Chloroplasts and bacterial cells divide by binary fission. the key protein in this constriction division is FtsZ, a self-assembling GtPase similar to eukaryotic tubulin. In prokaryotes, FtsZ is almost always encoded by a single gene, whereas plants harbor several nuclear-encoded FtsZ homologs. In seed plants, these proteins group in two families and all are exclusively imported into plastids. In contrast, the basal land plant Physcomitrella patens, a moss, encodes a third FtsZ family with one member. this protein is dually targeted to the plastids and to the cytosol. Here, we report on the targeted gene disruption of all ftsZ genes in R patens. Subsequent analysis of single and double knockout mutants revealed a complex interaction of the different FtsZ isoforms not only in plastid division, but also in chloroplast shaping, cell patterning, plant development, and gravity sensing. these results support the concept of a plastoskeleton and its functional integration into the cytoskeleton, at least in the moss R patens.

A recent research article published in Science by Ritter et *** that endosomal sorting complexes required for transport(ESCRt)proteins mediate repair of lesions in the cell membrane caused by the pore-forming toxin perforin at sites of cytotoxic t cell engagement.1 Subsequent entry of granzymes and initiation of apoptosis of cancer cells is thus limited,leading to reduced sensitivity against t cell‑mediated cytotoxicity.1 Along these lines,this could constitute a new critical resistance mechanism for t cellbased treatment approaches.

Accurate metrology of extreme ultraviolet (EUV) photomask is a crucial task. In this paper, two different methods for reference EUV photomask metrology are compared. One is the critical dimension atomic force microscopy (CD-AFM). In the measurements, the contribution of its AFM tip geometry is usually the dominant error source, as measured AFM images are the dilated results of measured structures by the AFM tip geometry. to solve this problem, a bottom-up approach has been applied in calibrating the (effective) AFM tip geometry where the result is traceably calibrated to the lattice constant of silicon crystals. the other is transmission electron microscopy (tEM). For achieving measurement traceability, structure features are measured in pairs in tEM images;thus the distance between the structure pair calibrated by a metrological AFM in prior can be applied to determine the magnification of the tEM image. In this study, selected photomask structures are calibrated by the CD-AFM, and then sample prepared and measured by high-resolution tEM nearly at the same location. the results are then compared. Of six feature groups compared, the results agree well within the measurement uncertainty, indicating excellent performance of the developed methodology. this research supports the development of a photomask standard, which is applied as a “reference ruler” with improved low measurement uncertainty in photomask fabs.

We present a detailed comparison of two approaches, the use of a precalculated database and simulated annealing （SA）, for fitting the continuum spectral energy distribution （SED） of astrophysical objects whose appearance is dominated by surrounding dust. While pre-calculated databases are commonly used to model SED data, only a few studies to date employed SA due to its unclear accuracy and convergence time for this specific problem. From a methodological point of view, different approaches lead to different fitting quality, demand on computational resources and calculation time. We compare the fitting quality and computational costs of these two approaches for the task of SED fitting to provide a guide to the practitioner to find a compromise between desired accuracy and available resources. to reduce uncertainties inherent to real datasets, we introduce a reference model resembling a typical circumstellar system with 10 free parameters. We derive the SED of the reference model with our code MC3 D at 78 logarithmically distributed wavelengths in the range [0.3 μm, 1.3 mini and use this setup to simulate SEDs for the database and SA. Our result directly demonstrates the applicability of SA in the field of SED modeling, since the algorithm regularly finds better solutions to the optimization problem than a precalculated database. As both methods have advantages and shortcomings, a hybrid approach is preferable. While the database provides an approximate fit and overall probability distributions for all parameters deduced using Bayesian analysis, SA can be used to improve upon the results returned by the model grid.

We present modeling work on three young stellar objects that are promis-ing targets for future high-resolution observations to investigate circumstellar disk evolution. the currently available data comprise the spectral energy distribution from optical to millimeter wavelengths which allow constraining the structure of the cir-cumstellar disk using self-consistent radiative transfer models. the results suggest that the assumption of well-mixed dust and gas leads to overestimation of flux in the far-infrared. Observational and theoretical arguments suggest that an overall decrease in far-infrared excess can be explained by dust settling towards the midplane. A new disk model is hence employed to take the effect of dust sedimentation into account. the extended model satisfactorily reproduces all existing observations. the three tar-gets studied here therefore deserve follow-up observations to reveal the evolutionary state of their protoplanetary disks.

Particles occur in almost all processes in chemical and life sciences. the particle size and shape influence the process performance and product quality, and in turn they are influenced by the flow behavior of the particles during production. Monitoring and controlling such characteristics in multiphase systems to obtain sufficient qualities will greatly facilitate the achievement of reproducible and defined distributions. So far, obtaining this information inline has been challenging, because existing instruments lack measurement precision, being unable to process overlapping signals from different particle phases in highly concentrated multiphase systems. However, recent advances in photo-optics made it possible to monitor such features(particle size distribution(PSD), aspect ratio and particle concentration) with advanced image analysis(IA) in real-time. New analysis workflows as well as single feature extractions from the images using multiple image analysis algorithms allowed the precise real-time measurements of size, shape and concentration of particle collectives even separated from each other in three phase systems. the performances, advantages and drawbacks with other non-photo-optical methods for assessing the particle size distribution are compared and discussed.
