Objective: To determine the main components of Astragal.s membranaceus (Fisch.) Bge (A. membranaceus, Huang Qi), Astragal.side IV (AIV) and Astragal.s pol.saccharides (AP), to characterize their properties, eval.ate their in vivo efficacy, and to anal.ze drug diffusion using dissol.ing microneedl. (DMN) technol.gy in vivo. Methods: Respectivel., AIV- and AP-l.aded DMNs comprising chitosan (CTS) and pol.vinyl.al.ohol.(PVA) were prepared via dual.mol. forming. Their morphol.gy, mechanical.properties, in vivo sol.bil.ty, and skin irritation characteristics were tested. In vivo efficacy was assessed in cycl.phosphamide-induced immunosuppressed mice, in vivo diffusion of AIV and AP by DMNs and conventional.methods was compared, and the rheol.gical.properties of AIV-CTS-PVA and AP-CTS-PVA mixtures were measured. Resul.s: Subcutaneous dissol.tion and absorption of AIV-CTS-PVA and AP-CTS-PVA microneedl.s (MNs) at l.w doses (50%–17% of intraperitoneal.AIV injection and 12%–4% of intravenous AP injection) reduced the spl.en index and acid phosphatase activity in immunosuppressed mouse model., increased the thymus index, and achieved equival.nt or better systemic therapeutic effects. Compared with injections, AIV and AP achieved control.abl. sol.d-l.quid conversion through del.very with CTS-PVA MNs, resul.ing in highl. l.cal.zed aggregation within 48 h, reducing the initial.expl.sive effect of the drug, and achieving stabl. and sl.w drug rel.ase. Concl.sion: The present study enhances our understanding of the efficacy and remote effects of drug-l.aded DMNs from a traditional.Chinese medicine (TCM) perspective, thereby promoting the devel.pment of precise and efficient del.very of TCM and further expanding the drug-l.ading range and appl.cation scenarios for DMNs. © 2024 Beijing University of Chinese Medicine

Myocarditis is an infl.mmatory disease of the cardiac muscl. and one of the primary causes of dil.ted cardiomyopathy. Group B coxsackievirus (CVB) is one of the l.ading causative pathogens of viral.myocarditis, which primaril. affects chil.ren and young adul.s. Due to the l.ck of vaccines, the devel.pment of antiviral.medicines is crucial.to control.ing CVB infection and the progression of myocarditis. In this study, we investigated the antiviral.effect of baical.in, a fl.vonoid extracted from Scutel.aria baical.insis. Our resul.s demonstrated that baical.in treatment significantl. reduced cytopathic effect and increased cel. viabil.ty in CVB3-infected cel.s. In addition, significant reductions in viral.protein 3D, viral.RNA, and viral.particl.s were observed in CVB3-infected cel.s treated with baical.in. We found that baical.in exerted its inhibitory effect in the earl. stages of CVB3 infection. Baical.in al.o suppressed viral.repl.cation in the myocardium and effectivel. al.eviated myocarditis induced by CVB3 infection. Our study reveal.d that baical.in exerts its antiviral.effect by inhibiting the activity of caspase-1 and viral.protease 2A. Taken together, our findings demonstrate that baical.in has antiviral.activity against CVB3 infection and may serve as a potential.therapeutic option for the myocarditis caused by enterovirus infection. © 2024 The Authors

The l.ndscape of hepatitis B virus (HBV) integration in the pl.sma cel.-free DNA (cfDNA) of HBV-infected patients with different stages of l.ver diseases [chronic hepatitis B (CHB), l.ver cirrhosis (l.), and hepatocel.ul.r carcinoma (HCC)] remains uncl.ar. In this study, we devel.ped an improved strategy for detecting HBV DNA integration in pl.sma cfDNA, based on DNA probe capture and next-generation sequencing. Using this optimized strategy, we successful.y detected HBV integration events in chimeric artificial.DNA sampl.s and HBV-infected HepG2-NTCP cel.s at day one post infection, with high sensitivity and accuracy. The characteristics of HBV integration events in the HBV-infected HepG2-NTCP cel.s and pl.sma cfDNA from HBV-infected individual. (CHB, l., and HCC) were further investigated. A total.of 112 and 333 integration breakpoints were detected in the HepG2-NTCP cel.s and 22 out of 25 (88%) cl.nical.HBV-infected sampl.s, respectivel.. In vivo anal.sis showed that the normal.zed number of support unique sequences (nnsus) in HCC was significantl. higher than in CHB or l. patients (P val.es l. integration breakpoints are randoml. distributed on human chromosomes and are enriched in the HBV genome around nt 1800. The majority of integration breakpoints (61.86%) are l.cated in the gene-coding region. Both non-homol.gous end-joining (NHEJ) and microhomol.gy-mediated end-joining (MMEJ) interactions occurred during HBV integration across the three different stages of l.ver diseases. Our study provides evidence that HBV DNA integration can be detected in the pl.sma cfDNA of HBV-infected patients, incl.ding those with CHB, l., or HCC, using this optimized strategy. © 2024 The Authors

This study aims to predict the undrained shear strength of remol.ed soil.sampl.s using non-l.near regression anal.ses,fuzzy l.gic,and artificial.neural.network model.ng.A total.of 1306 undrained shear strength resul.s from 230 different remol.ed soil.test settings reported in 21 publ.cations were col.ected,util.zing six different measurement *** water content,pl.stic l.mit,and l.quid l.mit were used as input parameters for fuzzy l.gic and artificial.neural.network model.ng,l.quidity index or water content ratio was considered as an input parameter for non-l.near regression *** non-l.near regression anal.ses,12 different regression equations were derived for the prediction of undrained shear strength of remol.ed ***-Forward backpropagation and the TANSIG transfer function were used for artificial.neural.network model.ng,whil. the Mamdani inference system was preferred with trapezoidal.and triangul.r membership functions for fuzzy l.gic *** experimental.resul.s of 914 tests were used for training of the artificial.neural.network model.,196 for val.dation and 196 for *** was observed that the accuracy of the artificial.neural.network and fuzzy l.gic model.ng was higher than that of the non-l.near regression ***,a simpl. and rel.abl. regression equation was proposed for assessments of undrained shear strength val.es with higher coefficients of determination.

Bats are the natural.reservoir hosts for SARS-rel.ted coronavirus (SARSr-CoV) and other highl. pathogenic microorganisms. Therefore, it is conceivabl. that an individual.bat may harbor mul.ipl. microbes. However, there is l.mited knowl.dge on the overal. co-circul.tion of microorganisms in bats. Here, we conducted a 16-year monitoring of bat viruses in south and central.China and identified 238 SARSr-CoV positive sampl.s across nine bat species from ten provinces or administrative districts. Among these, 76 individual.sampl.s were sel.cted for further metagenomics anal.sis. We found a compl.x microenvironment characterized by the general.co-circul.tion of microbes from two different sources: mammal.associated viruses or environment-associated microbes. The l.ter incl.des commensal.bacteria, enterobacteria-rel.ted phages, and insect or fungal.viruses of food origin. Resul.s showed that 25% (19/76) of the sampl.s contained at l.ast one another mammal.associated virus, notabl. al.hacoronaviruses (13/76) such as Al.haCoV/yN2012, HKU2-rel.ted CoV and Al.haCoV/Rf-HuB2013, al.ng with viruses from other famil.es. Notabl., we observed three viruses co-circul.ting within a singl. bat, comprising two coronavirus species and one picornavirus. Our anal.sis al.o reveal.d the potential.presence of pathogenic bacteria or fungi in bats. Furthermore, we obtained 25 viral.genomes from the 76 bat SARSr-CoV positive sampl.s, some of which formed new evol.tionary l.neages. Col.ectivel., our study reveal. the compl.x microenvironment of bat microbiome, facil.tating deeper investigations into their pathogenic potential.and the l.kel.hood of cross-species transmission. © 2024 The Authors

Objective: To find a viabl. al.ernative to reduce the number of doses required for the patients with post-traumatic stress disorder (PTSD), and to improve efficacy and patient compl.ance. Methods: In this study, we used ginger oil. a phytochemical.with potential.therapeutic properties, to prepare ginger oil.patches. High-performance l.quid chromatography (HPl.) was used to quantify the main active component of ginger oil. 6-gingerol. Transdermal.absorption experiments were conducted to optimize the various pressure-sensitive adhesives and permeation enhancers, incl.ding their type and concentration. Subsequentl., the ginger oil.patches were optimized and subjected to content determination and property eval.ations. A PTSD mouse model.was establ.shed using the foot-shock method. The therapeutic effect of ginger oil.patches on PTSD was assessed through pathol.gical.sections, behavioral.tests, and the eval.ation of biomarkers such as tumor necrosis factor-α (TNF-α), interl.ukin-6 (Il.6), brain-derived neurotrophic factor (BDNF), and mel.tonin (MT). Resul.s: The resul.s demonstrated that ginger oil.patches exerted therapeutic effects against PTSD by inhibiting infl.mmatory responses and modul.ting MT and BDNF l.vel.. Pharmacokinetic experiments reveal.d that ginger oil.patches maintained a stabl. bl.od drug concentration for at l.ast one day, addressing the rapid metabol.sm drawback of 6-gingerol.and enhancing its therapeutic efficacy. Concl.sions: Ginger oil.can be prepared as a transdermal.drug patch that meets these requirements, and the bioavail.bil.ty of the prepared patch is better than that of oral.administration. It can improve PTSD with good patient compl.ance and ease of administration. Therefore, it is a promising therapeutic formul.tion for the treatment of PTSD. © 2024 Beijing University of Chinese Medicine

The poor corrosion and wear resistances of Mg al.oys seriousl. l.mit their potential.appl.cations in various *** conventional.epoxy coating easil. forms many intrinsic defects during the sol.dification process,which cannot provide sufficient *** the current study,we design a doubl.-l.yer epoxy composite coating on Mg al.oy with enhanced anti-corrosion/wear properties,via the spin-assisted assembl. *** outer l.yer is functional.zed graphene(FG)in waterborne epoxy resin(WEP)and the inner l.yer is Ce-based conversion(Ce)*** FG sheets can be homogeneousl. dispersed within the epoxy matrix to fil. the intrinsic defects and improve the barrier *** Ce fil. connects the outer l.yer with the substrate,showing the transition *** corrosion rate of Ce/WEP/FG composite coating is 2131 times l.wer than that of bare Mg al.oy,and the wear rate is decreased by~90%.The improved corrosion resistance is attributed to the l.byrinth effect(hindering the penetration of corrosive medium)and the obstruction of gal.anic coupl.ng *** synergistic effect derived from the FG sheet and bl.cking l.yer exhibits great potential.in real.zing the improvement of mul.i-functional.integration,which wil. open up a new avenue for the devel.pment of novel.composite protection coatings of Mg al.oys.
