Recombinant outer membrane protein F-B subunit of LT protein as a prophylactic measure against Pseudomonas aeruginosa burn infection in mice

AIM: To study immunogenicity of outer membrane protein F(Opr F) fused with B subunit of LT(LTB), against Pseudomonas aeruginosa(P. aeruginosa). METHODS: The Opr F, a major surface exposed outer membrane protein that is antigenically conserved in various strains of P. aeruginosa, is a promising immunogen against P. aeruginosa. In the present study recombinant Opr F and Opr F-LTB fusion gene was cloned, expressed and purified. BALB/c mice and rabbits were immunized using recombinant Opr F and Opr F-LTB and challenged at the burn site with P. aeruginosa lethal dose of 104 CFU. The protective efficacy of rabbit anti Opr F Ig G against P. aeruginosa burn infection was investigated by passive immunization. RESULTS: It has been well established that the LTB is a powerful immunomodulator with strong adjuvant activity. LTB as a bacterial adjuvant enhanced immunogenicity of Opr F and anti Opr F Ig G titer in serum was increased. Experimental findings showed significantly higher average survival rate in burned mice immunized with Opr F-LTB than immunized with Opr F or the control group. Rabbits anti Opr F Ig G brought about 75% survival of mice following challenge with P. aeruginosa. Post challenge hepatic and splenic tissues of mice group immunized with Opr F-LTB had significantly lower bacterial load than those immunized with Opr F or the control groups. CONCLUSION: These results demonstrate that LTBfused Opr F might be a potential candidate protein for a prophylactic measure against P. aeruginosa in burn infection.

Adjuvant role of Pseudomonas flagellin for Acinetobacter baumannii biofilm associated protein

AIM To study immunogenicity of Pseudomonas N terminal flagellin as an adjuvant for Acinetobacter baumannii(A. baumanni) biofilm associated protein(Bap).METHODS The N terminal flagellin gene was amplified. The p ET28a(+) and polymerase chain reaction products weredigested with HindⅢ and Eco R Ⅰ. The ligation of N terminal flagellin into p ET28a(?+) was performed using T4 DNA ligase and was then transformed into Escherichia coli BL21(DE3) as a suitable expression host. p ET28a(?+) vector harboring a conserved region of Bap from our previous work was used. The recombinant proteins were expressed, analyzed by SDS-PAGE method and was purified by affinity chromatography with His-Tag residues followed by confirmation with western blotting. Mice were immunized with recombinant N terminal flagellin and Bap subunits. The immunized animals were intranasally(i.n) challenged with A. baumannii and Pseudomonas aeruginosa(P. aeruginosa).RESULTS The flagellin enhanced the immunogenicity of Bap causing an increase in specific Ig G titers in serum(P < 0.001). Internal organs, i.e., liver, lung and spleen of the BapFlagellin immunized group challenged with A. baumannii showed significantly lower bacterial load compared to the control group. The bacterial loads were studied in internal organs. A. baumannii infected immunized animals with Bap-Flagellin exhibited internal organs with minor bacterial load while P. aeruginosa PAO1 infected group showed heavy bacterial load of(4.3 ± 0.12) × 106,(1.1 ± 0.01) × 106 and(2.2 ± 0.22) × 106 per gram of lungs, liver and spleen respectively. Bacterial loads were detected per gram of lungs, liver and spleen of the mice group immunized with Bap were(1.2 ± 0.06) × 107,(11.1 ± 0.041) × 105 and(3.6 ± 0.42) × 106 respectively. In vivo neutralization assay indicated that all experimental mice groups, except for Flagellin administered group was significantly(P < 0.05) protected against A. baumannii. Objective： The complexity, heterogeneity and capacity of malignant neoplastic cells and tumors for rapid change and evolution suggest that living-cell-based biological-systems approaches to cancer treatment are merited. Testing this hypothesis, the tumor marker, metabolic activity, and overall survival（OS） responses, to the use of one such system, implantable macrobeads [RENCA macrobeads（RMBs）], in phase I and IIa clinical trials in advanced,treatment-resistant metastatic colorectal cancer（m CRC） are described ***： Forty-eight m CRC patients（30 females; 18 males）, who had failed all available, approved treatments,underwent RMB implantation（8 RMB/kg body weight） up to 4 times in phase I and phase IIa open-label ***, labs [tumor and inflammation markers, lactate dehydrogenase（LDH）] and positron emission tomography-computed tomography（PET-CT） imaging to measure number/volume and metabolic activity of the tumors were performed pre-and 3-month-post-implantation to evaluate safety and initial efficacy（as defined by biological responses）. PET-CT maximum standard uptake value（SUVmax）（baseline and d 90; SUVmax ≥2.5）, LDH,and carcinoembryonic antigen（CEA） and/or cancer antigen 19-9（CA 19-9） response（baseline, d 30 and/or d 60）were assessed and compared to ***： Responses after implantation were characterized by an at least 20% decrease in CEA and/or CA 19-9 in75% of patients. Fluorodeoxyglucose（FDG）-positive lesions（phase I, 39; 2 a, 82） were detected in 37/48 evaluable patients, with 35% stable volume and stable or decreased SUV（10） plus four with necrosis; 10, increased tumor volume, SUV. LDH levels remained stable and low in Responders（R）（d 0–60, 290.4–333.9）, but increased steadily in Non-responders（NR）（d 0–60, 382.8–1,278.5）（d 60, P=0.050）. 背景虽然目前植入型心脏复律除颤器(ICD)已经越来越多地应用到严重心衰患者(心功能III/IV级)中,但是其应用受到泵衰竭容易引起死亡以及数据较少的限制。本研究的目的是评估QRS波群宽度和疾病病因对植入ICD的严重心衰患者生存率和再入院率的影响。方法纳入了1996—2000年间出院的438例心衰患者(年龄56±15岁,心脏射血分数29%±15%,76%患者的心脏分级为III/IV级)进行回顾性研究。结果438例患者中有94例因非持续性(51.1%)或持续性室性心动过速(24.4%)、晕厥(16.0%)或出于预防目的(8.5%)植入了ICD。所有植入ICD患者的基线平均射血分数较低(25%vs30%,P=0.007),但符合心功能IV级症状的患者比例并不高(33%vs44%,P=0.164)。结果发现,植入ICD的患者具有较高的生存率(RR0.48,P=0.001),但其再入院率也较高(RR1.41,P=0.031)。在缺血性和非缺血性亚组分析中,植入ICD患者的生存率具有明显的优势(RR0.38,P=0.003;RR0.54,P=0.042)。QRS<120ms并植入ICD的患者具有生存率较高的趋势(RR0.42,P=0.119),但其再入院率也升高(RR2.13,P=0.021)。QRS≥120ms并植入ICD的患者具有较高的生存率(RR0.45,P=0.001),而再入院率不升高(RR1.22,P=0.314)。结论严重心衰患者植入ICD可以比未植入患者获得更高的生存率,但是其再住院率也升高。而且该治疗对QRS波群宽度延长患者的益处更明显。

Micro-computed tomography assessment of human femoral trabecular bone for two disease groups(fragility fracture and coxarthrosis):Age and gender related effects on the microstructure

The aim of this study was to identify three-dimensional microstructural changes of trabecular bone with age and gender, using micro-computed tomography. Human trabecular bone from two disease groups, osteoporosis and osteoarthritis was analyzed. A prior analysis of the effects of some procedure variables on the micro-CT results was performed. Preliminary micro-CT scans were performed with three voxel resolutions and two acquisition conditions. On the reconstruction step, the image segmentation was performed with three different threshold values. Samples were collected from patients, with coxarthrosis (osteoarthritis) or fragility fracture (osteoporosis). The specimens of the coxarthrosis group include twenty females and fifteen males, while the fragility fracture group was composed by twenty three females and seven males. The mean age of the population was 69 ± 11 (females) and 67 ± 10 years (males), in the coxarthrosis group, while in the fragility fracture group was 81 ± 6 (females) and 78 ± 6 (males) years. The 30 μm voxel size provided lower percentage difference for the microarchitecture parameters. Acquisition conditions with 160 μA and 60 kV permit the evaluation of all the volume’s sample, with low average values of the coefficients of variation of the microstructural parameters. No statistically significant differences were found between the two diseases groups, neither between genders. However, with aging, there is a decrease of bone volume fraction, trabecular number and fractal dimension, and an increase of structural model index and trabecular separation, for both disease groups and genders. The parameters bone specific surface, trabecular thickness and degree of anisotropy have different behaviors with age, depending on the type of disease. While in coxarthrosis patients, trabecular thickness increases with age, in the fragility fracture group, there is a decrease of trabecular thickness with increasing age.
