Regenerative potential of mouse embryonic stem cell-derived PDGFRα+ cardiac lineage committed cells in infarcted myocardium

BACKGROUND Pluripotent stem cell-derived cardiomyocytes(CMs) have become one of the most attractive cellular resources for cell-based therapy to rescue damaged cardiac *** We investigated the regenerative potential of mouse embryonic stem cell(ESC)-derived platelet-derived growth factor receptor-α(PDGFRα)+ cardiac lineagecommitted cells(CLCs), which have a proliferative capacity but are in a morphologically and functionally immature state compared with differentiated *** induced mouse ESCs into PDGFRα+ CLCs and αMHC+ CMs using a combination of the small molecule cyclosporin A, the rho-associated coiled-coil kinase inhibitor Y27632, the antioxidant Trolox, and the ALK5 inhibitor *** implanted PDGFRα+ CLCs and differentiated αMHC+ CMs into a myocardial infarction(MI) murine model and performed functional analysis using transthoracic echocardiography(TTE) and histologic *** Compared with the untreated MI hearts, the anterior and septal regional wall motion and systolic functional parameters were notably and similarly improved in the MI hearts implanted with PDGFRα+ CLCs and αMHC+ CMs based on *** histologic analysis, the untreated MI hearts contained a thinner ventricular wall than did the controls, while the ventricular walls of MI hearts implanted with PDGFRα+ CLCs and αMHC+ CMs were similarly thicker compared with that of the untreated MI hearts. Furthermore, implanted PDGFRα+ CLCs aligned and integrated with host CMs and were mostly differentiated into α-actinin+ CMs,and they did not convert into CD31+ endothelial cells or αSMA+ mural *** PDGFRα+ CLCs from mouse ESCs exhibiting proliferative capacity showed a regenerative effect in infarcted myocardium. Therefore, mouse ESC-derived PDGFRα+ CLCs may represent a potential cellular resource for cardiac regeneration.

Thermal stability of bimodal grain structure in a cobalt-based superalloy subjected to high-temperature exposure

The present work investigates the thermal stability and mechanical properties of a Co-20 Cr-15 W-10 Ni(wt%) alloy with a bimodal grain(BG) *** BG structure consisting of fine grains(FGs) and coarse grains(CGs) is thermally stable under high-temperature exposure treatments of 760℃ for 100 h and 870℃ for 100-1000 *** size of both FGs and CGs remains no significant changes after thermal exposure *** microstructural stability is associated with the slow kinetics of grain growth and the pinning of *** thermal stability enables to maintain the BG structures,leading to the same mechanical properties as the sample without thermal exposure *** particular,the BG alloy samples after thermal exposure treatment exhibit superior mechanical properties of both high strength and high ductility compared to the unimodal grain(UG) structured *** BG structure of the alloy samples after thermal exposure is capable of avoiding severe loss of ductility and retaining high *** specifically,the ductility of the BG alloy samples after thermal exposure treatments of 870℃ for 500-1000 h is ten times higher(44.6% vs.3.5% and 52.6% vs.5.0%) than that of the UG *** finding in the present work may give new insights into high-temperature applications of the Co-20 Cr-15 W-10 Ni alloy and other metallic materials with a BG structure.

Cambrian Stem-group Cnidarians with a New Species from the Cambrian Series 3 of the Taebaeksan Basin, Korea

Five species, Lipopora lissa Jell and Jell, 1976, Lipopora daseia Jell and Jell, 1976, Tretocylichne perplexa Engeibretsen, 1993 from Australia, Cambroctoconus orientalis Park, woo, Lee, Lee, Lee, Han and Chough, 2011 from China, and Cambroctoconus kyrgyzstanicus Peel, 2014 from Kyrgyzstan, belonging to the Cambrian stem-group cnidarians have been documented in the fossil record. Cambroctoconus coreaensis sp. nov., interpreted here as a stem-group cnidarian, from the Seokgaejae section in the Daegi Formation, Taebaek Group （Cambrian Series 3）, Taebaeksan Basin, central-eastern Korean Peninsula, has a slender cup-shaped skeleton. A cladistic analysis produced 21 most parsimonious trees, which invariably placed the six stem-group cnidarians below the crown-group, but their relationships within the stem-group are unresolved. Nine out of the 21 trees suggest a monophyletic relationship for the Cambrian stem-group cnidarians, whereas in six other trees a monophyly of Cambroctoconus and Tretocylichne appeared as the sister-group to the crown-group cnidarians with Lipopora at the most basal branch. This result may reflect the fact that crown-group cnidarians evolved in the Precambrian, and suggests that the diversity of stem-group cnidarians was a result of an independent radiation in the Cambrian.

Effects of ranitidine for exercise induced gastric mucosal changes and bleeding

AIM： To evaluate the effect of ranitidine on gastric mucosal changes and on GI bleeding in long distance runhers. METHODS： Twenty-four long distance runners （M： 16, F： 8, age： 18.2＋ 1.5 years） participated in this study. A symptom questionnaire, stool hemoccult test, and upper gastrointestinal （GI） endoscopy were performed on the subjects prior to the study. The subjects took oral ranitidine （150 mg, b.i.d.） for two weeks. The upper GI endoscopy and stool Hemoccult tests were repeated after the treatment. RESULTS： Twenty-two of the 24 runners had at least one upper G1 mucosal lesion before the medication. The Endoscopic improvements were seen in eleven of the 14 cases of erosive gastritis and four of the 5 cases of esophagitis. Six subjects were Heine occult positive prior to the study, but only one was positive after the medication. CONCLUSION： Gastric mucosal lesions and GI bleeding in long distance runners seem to be associated to acidrelated factors mediated by the high level of regular running. Ranitidine seems to be and effective prophylaxis to prevent gastric mucosal lesions and GI bleeding.

Dear Editor Hepatocellular carcinoma(HCC)is among the most com-mon cancers worldwide,causing about 600,000 deaths annully[1].In HCC,stem cell-like characteristics,which drive early recurrence and therapy resistance,are major contributors to poor prognosis[2].In this current study,we integrated and analyzed gene expression data from human fetal liver cells and primary HCC tumors(n=1231)*** two clinically and biologically distinct hepatic stem cell(HS)subtypes,potential biomarkers associated with these subtypes,and a potential new therapeutic inter-vention for these subtypes.

基于多模光纤或多芯光纤的无透镜超细光纤内窥成像技术近些年获得了快速发展,有望成为下一代的极微创、高分辨率内窥显微镜。通过对相干入射光场的时空调控,该技术可克服多模光纤中模式色散或多芯光纤中相位畸变的影响,在无需光纤末端透镜或扫描器件的情况下实现高分辨率的聚焦、成像及相关应用。此外,在无透镜光纤内窥成像或图像传输等场景下,通过构建物理或深度学习模型,从光纤输出测量中也能实现物体信息重建。对相干光纤无透镜成像技术的发展进行综述,首先说明无透镜光纤成像的基础原理,并从主动波前调控和被动目标重建这两类角度阐述无透镜光纤成像方法,接着介绍一些先进光纤成像模态和技术,列举光纤成像相关应用,最后分析该领域所面临的挑战,总结并展望其进一步发展方向和应用前景。

Interleukin-1 receptor-associated kinase 4(IRAK4)is a pivotal enzyme in the Toll-like receptor(TLR)/MYD88 dependent signaling pathway,which is highly activated in rheumatoid arthritis tissues and activated B cell-like diffuse large B-cell lymphoma(ABC-DLBCL).Inflammatory responses followed by IRAK4 activation promote B-cell proliferation and aggressiveness of ***,proviral integration site for Moloney murine leukemia virus 1(PIM1)functions as an anti-apoptotic kinase in propagation of ABC-DLBCL with ibrutinib *** developed a dual IRAK4/PIM1 inhibitor KIC-0101 that potently suppresses the NF-κB pathway and proinflammatory cytokine induction in vitro and in *** rheumatoid arthritis mouse models,treatment with KIC-0101 significantly ameliorated cartilage damage and ***-0101 inhibited the nuclear translocation of NF-κB and activation of JAK/STAT pathway in *** addition,KIC-0101 exhibited an anti-tumor effect on ibrutinib-resistant cells by synergistic dual suppression of TLR/MYD88-mediated NF-κB pathway and PIM1 *** results suggest that KIC-0101 is a promising drug candidate for autoimmune diseases and ibrutinib-resistant B-cell lymphomas.

BACKGROUND Acral persistent papular mucinosis(APPM)is a rare idiopathic subtype of localized lichen *** date,there have been 40 APPM cases reported worldwide;however,only 7 cases have been reported in the Korean *** SUMMARY A 70-year-old man was referred to our hospital with a solitary pinkish nodule on the dorsum of his right *** the absence of symptoms,the patient wanted to know the exact diagnosis;thus,a biopsy was *** examination of a biopsy specimen obtained from the nodule on the dorsum of his hand revealed orthokeratotic hyperkeratosis with patchy parakeratosis,prominent hypergranulosis,and diffuse dissecting mucinous deposition between collagen bundles,along with some bland-looking spindle cells throughout the *** nodule was histologically diagnosed as an APPM,and an intralesional triamcinolone injection(2.5 mg/mL)was started every 2 *** three sessions of treatment,the patient showed marked *** To the best of our knowledge,this is the first case of a Korean APPM presenting as a solitary nodule that showed a marked response to triamcinolone intralesional *** it is a rare disease,we report this case to contribute to future research on the pathogenesis and treatment of APPM.

Optimal laboratory testing protocol for patients with acne taking oral isotretinoin

BACKGROUND Isotretinoin is a widely prescribed drug for various dermatological conditions,especially *** tests are routinely performed to monitor its side ***,the optimal testing schedule remains *** To evaluate the laboratory monitoring tests and schedules of patients with acne taking isotretinoin to determine the most efficient testing *** We retrospectively reviewed electronic medical records from the Dermatology Department of Dongguk University Ilsan Hospital from 2005 to 2020 for patients prescribed isotretinoin for acne who underwent monthly blood *** Aspartate aminotransferase(AST),alanine aminotransferase(ALT),and trigly-ceride(TG)levels significantly changed between 5 and 6 mo when the total prescription period and dose variables were considered *** high-density lipoprotein level also significantly changed between 5 and 6 ***,low-density lipoprotein(LDL)and total cholesterol levels significantly changed between 1 and 2 *** We recommend testing AST,ALT,and TG levels once every 5 to 6 *** also suggest testing LDL and total cholesterol levels during the first and second months.
