cDNA cloning,sequence analysis,and recombinant expression of akitonin beta,a C-type lectin-like protein from Agkistrodon acutus

AIM: To clone the cDNA of a new member of snake venom C-type lectin-like proteins, to study its structure- function relationships and to achieve its recombinant production. METHODS: PCR primers were designed based on the homology and cDNA was amplified by RT-PCR using total RNA from snake venom gland as the template. The PCR products were cloned into the plasmid pGEM-T and sequenced. The deduced protein sequence was analyzed with some bioinformatic programs. A recombinant expression plasmid was constructed using pBAD- TOPO as vector and transformed into *** TOP10 competent cells. RESULTS: A novel cDNA sequence encod- ing akitonin β was found and accepted by GenBank (accession number AF387100). Akitonin β consists of a typical carbohydrate recognition domain (CRD) of C-type lectins, and it is homologous with other snake venom C-type lectin-like proteins. It was predicted to be a platelet antagonist. Upon induction with arabinose rAkitonin β express- ing in E coli was achieved at a high level (superior to 150 mg/L). The recombinant fusion protein exhibited inhibitory activities on rat platelet aggregation in vitro. CONCLUSION: A new member of snake venom C-type lectin-like proteins was discovered and characterized, and an efficient recombinant expression system was estab- lished for its production.

Effects of Nitrogen Doping on Microstructures and Irradiation Resistance of Ti-Zr-Nb-V-Mo Refractory High-Entropy Alloy

Interstitial strengthening with nitrogen(N)is one of the effective ways to improve the mechanical properties of HEAs,but the effects of N on the microstructures and mechanical properties of the irradiated HEAs have not been studied ***,the microstructures and mechanical properties of N-free and N-doped Ti_(2)ZrNbV_(0.5)Mo_(0.2)HEAs before and after He irradiation were *** results showed that the solid solution strengthening caused by interstitial N improved the yield strength at room temperature and 1023 K without significantly reducing plasticity.N doping significantly promoted the growth,aggregation and wider spatial distribution of He bubbles by enhancing the mobility of He atoms/He-vacancy complexes,with the average size of He bubbles increasing from 10.4 nm in N-free HEA to 31.0 nm in N-doped *** addition,N-doped HEA showed a much higher irradiation hardness increment and hardening fraction than N-free *** to conventional materials doped with N,the introduction of N into Ti_(2)ZrNbV_(0.5)Mo_(0.2)HEA had adverse effects on its resistance to He bubble growth and irradiation *** results of this study indicated that N doping may not improve the irradiation resistance of HEAs.

TLR3 Ligand-Induced Accumulation of Activated Splenic Natural Killer Cells into Liver

It has been revealed that poly I：C is a potent stimulator for NK cells, which can induce NK cell rapid activation and preferential accumulation into liver. However, the process mediating the influx of NK cells remains obscure. In this study, we found that poly I：C administration increased the portion and absolute number of NK cells in liver, but largely decreased those in spleen. There were no obvious changes of these lymphocytes in other immune organs. The results from splenic adoptive transfer and splenectomy showed that the recruited spleen NK cells contributed to the accumulation of NK cells in liver, and this process was regulated by the production of chemokines and the presence of T cells. This investigation will help to understand the enhanced immune cell recruitment in liver upon viral infection. Cellular ＆ Molecular Immunology. 2005;2（6）：449-453.

Syntheses of amphiphilic biodegradable copolymers of poly(ethyl ethylene phosphate) and poly(3-hydroxybutyrate) for drug delivery

Biodegradable and amphiphilic triblock copolymers poly(ethyl ethylene phosphate)-poly(3-hydroxybutyrate)-poly(ethyl ethylene phosphate) (PEEP-b-PHB-b-PEEP) have been successfully synthesized through ring-opening polymerization. The structures are confirmed by gel permeation chromatography and NMR analyses. Crystallization investigated by X-ray diffraction reveals that the block copolymer with higher content of poly(ethyl ethylene phosphate) (PEEP) is more amorphous, showing decreased crystallizability. The obtained copolymers self-assemble into biodegradable nanoparticles with a coreshell micellar structure in aqueous solution, verified by the probe-based fluorescence measurements and transmission electronic microscopy (TEM) observation. The hydrophobic poly(3-hydroxybutyrate) (PHB) block serves as the core of the micelles and the micelles are stabilized by the hydrophilic PEEP block. The size and size distribution are related to the compositions of the copolymers. Paclitaxel (PTX) has been encapsulated into the micelles as a model drug and a sustained drug release from the micelles is observed. MTT assay also demonstrates that the block copolymers are biocompatible, rendering these copolymers attractive for drug delivery.

Alterations in circadian rhythms of melaton in and cortisol in patients with bronchial asthma

AIM: To investigate the possible relationships between alterations in circadian rhythm of melatonin, cortisol andbronchial asthma. METHODS: Salivary melatonin and cortisol were measured simultaneously by radioimmunoas-say in 10 mild intermittent or persistent patients, 11 moderate-to-severe persistent asthma patients, and 15 controlsubjects. Twelve salivary samples were collected in a series during a 24-h period in each subject. RESULTS: Theresults showed overall lower levels of salivary melatonin in asthma patients compared with control subject(P24-h mean level of salivary cortisol was greatly lower and the acrophase was markedly delayedin patients with mild intermittent or persistent asthma (P<0.01) and moderate-to-severe persistent asthma (P<0.05, P<0.01) compared with control subject. CONCLUSION: Disordered circadian rhythms of salivary melato-nin and cortisol were found in asthma patients, which may be involved in the pathogenesis of bronchial asthma.

Fast parallel algorithms for discrete Gabor expansion and transform based on multirate filtering

The Gabor transform has long been recognized as a very useful tool for the joint time and frequency analysis in signal *** real time applications,however,were limited due to the high computational complexity of the Gabor transform *** this paper,some novel and fast parallel algorithms for the finite discrete Gabor expansion and transform are presented based on multirate *** analysis filter bank is designed for the finite discrete Gabor transform(DGT)and a synthesis filter bank is designed for the finite discrete Gabor expansion(DGE).Each of the parallel channels in the two filter banks has a unified structure and can apply the FFT and the IFFT to reduce its computational *** computational complexity of each parallel channel does not change as the oversampling rate *** fact,it is very low and depends only on the length of the input discrete signal and the number of the Gabor frequency sampling *** computational complexity of the proposed parallel algorithms is analyzed and compared with that of the major existing parallel algorithms for the finite DGT and *** results indicate that the proposed parallel algorithms for the finite DGT and DGE based on multirate filtering are very attractive for real time signal processing.

The effect of Dzialoshiski-Moriya (DM) interaction on the violation of Bell inequality for thermal states of interacting qubits via a two-qubit XXZ spin chain is *** results imply that the DM interaction and anisotropy taking a large positive value can enhance the Bell violation and improve the threshold temperatures of *** the comparison between the Bell violation and thermal entanglement,we find that the threshold temperatures of thermal entanglement are higher than those of the Bell *** implies that some states are entangled but the Bell inequality is not violated.

This investigation developed a new method for determining metal complex's property containing the stepwise real absorptivity(epsilon) and stability constant (K-m). The correction equation of the multi-wavelength spectral absorption was established for the simultaneous determination of various complexes to give high accuracy for trace analysis. This method was more acceptable in theory and simpler in operation than the classical methods.

Antisense-Induced Blockade of GATA-3 Expression Could Inhibit Th2 Excursion of Tumor Cells in vitro and in vivo

Previous studies have shown that tumor cells predominantly express Th2 type cytokines and transcription factors. GATA-3, as a Th2-specific transcription factor, plays a central role in positive-regulating Th2 development. So whether the expression of GATA-3 in tumor cells has any effect on tumor development is a question of interest. In the present study, we inhibited the expression of GATA-3 in tumor cells through antisense RNA blockade technique, and observed its effects on tumor in vitro and in vivo. Our results showed that antisense GATA-3 treatment could inhibit the expression of TNF-α and Th2 cytokines in tumor cells, and antisense-induced blockade of GATA-3 could also depress tumor growth in tumor-bearing mice. We suggest that the ratio of T-bet/GATA-3 can be evaluated as a more important marker of the status of Thl/Th2 type. And our results might provide some evidence about the molecular regulatory mechanisms in tumor cell development. Cellular ＆ Molecular Immunology. 2005 ;2（3 ）： 189-196.

Dynamic behavior of micro-diaphragms and its characterized description

Diaphragm structures with micron scale play a significant role in microtransducers and micro-nano devices, and the performance of these devices depends mainly on the dynamic behaviour of diaphragms. Micro-diaphragms are treated commonly as membranes and in some cases as plates or plates in tension (called TD plates for short), but they also show in many cases the behaviour of plates in tension and supported by air spring (called TDK plates for short). Therefore, it is necessary to perform systematic research on the dynamic behaviour of micro-diaphragms, and establish a characterized mathematical description. This paper focuses on the TDK plates since they possess universality, gives the corresponding basic equations, and then derives analytical solutions of TDK circular plates under clamped and simply supported boundary conditions. This paper also gives a 3D plot representation of characteristic curved surfaces, revealing the transition from the TDK and TD plate to the pure plate or pure membrane behaviour; and further uses the value φ to determine the property of diaphragms. Its two extreme cases, i.e. φ = 0 and φ = ∞ , correspond to pure plate or pure membrane, respectively. Thus, membrane, plate and TD plate can be treated as special cases of TDK plate. In addition, this paper reveals that the presence of air-spring not only enhances the restoring force of diaphragm such that increases its natural frequencies, but also results in the resonance of a dynamic system consisting of diaphragm and air-spring. These analytical and computational results are significant for the understanding of the operation mechanism of capacitive microtransducers and their optimized design.
