BACKGROUND Colorectal cancer(CRC) is the second most common cause of cancer death worldwide. It is broadly described that cyclooxygenase-2(COX-2) is mainly overexpressed in CRC but less is known regarding post-translational modifications of this enzyme that may regulate its activity, intracellular localization and stability. Since metabolic and proteomic profile analysis is essential for cancer prognosis and diagnosis, our hypothesis is that the analysis of correlations between these specific parameters and COX-2 state in tumors of a high number of CRC patients could be useful for the understanding of the basis of this cancer in *** To analyze COX-2 regulation in colorectal cancer and to perform a detailed analysis of their metabolic and proteomic *** Biopsies from both healthy and pathological colorectal tissues were taken under informed consent from patients during standard colonoscopy procedure in the University Hospital of Bellvitge(Barcelona, Spain) and Germans Trias i Pujol University Hospital(Campus Can Ruti)(Barcelona, Spain). Western blot analysis was used to determine COX-2 levels. Deglycosylation assays were performed in both cells and tumor samples incubating each sample with peptide N-glycosidase f(PNGase f). Prostaglandin E2(PGE2) levels were determined using a specific ELISA. 1 H high resolution magic angle spinning(HRMAS) analysis was performed using a Bruker AVIII 500 MHz spectrometer and proteomic analysis was performed in a nano-liquid chromatography-tandem mass spectrometer(nano LC-MS/MS) using a QExactive Hf orbitrap *** Our data show that COX-2 has a differential expression profile in tumor tissue of CRC patients vs the adjacent non-tumor area, which correspond to a glycosylated and less active state of the protein. Stability Analysis of a Neurocontroller with Kalman Estimator for the Inverted Pendulum Case

In this paper, a practical analysis of stability by simulation for the effect of incorporating a Kalman estimator in the control loop of the inverted pendulum with a neurocontroller is presented. The neurocontroller is calculated by approximate optimal control, without considering the Kalman estimator in the loop following the Theorem of the separation. The results are compared with a time-varying linear controller, which in noiseless conditions in the state or in the measurement has an acceptable performance, but when it is under noise conditions its operation closes into a state space range more limited than the one proposed here.

Water Quality Assessment of Jose Antonio Alzate Dam, the Lerma River and Its Tributaries in the State of Mexico, Mexico

Water quality was evaluated at sites of the Jose Antonio Alzate Dam, the rivers Lerma, Tejalpa and Temoaya in the State of Mexico, Mexico. The variables measured included pH, conductivity, temperature, dissolved oxygen (DO), ammonia, nitrates, sulfates, sulfides and metal concentrations (Cu and Pb). Chemical speciation for dissolved metals in the aqueous phase was obtained using measured concentrations and the software MINEQL+ (4.5);metal partition coefficients was also obtained using dissolved and particulate concentrations of metals. Hydraulic behavior of the studied water bodies was investigated and the fate of Cu and Pb ions was simulated. The simulation results indicate that the Lerma River provides the majority of organic matter and metal contaminants as a result of a poor oxygen saturation velocity, and showed a slight improvement when the Lerma River is subjected to its tributaries Tejalapa and Temoaya Rivers. Under the reducing conditions predominating along the dam and from the results of aqueous phase chemical speciation, it was found that sulfide metal-associated species are the ones that predominate. It was demonstrated that in general, the rivers Tejalapa and Temoaya presented the best water quality. The Lerma River oxygen saturation improves after the rainy season, but it worsens during the dry season. Due to irregular topography, the small slope prevailing in the Lerma River, and the almost nonexisting water flow, zones with different characteristics are formed within the dam. The sedimentation contributes in the generation of local eddies causing the existence of a small diffusive flow. The order of partition coefficient was Pb > Cu. Cu presented the highest risk levels in the dissolved phase.

Methicillin-resistant Staphylococcus aureus is an increasingly important cause of nosocomial bacterial infections worldwide. S. aureus is responsible for several pathologies, including skin infections, endocarditis, meningitis, deep-tissue ulcers, and sepsis. S. aureus biofilm formation on catheters and other medical devices is a major post-operative concern, because biofilms are often the source of persistent and difficult to treat bacterial infections. While catheter-related S. aureus infections have been reported, the strains responsible for these infections have not been genetically characterized. We genetically characterized S. aureus strains isolated from hemodialysis catheters in Mexican patients. The frequency of 35 genes coding for adhesins, toxins, and other virulence-associated products in the 55 isolated S. aureus strains was determined using PCR, while real-time PCR was used to examine the level of gene expression. Of the 55 S. aureus strains isolated from 109 patients, 45 (81.8%) were determined to be methicillin-resistant. The icaA, rbf, sarA, and agr genes are involved in biofilm formation and bacterial dispersion and were detected in 96.3%, 40.0%, 74.5%, and 100% of S. aureus strains, respectively, and 70.9% of the strains formed a detectable biofilm. Interestingly, 67.2% of the strains contained the icaA, agr, spa, clfA, sdrC, sdrD, sdrE, seg, seh, and sei genes, suggesting that this gene combination is important for successful catheter colonization. The results of this study provide significant insight into the virulence gene make-up of catheter-colonizing S. aureus strains, and will assist in developing a more targeted treatment approach for persistent S. aureus biofilm contamination of medical devices.

Density,Sex Ratio,Size,Weight,and Recruitment of Plicopurpura pansa(Gastropoda:Muricidae)in Costa Chica,Guerrero,Mexico

Plicopurpura pansa is a cultural resource with economic importance since ink can be extracted from it to dye clothes. This snail inhabits the rocky intertidal zone of Mexican Pacific, is found attached to rocks in crevices and holes. The objective was to analyze the P. pansa population structure in Costa Chica of the State of Guerrero based in density, sex ratio, size and weight, besides to realize observations on mating and recruitment, for which quarterly sampling was made during an annual cycle in three beaches. The density was 4.83 snails/m2. Sex ratio ranged from 0.76:1.00 to 1.00: 0.96 (f:M). Average size was 20.91 mm. The best represented sizes in females were from 10.5 to 28.5 mm and in males 13.5 to 28.5 mm. Averaged weight was 1.99 g, females showed a coefficient of variation in weight of 96.53%. Recruitment occurred for the entire year. In December 2006 and September 2007 a larger frequency of recruits was shown. Sex ratio showed a 1:1 tendency. Snail population was characterized by being small sized and in high densities. Weight variations in females were found related to their reproductive cycle.

A single dose of caffeic acid phenethyl ester prevents initiation in a medium-term rat hepatocarcinogenesis model

AIM： To study of the effect of caffeic acid phenethyl ester （CAPE） on the initiation period in a medium-term assay of hepatocarcinogenesis. METHODS： Male Wistar rats were subjected to a carcinogenic treatment （CT） and sacrificed at 25^th d; altered hepatic foci （AHf） were generated efficiently. To a second group of rats a single 20 mg/kg doses of CAPE was given 12 h before initiation with CT and were sacrificed at 25^th d. We evaluated the expression of preneoplastic markers as Y-glutamyltranspeptidase （GGT） and glutathione S-transferase type pi protein （GSTp） by histochemistry, RT-PCR and Western blot analyses, respectively. We measured thiobarbituric acid reactive substances （TBARS） in homogenates of liver and used Unscheduled DNA Synthesis （UDS） assay by incorporation of [^3H] thymidine （^3HdT） in primary hepatocyte cultures （PHC）. RESULTS： At 25^th d after CT CAPE reduced the observed increase of GGT^＋AHf by 84% and liver expression ofggt mRNA by 100%. In case of the GSTp protein, the level was reduced by 90%. As indicative of oxidative stress generated by diethylnitrosamine （DEN） 12 h after its administration, we detected a 68% increase of TBARS. When CAPE was administered before DEN, it completely protected from liver TBARS induction. To have an indication of the sole effect of CAPE on initiation, two carcinogens were tested in a UDS assay in PHC, we used methyl-n-nitrosoguanidine as a direct carcinogen and DEN, as indirect carcinogen. In this assay, genotoxic damage caused by carcinogens was abolished at 5μM CAPE concentration. CONCLUSION： Our results demonstrated that CAPE possesses anti-genotoxic and antineoplastic capabilities, by an anti-oxidative and free-radical scavenging mechanism.

Innovate combination of sevoflurane dilution in dimethyl sulfoxide: A stability study by gas chromatography and nuclear magnetic resonance

To investigate physicochemical stability of sevofuranein dimethyl sulfoxide using gas chromatography with a fame ionization detector and nuclear magnetic reso-nance （NMR）.METHODSUndiluted sevoflurane, plus dilutions 1：2, 1：5, 1：10, 1：25, and 1：50 in dimethyl sulfoxide were prepared in a vertical laminar fow cabinet class Ⅱ type B and stored at different temperatures （23 ℃, 6 ℃, and -10 ℃） for 45 d. Sterile 1 mL polypropylene amber syringes to minimize light degradation, caps and needles were used. The presence of sevofurane and its degradation products in the samples was determined by gas chroma-tography with flame ionization detector （260 ℃, 40min）, and by 1H, 19f, and proton-decoupled 19f nuclearmagnetic *** The gas chromatography analysis showed sevofluraneand dimethyl sulfoxide （DMSO） retention times were 2.7and 13.0 min, respectively. Pure DMSO injection into thecolumn resulted in two additional peaks at 2.1 and 2.8min. The same sevofurane peak at 2.7 min was observedin all the dilutions at -10 ℃, 4 ℃ and 25 ℃. The NMRspectra showed signals consistent with the sevoflurane structure in all the dilutions at -10 ℃, 4 ℃ and 25 ℃. In the 1H spectrum, two signals corresponding to the sevoflurane molecule were observed at 5.12 and 4.16 parts per million （ppm5）. In the 19f-NMR spectrum, two signals were observed at -76.77 ppm and -157.13 ppm. In the 19f NMR CPD, two signals were observed at -76.77 ppm and -157.13 ppm. The first one showed a doublet （Jf-f = 3.1 Hz） which integrated by six fluorine nuclei from the hexafluoro-isopropyl group. The second signal was integrated by a fuorine atom and showed a septuplet （Jf-f = 3.1 Hz）.CONCLUSIONThis study shows that different concentrations ofsevofurane in dimethyl sulfoxide retain their chemicalcomposition after exposure to different temperaturesfor a period of 45 d.

Risk of Second Solid Malignancy among 1000 Kidney Transplanted Patients: A Single Center Study

In this study we focus on kidney-transplanted patients in order to investigate the correlation between immunodeficiency (ID) induced by immunosuppressive treatments and the incidence of solid multiple primary malignancies (MPM). Material of study: from 1980 to 2010 we followed up 1000 kidney-transplanted patients (637M, 363f). This group was homogeneous for age, immunology (e.g. miss-match index), number of rejection events and for immunosuppressive therapy. Results: Out of 1000 kidney-transplanted patients we observed 70 patients (53M, 17f) with cancer disease and in 22 of them a multiple cancer has been found. Most of multiple cancer were synchronous and the association between cancer and rejection episodes were not significant. Discussion: In general population 1 out of 9 patients with a cancer would develop a second neoplasia during the course of his life, so, it would be logical to conclude that, from a merely theoretical and statistical point of view, long term transplanted patients potentially have a higher risk to develop MPM. But our series and literature review did not confirm it, probably because these patients die before the appearance of a second primary malignancy. Conclusions: Despite many observations regarding different types of tumors/pre-cancerous lesions and their increased incidence in ID patients and despite the fact that immune suppression is a predisposing factor for the multicancer syndrome, at least theoretically, nowadays there are no significant statistical data favouring a correlation between ID and MPM in kidney transplanted patients.

The effect of chronic administration of the sigma agonist: 1,3-di-o-tolyl-guanidine (DTG), in neonatal ventral hippocampal lesioned (nVHL) rats, on the immobility by clamping and dorsal immobility, were investigated. The nVHL increases the duration of immobility by clamping, but does not affect the duration of the dorsal immobility. We found that DTG augments the duration of the dorsal immobility in the unlesioned rats, but does not modify the duration of immobility induced by clamping the neck. DTG also counteracts the increase in the duration of the immobility by clamping produced by nVHL. However, the increase in the duration of the dorsal immobility produced by DTG is counteracted by nVHL. These results are discussed with respect to the differential effect on the two immobility responses tested, suggesting that they are different forms of immobility mediated by different mechanisms although they behaviorally share common characteristics.
