasis.Children with the R652G SNP in Guangxi of China may have reduced susceptibility to infant intrahepatic cholestasis.

In the present study, two new limonoids, 1α, 7α-dihydroxyl-3α-acetoxyl-12α-ethoxylnimbolinin(1) and 1α-tigloyloxy-3α-acetoxyl-7α-hydroxyl-12β-ethoxylnimbolinin(2), together with other four known limonoids(3-6), were isolated from the fruits of Melia toosendan. Their structures were elucidated by means of extensive spectroscopic analyses(NMR and ESI-MS) and comparisons with the data reported in the literature. The isolated compounds were evaluated for their antibacterial activities. Compound 4 exhibited significant antibacterial activity against an oral pathogen, Porphyromonas gingivalis ATCC 33277, with an MIC value of 15.2 μg·m L-1. Compound 2 was also active against P. gingivalis ATCC 33277, with an MIC value of 31.25 μg·m L-1. In conlcusion, our resutls indicate that these compounds may provide a basis for future development of novel antibiotics.

A new beam element for analyzing geometrical and physical nonlinearity

Based on Timoshenko＇s beam theory and Vlasov＇s thin-walled member theory, a new model of spatial thin-walled beam element is developed for analyzing geometrical and physical nonlinearity, which incorporates an interior node and independent interpolations of bending angles and warp and takes diversified factors into consideration, such as traverse shear deformation, torsional shear deformation and their coupling, coupling of flexure and torsion, and the second shear stress. The geometrical nonlinear strain is formulated in updated Lagarange （UL） and the corresponding stiffness matrix is derived. The perfectly plastic model is used to account for physical nonlinearity, and the yield rule of von Mises and incremental relationship of Prandtle-Reuss are adopted. Elastoplastic stiffness matrix is obtained by numerical integration based on the finite segment method, and a finite element program is compiled. Numerical examples manifest that the proposed model is accurate and feasible in the analysis of thin-walled structures.
