Based on the mass balance equations of solute transfer in the radial chroma-tog.aphic column,the theoretical expression to describe the column efficiency and shape of elu-tion profile is obtained under linear isotherm ***,the tendency for the variation of column efficiency and symmetry of peak profile is systematically *** results showed that in radial chromatog.aphy the relationship between the column efficiency and volumetric flow rate is similar with that relationship in axial chromatog.aphy;relatively hig. column efficiency still can be obtained under hig. flow rate in radial *** the increase of retention factor of solutes and injection time,the column efficiency decreases *** effect of column diameter and column leng.h on the column efficiency interfere with each *** is more advantag.ous to increase the column efficiency by applying.columns with larg.r column diameter and shorter column *** to the discussion of the effect of diffusion on the column efficiency,radial chromatog.aphy is proved to be suitable for the separation of samples with relatively hig. diffusion coefficient,which predicts its obvious advantag. in the preparative separation of samples such as proteins and DNA.

Based on the mechanism of chromatog.aphic retention (the relationship between the retention of solute and the mobile phase conditions) and method of resolution map, several methods of optimizing.multi-seg.ent linear g.adient elution conditions were proposed according.to the different separation requirements of various samples. These methods were verified using.literature data. Moreover, the advantag.s and disadvantag.s of these methods were compared. It was proved that the third method is a fast optimization method which is capable of separating.all the components with relatively hig. resolution.

An assimilation data set based on the g.DL MOM3 model and the NODC XBT data set is used to examine the circulation in the western tropical Pacific and its seasonal variations. The assimilated and observed velocities and transports of the mean circulation ag.ee well. Transports of the North Equatorial Current (NEC), Mindanao Current (MC), North Equatorial Countercurrent (NECC) west of 140°E and Kuroshio orig.n estimated with the assimilation data display the seasonal cycles, roug.ly strong.in boreal spring.and weak in autumn, with a little phase difference. The NECC transport also has a semi-annual fluctuation resulting.from the phase lag.between seasonal cycles of two tropical g.res’ recirculations. Strong.in summer during.the southeast monsoon period, the seasonal cycle of the Indonesian throug.flow (ITF) is somewhat different from those of its upstreams, the MC and New g.inea Coastal Current (Ng.C), implying.the monsoon’s impact on it.

Photoconvertible fluorescent proteins such as Kaede are routinely used for tracking.proteins, org.nelles, and whole cells. Kaede was the first identified photoconvertible fluorescent protein and has since become the most commonly used photoconvertible fluorescent protein in vertebrates. Kaede can be irreversibly converted from a g.een to a red fluo- rescent form upon UV/blue lig.t irradiation and fluorescence of each form can be isolated separately by appropriate filter sets. Spectral properties of the Kaede forms allow F6rster resonance energ. transfer （FRET） from the g.een form as donor to the red form as acceptor. As a sample containing.olig.merized Kaede-containing.proteins is exposed to UV or blue lig.t, FRET first increases as g.een Kaede is converted to red and then decreases as the g.een donor becomes depleted. Thus, FRET information is potentially obtained from a number of independent measurements taken as photoconversion proceeds. We demonstrate here the application of this approach to detect homo-ag.reg.tion and conformational dynamics of plant pro- tein constructs. Structural alterations of 2-cys peroxiredoxin-Kaede were successfully detected depending.on the redox state in living.plant cells. Photoconversion was performed g.adually and donor emission, acceptor emission, and FRET-derived sensitized acceptor emission were measured at each step of conversion. Since photoconvertible proteins have not been rou- tinely used in plants, two plasmids have been desig.ed to facilitate plant applications. The plasmids allow either transient expression of Kaede-containing.protein constructs in plant cells or g.teway cloning.and stable transformation of plants.

A hig. quality factor is preferred for a microresonator sensor to improve the sensitivity and resolution. In this paper we systematically investig.te the performance of the microcantilever in different resonance modes, which are the first three flexural modes, the first lateral mode, and the first and the second torsional modes. An aluminum nitride-based piezoelectric cantilever is fabricated and tested under controlled pressure from an ultra-hig. vacuum to a normal atmosphere, using.a custom-built vacuum chamber. From the experiment results, it can be seen that the torsional modes exhibit better quality factors than those of the flexural and lateral ones. Finally, an analytical model for the air damping.characteristics of the torsional mode cantilever is derived and verified by comparing.with experimental results.
