Asian cultivated rice shows allelic variation in sodium transporter,OsHKT1;5,correlating with shoot sodium exclusion(salinity tolerance).These changes map to intra/extracellularly-oriented loops that occur between four transmembrane-P loop-transmembrane(MPM)motifs in OsHKT1;5.HKT1;5 sequences from more recently evolved Oryza species(O.sativa/O.officinalis complex species)contain two expansions that involve two intracellularly oriented loops/helical regions between MPM domains,potentially governing transport characteristics,while more ancestral HKT1;5 sequences have shorter intracellular loops.We compared homology models for homoeologous OcHKT 1;5-K and OcHKT1;5-L from halophytic O.coarctata to identify complementary amino acid residues in OcHKT1;5-L that potentially enhance affinity for Na+.Using haplotyping,we showed that Asian cultivated rice accessions only have a fraction of HKT1;5 diversity available in progenitor wild rice species(O.nivara and O.rufipogon).Progenitor HKT1;5 haplotypes can thus be used as novel potential donors for enhancing cultivated rice salinity tolerance.Within Asian rice accessions,10 non-synonymous HKT1;5 haplotypic groups occur.More HKT1;5 haplotypic diversities occur in cultivated indica gene pool compared to japonica.Predominant Haplotypes 2 and 10 occur in mutually exclusive japonica and indica groups,corresponding to haplotypes in O.sativa salt-sensitive and salt-tolerant landraces,respectively.This distinct haplotype partitioning may have originated in separate ancestral gene pools of indica and japonica,or from different haplotypes selected during domestication.Predominance of specific HKT1;5 haplotypes within the 3000 rice dataset may relate to eco-physiological fitness in specific geo-climatic and/or edaphic contexts.

Variegated Roles of and Relationships Between Private Tutoring and Schooling: Insights From the State of Maharashtra, India

Purpose:The relationship between private tutoring(PT)and mainstream education is among the complex themes characterizing PT discourses in the literature.This study examined the complications of practices and processes in tutoring and schooling to elucidate different roles played by PT and its relationship with mainstream education.Design/Approach/Methods:This study used qualitative data from a diverse set of 37 PT providers from the State of Maharashtra,India,to delineate their roles and explore their relationship with schooling in this context.Findings:The classification of PT providers roles into complementary,accommodating,competing,and substitutive ones demonstrated a diverse range of relationships between PT and mainstream education.Further analysis showed that these relationships are dynamic in nature,and the boundaries between them are blurred.Originality/Value:Research in the field of PT has been consistently pointing toward a perplexing mixture of positive and negative outcomes resulting from its relationship with mainstream education.This study transcended the positive vis-a-vis negative binary approach by contributing to the deeper understanding of PT relationships.Furthermore,it exemplified how future studies can disentangle the complexities of such relationships by deploying flexible,context-specific theoretical approaches.

Prevention and treatment of hypoxia-induced colorectal cancer damage in albino Wister rats

Background:In the early metastasis of colon cancer,cancer cells detach,migrate,and infiltrate surrounding tissues,including lymph vessels and blood vessels.Tumor heterogeneity arises from both tumor cells and distinct microenvironments.Maldistribution of blood vessels,creates hypoxic regions within the tumors,fostering cancer stem cell-like properties due to reduced oxygen and nutrient supply.Under hypoxia,tumor cells shift to a glycolytic pathway,producing more lactic acid that acidifies the microenvironment and leads to unstable heart rate variability(HRV)factors,weight disparity,and a higher incidence of aberrant crypt foci(ACF).These hypoxic-induced parameters promote cancer cell invasion,increase radiation resistance,and facilitate cancer cell migration.Methods:In this study,we induced hypoxia-preneoplastic colon damage in albino Wister rats by administrating 1,2-dimethyl hydrazine(DMH).After successfully creating a hypoxic environment in albino Wister rats,resulting in preneoplastic colon damage,we randomly allocated Wistar albino rats into seven groups,each containing 8 animals,and conducted a 6-week study.Group 1-Normal control(administered 1 mM EDTA+saline,2 ml/kg/day,p.o.);group 2-Toxic control(administered DMH,30 mg/kg/week,s.c.);group 3-Standard treatment(DMH,30 mg/kg/week,s.***.for 6 weeks),followed by 5-fluorouracil and Leucovorin(25 mg/kg each on 1^(st),3^(rd),7^(th),and 10^(th) days,i.***.after 6 weeks administration of DMH);group 4-Low dose of P1(DMH,30 mg/kg/week,s.c.+P1,2 mg/kg,i.v.,weekly for 3 weeks);group 5-High dose P1(DMH,30 mg/kg/week,s.c.+P1,4 mg/kg,i.v.,weekly for 3 weeks),group 6-Low dose of P2(DMH,30 mg/kg/week,s.c.,+P2,2 mg/kg,i.v.,weekly for 3 weeks),group 7-High dose of P2(DMH,30 mg/kg/week,s.c.,+P2,4 mg/kg,i.***.weekly for 3 weeks).Results:DMH-treated rats exhibited alterations in HRV factors,weight disparity,elevated gastric pH,increased total acidity,a higher incidence of ACF,and changes in antioxidant markers(TBARs,SOD,catalase,GSH).Brightfield microscopy at 40x magnification reveale

Gasification kinetic studies of low volatile weakly caking coal

Present investigation focuses on the fractional conversion of low volatile weakly caking coal(LVWC)under the standard set of operating conditions for gasification.For this purpose,samples of LVWC of different ash content have been collected from Kusunda Area of Bharat Coking Coal Ltd.Gasification results were validated using Homogenous and Shrinking Core kinetic models and the suitability of selected samples for gasification was assessed by estimating the activation energy.The values of activation energy for LVWC samples were obtained in the range of 25.17-44.09 kJ/mol.Further,empirical models were developed to correlate the response of interest with the input variables(temperature,residence time and CO_(2) flow rate).The significance of these developed empirical models was checked using analysis of variance(ANOVA).

Biology of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and the humoral immunoresponse:a systematic review of evidence to support global policy-level actions and research

Background Both population-level epidemiological data and individual-level biological data are needed to control the coronavirus disease 2019(COVID-19)pandemic.Population-level data are widely available and efforts to combat COVID-19 have generated proliferate data on the biology and immunoresponse to the causative pathogen,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).However,there remains a paucity of systemized data on this subject.Objective In this review,we attempt to extract systemized data on the biology and immuno-response to SARS-CoV-2 from the most up-to-date peer-reviewed studies.We will focus on the biology of the virus and immunological variations that are key for determining long-term immunity,transmission potential,and prognosis.Data Sources and Methods Peer-reviewed articles were sourced from the PubMed database and by snowballing search of selected publications.Search terms included:“Novel Coronavirus”OR“COVID-19”OR“SARS-CoV-2”OR“2019-nCoV”AND“Immunity”OR“Immune Response”OR“Antibody Response”OR“Immunologic Response”.Studies published from December 31,2019 to December 31,2020 were included.To ensure validity,papers in pre-print were excluded.Results Of 2889 identified papers,36 were included.Evidence from these studies suggests early seroconversion in patients infected with SARS-CoV-2.Antibody titers appear to markedly increase two weeks after infection,followed by a plateau.A more robust immune response is seen in patients with severe COVID-19 as opposed to mild or asymptomatic presentations.This trend persists with regard to the length of antibody maintenance.However,overall immunity appears to wane within two to three months post-infection.Conclusion Findings of this study indicate that immune responses to SARS-CoV-2 follow the general pattern of viral infection.Immunity generated through natural infection appears to be short,suggesting a need for long-term efforts to control the pandemic.Antibody testing will be essential to gauge the epidemic and inform decision-making on effect

Impact of the coronavirus disease 2019 pandemic on breastfeeding during and at discharge from neonatal care:An observational cohort study

Importance:During the coronavirus disease 2019(COVID-19)lockdown,changes in the visiting rules in neonatal units might have affected the initiation and continuation of breastfeeding.Objective:To investigate the effects of the implementation of the COVID-19 lockdown in the UK on mother’s own milk(MOM)feeding in hospital and at the time of discharge in two UK neonatal units.Methods:Retrospective cohort study using routinely recorded data from electronic patient records.Data were retrieved from two neonatal services in the UK East Midlands region.Adjusted logistic regression was used to compare the odds of MOM feeding before,and after the implementation of the UK lockdown.Results:Among 2073 infants,after adjusting for maternal and infant characteristics and underlying trends over time,there were no differences in the odds of infants receiving any MOM during admission;any MOM at discharge or exclusive MOM at discharge before and after the imposition of the lockdown.Infants with birthweight2500 g(adjusted odds ratio[OR]0.33,95%confidence interval[CI]:0.22–0.50).Younger mothers were less likely,and Black British mothers more likely,to be feeding MOM to their infants at discharge,while women in the least deprived Index of Multiple Deprivation(IMD)quintiles were 2–4 times more likely to do so,compared to those in the most deprived IMD quintile(adjusted OR 2.78,95%CI:1.97–3.90).Interpretation:Despite the difficulties faced during COVID-19 pandemic-induced restrictions,infants in the participating neonatal units continued to receive MOM in similar proportions as before the pandemic.

Role of beneficial microbial gene pool in mitigating salt/nutrient stress of plants in saline soils through underground phytostimulating signalling molecules

Soil salinity diminishes soil health and reduces crop yield,which is becoming a major global concern.Salinity stress is one of the primary stresses,leading to several other secondary stresses that restrict plant growth and soil fertility.The major secondary stresses induced in plants under saline-alkaline conditions include osmotic stress,nutrient limitation,and ionic stress,all of which negatively impact overall plant growth.Under stressed conditions,certain beneficial soil microflora are known to have evolved phytostimulating mechanisms,such as the synthesis of osmoprotectants,siderophores,1-aminocyclopropane-1-carboxylic acid(ACC)deaminase activity,phosphate solubilization,and hormone production,which enhance plant growth and development while mitigating nutrient stress.Beneficial soil-borne bacterial species such as Bacillus,Pseudomonas,and Klebsiella and fungal strains such as Trichoderma,Aspergillus,Penicillium,Alternaria,and Fusarium also aid in reducing salinity stress.Phosphate-solubilizing microorganisms also assist in nutrient acquisition via both enzymatic and non-enzymatic processes.In the case of enzymatic processes,they produce different enzymes such as alkaline phosphatases and phytases,whereas non-enzymatic processes produce organic acids such as gluconic,citric,malic,and oxalic acids.The native halotolerant/halophilic soil microbial gene pool with multifunctional traits and stress-induced gene expression can be developed as suitable bio-inoculants to enhance stress tolerance and optimize plant growth in saline soils.

Antioxidant and antiarthritic potential of berberine: In vitro and in vivo studies

Objective: To extract and isolate berberine from Berberis aristata(Berberidaceae). Isolated berberine was characterised using spectroscopy and its antioxidant and antiarthritic activity was analyzed.Methods: The berberine was isolated from B. aristata using microwave-assisted extraction(MAE) and characterised by a spectroscopic technique. The isolated berberine was evaluated for its antioxidant activity in DPPH, nitric oxide, and superoxide scavenging assays, while antiarthritic activity was evaluated in the complete freund’s adjuvant(CFA)-induced arthritis rat model.Results: The antioxidant activity of berberine revealed potent antioxidant activity in DPPH, nitric oxide,and superoxide scavenging assays. The in vivo antiarthritic activity of berberine in the CFA-induced arthritis rat model showed a significant reduction in paw diameter, arthritic score, and an increase in body weight. Furthermore, a concentration-dependent ameliorating action of berberine on haematological parameters was noticed. Proinfammatory biomarkers, including IL-6, IL-10, and TGF-b in serum were reported, and histopathology examination revealed that berberine decreased pannus formation, synovial hyperplasia, and bone erosion. Radiographic investigation showed soft tissue infammation, bone resorption and erosion, joint gap reduction, and substantial connective tissue expansion after treatment with berberine.Conclusion: The ameliorating action on haematological parameters and proinfammatory biomarkers of berberine makes them a suitable remedy for the treatment of arthritis.
