Human CD4 ＋ effector T lymphocytes generated upon TCR engagement with self-peptides respond defectively to IL-7 in their transition to memory cells

The peripheral repertoire of CD4＋ T lymphocytes contains autoreactive cells that remain tolerant through several mechanisms. However, nonspecific CD4＋ T cells can be activated in physiological conditions as in the course of an ongoing immune response, and their outcome is not yet fully understood. Here, we investigate the fate of human naive CD4＋ lymphocytes activated by dendritic cells （DCs） presenting endogenous self-peptides in comparison with lymphocytes involved in alloresponses. We generated memory cells （Tmem） from primary effectors activated with mature autologous DCs plus interleukin （IL）-2 （Tmauto）, simulating the circumstances of an active immune response, or allogeneic DCs （Tmallo）. Tmem were generated from effector cells that were rested in the absence of antigenic stimuli, with or without IL-7. Tmem were less activated than effectors （demonstrated by CD25 downregulation） particularly with IL-7, suggesting that this cytokine may favour the transition to quiescence. Tmauto and TmaHo showed an effector memory phenotype, and responded similarly to polyclonal and antigen-specific stimuli. Biochemically, IL-7-treated Tma^o were closely related to conventional memory lymphocytes based on Erk-l/2 activation, whereas Tmauto were more similar to effectors. Autologous effectors exhibited lower responses to IL-7 than allogeneic cells, which were reflected in their reduced proliferation and higher cell death. This was not related to IL-7 receptor expression but rather to signalling deficiencies, according to STAT5 activation These results suggest that ineffective responses to IL-7 could impair the transition to memory cells of naive CD4＋ T lymphocytes recognizing self-peptides in the setting of strong costimulation.

Risk Groups and Mortality in patients with SARS-CoV-2 from Family Medicine Unit no. 43 in Southeastern Mexico

SARS-CoV-2 infection is a viral disease that causes respiratory infections that can cause multiple complications. patients with adjacent morbidities have a higher chance of complications and mortality. The national Health and nutrition Survey (EnSAnUT) in 2018 reported that in Mexico, 75.2% of adults aged 20 years and over are overweight and obese, both situations are closely related to the cases reported as severe COVID and even with a higher risk of mortality. The main objective was to evaluate the risk and mortality groups in patients with SARS-CoV-2 from family medicine in the south of México. The study was carried out during the 2020 year, in the Family Medicine Unit no. 43 of Villahermosa, Tabasco, a cross-sectional, retrospective investigation was carried out with an analytical approach, and the sample was obtained using the formula of n for infinite populations that correspond to 215, patients were included who met the selection criteria;3 groups were integrated: DM2, HAS and obesity. Descriptive statistics andχp>2p> formula were used, using the SpSS version 25 Windows program. The female gender presented the highest number of infections 122 (56.7%) and mortality: 5 (2.32%), obesity comorbidity presented the highest number of infections: 194 (90.2%), HAS: 153 (71.2%), and MD: 44 (20.46%). Complications were observed in 23 (10.69%), distributed in: SIRA: 7 (3.3%), multiple organ failure: 6 (2.8%), heart failure: 6 (2.8%) and pneumonia 4 (1.9%). The observed mortality was 8 (3.72%). The comorbidities of patients with the highest mortality were: SAH: 6 (75%) (χp>2p> = 9.169;df = 1;p = 0.010) and obesity: 6 (75%) (χp>2p> = 9.169;df = 1;p = 0.010). The group with the highest risk for SARS-CoV-2 was patients with overweight/obesity comorbidities. Mortality was 3.7%. The patients who died had at least one or more comorbidities. The main complications were ARIS, heart failure, and multiple organ failure.

The starting point of this research is the urban model promoted by the United nations through the Sustainable Development Goals(SDGs).The importance of public spaces is especially highlighted in Goal 11.7:“provide universal access to safe,inclusive,and accessible,green,and public spaces.”The quality of public spaces can be properly assessed by reconsidering the quality indicators and investigating their development potential,which is the main focus of our *** study is based on a theoretical and methodological reflection on the quality of public space,considering the contributions of some contemporary urban traditions and a few recent *** this perspective,a variety of public space projects developed in the European sphere have been selected and *** quality parameters identified in this initial analysis were then applied to the analysis of nine representative case studies in three Spanish cities(Madrid,Barcelona,and Zaragoza).The main goal is to propose a methodology that helps to evaluate the quality of those public spaces and determine its relationship with the safety,accessibility,and inclusiveness conditions they offer.

Reproductive patterns in Araucaria araucana forests in the Andean range, Chile

Background Araucaria araucana is a mast species that presents a high variability in annual cone production. Researchers have recorded synchronization events in cone production in different populations, which allows the seed production to be concentrated, reducing the percentage of seeds consumed by different animal *** We sampled three populations located in the Andes Mountains, Araucanía Region, Chile. In 2004 we began the collection of data on cone production, for which we installed permanent plots（1200 m2each） at each location. We identified and labeled each female tree in each plot to monitor its cone production. In 2012 we selected a total of 30 trees near the plots to evaluate the number of seeds per cone. In each February from 2012 to 2014 we selected two mature cones and covered them with a porous mesh for subsequent collection and storage in March. At the beginning of June, we counted and weighed the seeds, determining the average weight, the number of seeds per cone, the germination capacity（GC）, and the germination speed（GS）.Results Cone production was synchronous across the three locations. We observed significant differences among the locations and years evaluated. The cones had fewer seeds in 2013（high production） compared to those in 2012（low production）, but their weights were similar. In 2014 the cones produced smaller seeds in fewer quantities. The difference between the years 2013 and 2014 resulted from the high-energy expenditure in 2013. The hydraulic fracturing is a nonlinear,fluid-solid coupling and transient problem,in most cases it is always time-consuming to simulate this process *** recent years,although many numerical methods were proposed to settle this problem,most of them still require a large amount of computer *** it is a high demand to develop more efficient numerical approaches to achieve the real-time monitoring of the fracture geometry during the hydraulic fracturing *** this study,a reduced order modeling technique namely proper Generalized Decomposition(pGD),is applied to accelerate the simulations of the transient,non-linear coupled system of hydraulic fracturing problem,to match this extremely tight response time *** separability of the solution in space and time dimensions is studied for a simplified model *** solid and fluid equations are coupled explicitly by inverting the solid discrete problem,and a simple iterative procedure to handle the non-linear characteristic of the hydraulic fracturing problem is proposed in this *** validation illustrates that the results of pGD match well with these of standard finite element method in terms o f fracture opening and fluid pressure in the ***,after the off-line calculations,the numerical results can be obtained in real time.

performance evaluation of lateral canopy shakers with catch frame for continuous harvesting of oranges for juice industry

Citrus is mainly oriented to fresh consumption,and harvesting is usually performed ***,the high cost of harvesting and the low availability of labour can compromise the profitability of the crop when it is destined to juice *** development of citrus mechanical harvesting for industrial processing is conditioned to reach a high fruit removal efficiency,with a reduced damage to both fruit and *** current machines available are very large,requiring extensive plantations with long rows of trees to be *** this study,two lateral canopy shakers equipped with a catch frame were evaluated to harvest independently both sides of the hedge on intensive citrus plantations with the main objective of determining their performance and *** lateral canopy shakers tested were three tractor-drawn machines,one commercial machine and two *** tested machines reached a mean value of 78%of fruit ***,the prototypes,equipped with a catch frame,were able to recover a mean value of 70%of *** the results were promising,for achieving an efficient result,the application of this harvesting technology still requires a process of improvement,and the adaptation of both the machine and the *** machines should reduce the amount of post-harvest ground fruit(5.9%-10.4%).Tree damages generated by the contact of the catch frame with the trunk and the metal rods with main branches were the most ***,it is still necessary to increase the ground speed of the machinery and improving the design of the rods,regulating the rod penetrating deep in the canopy to improve the fruit recovery and limit the damage caused to the trees.

Differentiated payments for environmental services schemes: A methodology proposal

payment schemes for environmental services face serious budget constraints in Mexico. payments are equally distributed among forest owners, and the governmental agencies that make payments to beneficiaries do not evaluate the amount of environmental services produced by forested areas. This lack of evaluation could lead to economic and environmental deficiencies that restrict the possible benefits of such programmes. In general, these programmes do not achieve their conservation objectives, and local beneficiaries are not involved in the design or application of these programmes. Similarly, the payments for Hydrological Environmental Services(pHES) programme of the State of Mexico, Mexico, makes equal payments to beneficiaries of forested areas considering tree density. The objective of the present work was to create a methodology for determining differentiated payments schemes for hydrological environmental services, prioritising areas that contribute the most to groundwater recharge and promote the participation of society. In the nevado de Toluca natural protected Area, the environmental criteria of forest type, tree density and aquifer overexploitation as well as social participation were considered in a multi-criteria analysis implemented in a Geographic Information System. A methodology was developed to determine differentiated payments based on specific forest characteristics that influence the potential capacity of forests to recharge groundwater reserves in addition to the level of social participation. In particular, social participation is key as it could ensure the viability of pHES programmes as conservation alternatives in forested areas with high groundwater recharge potential.

Diet composition and its ecological drivers are rarely investigated in coexisting closely related *** used a molecular approach to characterize the seasonal variation in diet composition in four spiny lizard species inhabiting a mountainous *** metabarcoding revealed that the lizards Sceloporus aeneus,***,***,and *** mostly consumed arthropods of the orders Hemiptera,Araneae,Hymenoptera,and *** terrestrial lizards *** and *** mostly predated ants and spiders,whereas the arboreal–saxicolous *** and saxicolous *** largely consumed grasshoppers and *** taxonomic and phylogenetic diversity of the prey was higher during the dry season than the rainy season,likely because reduced prey availability in the dry season forced lizards to diversify their diets to meet their nutritional *** and phylogenetic composition varied seasonally depending on the species,but only dietary composition varied with *** dietary turnover was greater in *** than in ***,suggesting site-specific seasonal variability in prey availability;no other differences among species were ***,which lives at the highest altitude in our study site,displayed interseasonal variation in diet *** differences were correlated with the species’feeding strategies and elevational distribution,which likely contributed to the coexistence of these lizard species in the studied geographic area and beyond.
