cyclooxygenases are encoded by cOX-1 and cOX-2. They share over sixty percent sequence identity in human and are similar to each other in their crystallographic structures. One major difference in the primary structure of these two isozymes is the presence of eight amino acids in the amino-terminal region of cOX-1 that are not present in cOX-2. The function of this amino acid sequence is unknown. In this study, a human cOX-1 mutant （Δ7aa） with this sequence removed was studied in parallel with cOX-1. Signal peptide cleavage, N-linked glycosylation, protein expression, distribution and dimerization were not affected by the mutation. The mutant was enzymati-cally active and showed the same sensitivity toward aspirin. The KM for the enzyme remained the same as cOX-1. However, the Vmax of the cOX-1 mutant decreased by 3.3-fold. We conclude that the cOX-1 specific amino-terminal sequence has a subtle but detectable effect on cOX-1 catalysis.

Phosphine-catalyzed [4＋2] annulations of 2-（acetoxymethyl）buta-2,3-dienoates with α-aminonitriles have been developed, in which a-aminonitriles serve as c, N-bisnucleophilic reaction partners and 2-（acetoxymethyl）buta- 2,3-dienoates as ＂c4 synthons＂ respectively. A number of a-aminonitriles could be successfully applied to giving multifunctional desired products using PPh3 as catalyst. This method provides a facile entry to access polysubstituted tetrahydropyridines bearing quaternary carbon centers. The possible reaction mechanism was also proposed.

Purpose A portable fast-neutron imaging system is being developed to provide complementary information to field X-ray *** include inspection of vehicles and infrastructure for corrosion,measurement of material levels in containers,and inspection of munitions and suspicious *** fast-neuron imaging generally provides lower imaging resolution compared to X-rays,fast-neutron interaction cross-sections have a weak dependence on material *** enables imaging of low-Z materials inside high-Z ***,we discuss the limitations and current improvements in fast-neuron *** Limitations in portable fast-neutron imaging systems include low D-T neutron generator output,low light pro-duction in ZnS(cu)imaging scintillators,low resolution due to scintillator thickness and D-T spot size,and digital-panel darknoise that varies in time and position and that can be 100×larger than the neutron *** have made improvements in these areas through development of a segmented high light yield scintillator,panel noise mitigation techniques,and testing of new high-output,small spot size D-T neutron *** The segmented high light yield fast-neutron scintillator demonstrated 5×increase in light compared to ZnS(cu).An additional 2×improvement in signal-to-noise was demonstrated with panel-noise mitigation *** McNP calculations also show good agreement with neutron imaging results conclusions We have demonstrated improvements in fast-neutron imaging through development of a segmented high light yield neutron scintillator,mitigation of digital panel noise,and preliminary testing with new high-output,small spot size D-T neutron *** have also demonstrated good results modeling fast-neutron images and scatter effects using McNP.

BAcKGROUND:complications from gallstones and laparoscopic cholecystectomy can be serious and fatal if there is a delay in recognition and treatment. We aim to present two unusual, life threatening vascular complications as a result of gallstones and laparoscopic cholecystectomy. Their management is highlighted with a brief review of literature. METHODS:Data for the article were gathered from clinical case note review. Radiology database was used for images. A brief literature review was undertaken using Pubmed search. The keyword used included hemobilia, pseudoaneurysm, arterio-biliary fistula and laparoscopic cholecystectomy. RESULTS:The article highlights two individual case reports. The first case constitutes an 81-year woman who had cystic arterial erosion causing hematemesis, while the second patient was a 57-year man who presented with hemobilia from a pseudoaneurysm of right hepatic artery (RHA) following laparoscopic cholecystectomy. cystic arterial erosion was treated with subtotal cholecystectomy with duodenal defect closure while the pseudoaneurysm underwent radiological intervention. cONcLUSIONS:cystic artery erosion and pseudoaneurysm causing arteriobiliary fistula are rare vascular complications related to the biliary tree. A high index of suspicion and timely intervention is important. Trauma to arteries should be avoided during laparoscopic cholecystectomy.

Background: Increasing evidence is demonstrating thatinfants born early on during the term period are at increasedrisk of morbidity compared with infants born closer to acomplete 40 week gestational pregnancy. The purpose of thisstudy was to compare early term [gestation age (GA): 37-376/7 weeks] neonatal outcomes with those of other full termneonatal intensive care unit (NIcU) ***: Retrospective chart review of all term infantsadmitted to the NIcU at New York University LangoneMedical center over a 17 month period. Subjects weregrouped and analyzed according to their GA at birth: 1)early term infants (GA between 37 0/7 to 37 6/7 weeks) and2) other term infants (38 0/7 weeks and older).Results: Early term infants were more likely to requireNIcU care than other term infants [relative risk: 1.42,95% confidence interval (cI)=1.07-1.88), P=0.01]. In theNIcU, they are more likely to manifest respiratory distresssyndrome [odds ratio (OR)=5.7, 95% cI=1.6-19.8, Pcemia (OR=4.6, 95% cI=2.0-10.4, Pctive cesarean section than other term neonates(OR=4.1, 95% cI=2.0-8.5, Pconclusions: Being born at early term is associatedwith increased risk of respiratory disease and hypoglycemiarequiring neonatal intensive care. Further efforts directedat decreasing early term deliveries may be warranted.

cellulose and hemicelluloses are the most prevalent sources of carbon in nature. currently many approaches employ micro-organisms and their enzyme products to degrade plant feedstocks for production of bioenergy. Scarab larvae are one such model. They consume celluloses from a variety of sources including plant roots, soil organic matter and decaying wood, and are able to extract nutrients and energy from these sources. In this paper, we review the physicochemical properties of the scarab larval gut, the diversity and digestive role that microflora play in the scarab gut and discuss the potential for applying these digestive processes in bioreactors for improving bio-fuel production. Scarab larvae are characterised by their highly alkaline midgut which is dominated by serine proteinase enzymes, and a modified hindgut which harbors the majority of the intestinal microbiota under anaerobic conditions. Evidence suggests that digestion of recalcitrant organic matter in scarab larvae likely results from a combination of endogenous gut proteinases and cellulolytic enzymes produced by symbiotic micro-organisms. Most of the easily digestible proteins are mobilized and absorbed in the midgut by endogenous proteinases. The hindgut contents of scarab larvae are characterized by high concentrations of volatile fatty acids, the presence of fermenting bacteria, and typical anaerobic activities, such as methanogenesis. The hindgut typically contains a wide diversity of micro-organisms, some of which appear to be obligate symbionts with cellulolytic potential. As a result, the scarab larval gut can be regarded as a small bioreactor resembling the rumen of sheep or cattle, where solid food particles composed of cellulose, hemicellulose, pectin and polysaccharides are degraded through enzymatic and fermentation processes. A magnesium alloy containing essential,non-toxic,biodegradable elements such as ca and Zn has been fabricated using a novel twin-roll casting process(TRc).Microstructure,mechanical properties,in vivo corrosion and biocompatibility have been assessed and compared to the properties of the rare earth(RE)element containing WE43 *** Mg-0.5 wt% Zn-0.5 wt% ca exhibited fine grains with an average grain size ranging from 70 to 150μ*** properties of a TRc Mg-0.5Zn-0.5ca alloy showed an ultimate tensile strength of 220 MPa and ductility of 9.3%.The TRc Mg-0.5Zn-0.5ca alloy showed a degradation rate of 0.51±0.07 mm/y similar to that of the WE43 alloy(0.47±0.09 mm/y)in the rat model after 1 week of *** week 4 the biodegradation rates of both alloys studied were lowered and stabilized with fewer gas pockets around the *** histological analysis shows that both WE43 and TRc Mg-0.5Zn-0.5ca alloy triggered comparable tissue healing responses at respective times of *** presence of more organized scarring tissue around the TRc Mg-0.5Zn-0.5ca alloys suggests that the biodegradation of the RE-free alloy may be more conducive to the tissue proliferation and remodelling process.
