Flashing light in sponges through their siliceous fiber network:A new strategy of “neuronal transmission” in animals

Agrobacterium tumefaciens mediated plant transformation is a versatile tool for plant genetic engineering following its discovery nearly half a century *** modifications were made in its application to increase efficiency,especially in the recalcitrant major cereals *** breakthroughs in transformation efficiency continue its role as a mainstream technique in cRISPR/cas-based genome editing and gene *** modifications led to higher transformation frequency and lower but more stable transgene copies with the capability to revolutionize modern *** this review,we provide a brief overview of the history of Agrobacterium-mediated plant transformation and focus on the most recent progress to improve the system in both the Agrobacterium and the host recipient.A promising future for transformation in biotechnology and agriculture is predicted.

There is limited knowledge about microbial communities and their ecological functions in karst caves with high cO_(2)***,we studied the microbial community compositions and functions in Shuiming cave( "SMc",cO_(2)concentration 3 303 ppm) and Xueyu cave( "XYc",cO_(2)concentration 8 753 ppm) using Illumina MiSeq high-throughput sequencing in combination with BIOLOG *** results showed that Proteobacteria,Actinobacteria and Bacteroidetes were dominant phyla in these two caves,and Thaumarchaeota was the most abundant in the rock wall samples of *** microbial diversity in the water samples decreased with increasing HcO_(3)^(-)concentration,and it was higher in XYc than that in *** microbial community structures in the sediment and rock wall samples were quite different between the two *** concentrations of cO_(2)can reduce the microbial diversity on the rock walls in karst caves,probably through changing microbial preference for different types of carbon sources and decreasing the microbial utilization rate of carbon *** results expanded our understanding of microbial community and its response to environments in karst caves with high cO_(2).

Sponges are the oldest and the simplest but not primitive multicellular animals. They represent the earliest evolutionary metazoan phylum still extant. It was a long and painful scientific process to posi-tion the most enigmatic and mysterious metazoan,the Porifera,into their correct phylogenetic place among the eukaryotes in general and multicellular animals in particular. As living fossils,sponges provide the best evidence for the early evolution of Metazoa. More recently,interest has been focused on the bionic applications of sponges' siliceous spicules,after the discovery of their unique structure and high fiber performance. In this review,the emergence of sponges,evolutionary novelties found in sponges,and the phylogenetic position of sponges in early metazoan evolution are highlighted. In ad-dition,the present state of knowledge on silicatein-mediated "biosilica" formation in marine sponges,including the involvement of other molecules in silica metabolism and their potential application in nanobiotechnology and medicine,is given.

Deep-sea minerals in polymetallic nodules and seamount co-rich crusts are not only formed by mineralization but also by biologically driven processes involving microorganisms （biomineralization）. Within the polymetallic nodules, free-living and biofilm-forming bacteria provide the matrix for manganese deposition, and in seamount co-rich crusts, coccolithophores represent the dominant organisms that act as bio-seeds for an initial manganese deposition. These （bio）minerals are economically important： manganese is an important alloying component and cobalt forms part of special steels in addition to being used, along with other rare metals, in plasma screens, hard-disk magnets and hybrid car motors. Recent progress in our understanding of the participation of the organic matrices in the enrichment of these metals might provide the basis for feasibility studies of biotechnological applications.

Sterile signals generate weaker and delayed macrophage NLRP3 inflammasome responses relative to microbial signals

Inflammation is the host response to microbial infection or sterile injury that aims to eliminate the insult, repair the tissue and restore homeostasis. Macrophages and the NLRP3 inflammasome are key sentinels for both types of insult. Although it is well established that the NLRP3 inflammasome is activated by microbial products and molecules released during sterile injury, it is unclear whether the responses elicited by these different types of signals are distinct. In this study, we used lipopolysaccharide and tumor necrosis factor as prototypical microbial and sterile signal 1 stimuli, respectively, to prime the NLRP3 inflammasome. We then used the bacterial toxin nigericin and a common product released from necrotic cells, ATP, as prototypical microbial and sterile signal 2 stimuli, respectively, to trigger the assembly of the NLRP3 inflammasome complex in mouse and human macrophages. We found that NLRP3 inflammasome responses were weakest when both signal 1 and signal 2 were sterile, but responses were faster and stronger when at least one of the two signals was microbial. Ultimately, the most rapid and potent responses were elicited when both signals were microbial. Together, these data suggest that microbial versus sterile signals are distinct, both kinetically and in magnitude, in their ability to generate inflammasome-dependent responses. This hierarchy of NLRP3 responses to sterile versus microbial stimuli likely reflects the urgent need for the immune system to respond rapidly to the presence of infection to halt pathogen dissemination.

The physiological polyphosphate as a healing biomaterial for chronic wounds:crucial roles of its antibacterial and unique metabolic energy supplying properties

In the last 15 years biomineralization, in particular biosilicification （i.e., the formation of biogenic silica, SiO2）, has become an exciting source of inspiration for the development of novel bionic approaches, following ＂Nature as model＂. Among the silica forming organisms there are the sponges that have the unique property to catalyze their silica skeletons by a specific enzyme termed silicatein. In the present review we summarize the present state of knowledge on silicatein-mediated ＂biosilica＂ formation in marine sponges, the involvement of further molecules in silica metabolism and their potential application in biomedicine. Recent advancements in the production of bone replacement material and in the potential use as a component in the treatment of osteoporosis are highlighted.
