Identification, Isolation and Structure Confirmation of Forced Degradation Products of Sofosbuvir

A new stability indicating reverse phase chromatographic method was developed for the analysis of Heptisis C Drug Sofosbuvir. The developed UPLC method was superior in technology to conventional RP-HPLC with respect to resolution, speed, solvent consumption and analysis cost. Sofosbuvir was subjected to the thermal, hydrolytic, oxidative, and photolytic degradation, according to ICH guidelines. The drug depicted degradation in acidic, basic and oxidative conditions and it was stable to other stress conditions (thermal and photolytic). Identified degradation products were isolated using mass supported auto purification system and characterized by NMR techniques (1H NMR, D2O Exchange, 13C NMR, 31P NMR, 19F NMR, HSQC and HMBC) and HRMS experiments. Isolated acid degradation impurity was showing molecular weight of 416.08, molecular formula C16H18FN2O8P and its name as (R)-((2R, 3R, 4R, 5R)-5-(2,4-dioxo-3,4-dihydropyrimidin-1(2H)-yl)-4-fluoro-3-hydroxy-4-methyltetrahydrofuran-2-yl)methyl phenyl hydrogen phosphate. Isolated base degradation impurity-A was showing molecular weight of 453.13, molecular formula C16H25FN3O9P and its name as (S)-isopropyl 2-((R)-(((2R, 3R, 4R, 5R)-5-(2, 4-dioxo-3,4-dihydropyrimidin-1(2H)-yl)-4-fluoro-3-hydroxy-4-methyltetrahydrofuran-2-yl)methoxy)(hydroxy)phosphorylamino)propanoate. Isolated base degradation impurity-B was showing molecular weight of 411.08, molecular formula C13H19FN3O9 P and its name as (S)-2-((R)-(((2R, 3R, 4R, 5R)-5-(2,4-dioxo-3,4-dihydropyrimidin-1(2H)-yl)-4-fluoro-3-hydroxy-4-methyltetrahydrofuran-2-yl)methox-y)(hydroxy)phos-phorylamino)propanoic acid. AIM： To investigate the effects of mangiferin on gas- trointestinal transit （GIT） in normal and constipated mice, together with the possible ***： Intragastrically-administered charcoal mealwas used to measure GIT in overnight starved Swiss mice. In the first experiments, mangiferin （3 mg/kg, 10 mg/kg, 30 mg/kg, and 100 mg/kg, po） or tegaserod （1 mg/kg, ip） were administered 30 min before the char- coal meal to study their effects on normal transit. In the second series, mangiferin （30 mg/kg） was tested on delayed GIT induced by several different pharma- cological agonists （morphine, clonidine, capsaicin） or antagonists （ondansetron, verapamil, and atropine） whereas in the third series, mangiferin （30 mg/kg, 100 mg/kg and 300 mg/kg） or tegaserod （1 mg/kg） were tested on 6 h fecal pellets outputted by freely fed mice. The ratio of wet to dry weight was calculated and used as a marker of fecal water content. RESULTS： Mangiferin administered orally significantly （P 〈 0.05） accelerated GIT at 30 mg/kg and 100 mg/kg （89% and 93%, respectively）, similarly to 5-hydroxytrypta- mine4 （5-H%） agonist tegaserod （81%） when compared to vehicle-treated control （63%）. Co-administered man- giferin （30 mg/kg） totally reversed the inhibitory effect of opioid agonist morphine, 5-HT3-receptor antagonist ondansetron and transient receptor potential vanilloid-1 receptor agonist capsaicin on GIT, but only to a partial extent with the GIT-delay induced by ~2-adrenoceptor agonist clonidine, and calcium antagonist verapamil. However, co-administered atropine completely blocked the stimulant effect of mangiferin on GIT, suggesting the involvement of muscarinic acetylcholine receptor activation. Although mangiferin significantly enhanced the 6 h fecal output at higher doses （245.5±10.43 mg vs 161.9±10.82 mg and 227.1±20.11 mg vs 161.9±10.82 mg of vehicle-treated control, at 30 and 100 mg/ kg, P 〈 0.05, respectively）, the effect of tegaserod was more potent （297.4±7.42 mg vs 161.9±1

Micro-scale Dispersion of Air Pollutants over an Urban Setup in a Coastal Region

The dispersion is mainly governed by wind field and depends on the planetary boundary layer (PBL) dynamics. Accurate representation of the meteorological weather fields would improve the dispersion assessments. In urban areas representation of wind around the obstacles is not possible for the pollution dispersion studies using Gaussian based modeling studies. It is widely accepted that computational fluid dynamics (CFD) tools would provide reasonably good solution to produce the wind fields around the complex structures and other land scale elements. By keeping in view of the requirement for the micro-scale dispersion, a commercial CFD model PANACHE with PANEPR developed by Fluidyn is implemented to study the micro-scale dispersion of air pollution over an urban setup at Indira Gandhi Centre for Atomic Research (IGCAR), Kalpakkam a coastal station in the east coast of India under stable atmospheric conditions. Meso-scale module of the PANACHE model is integrated with the data generated at the site by IGCAR under RRE (Round Robin Exercise) program to develop the flow fields. Using this flow fields, CFD model is integrated to study the micro-scale dispersion. Various pollution dispersion scenarios are developed using hypothetical emission inventory during stably stratified conditions to understand the micro-scale dispersion over different locations of coastal urban set up in the IGCAR region of Kalpakkam.

Altered Serum Lipids in the Cases of Head and Neck Cancer Associated with the Habit of Tobacco Consumption

Alterations in serum lipid profile patterns have long been associated with malignancies, and their role remains controversial with respect to head and neck cancer. Due to an increased rate of neoplastic cell multiplication and reduced supply, there is increased utilization of lipids causing Hypolipidemia. Adding to this, tobacco contains carcinogens capable of damaging the cell membrane components including lipids resulting in further hypolipidemia. Thus the purpose of the present case control study is to discuss the alterations in plasma lipid profile in head and neck cancer patients in association with tobacco consumption. This hospital based study includes 80 cases of head and neck cancer patients and 50 controls. Plasma Lipids included are total cholesterol (TC), high density lipoprotein cholesterol (HDL), low density lipoprotein cholesterol (LDL), very low density lipoprotein cholesterol (LDL), triglycerides (TG). Student’s “t test” was applied to the data acquired. Values of P 0.05 were considered statistically insignificant.

Derivation of Specific Velocity of Body Moving under Gravity with Zero Total Energy

Mathematical solutions predict abstract conditions that indicate limits or bounds for physical processes. Generally, experimental verifications and physical observations on physical processes validate the mathematical predictions. Sometimes these predictions lead to new theories and concepts that form basis of better understanding of the natural processes. Gravitational interactions between bodies are natural physical processes. A smaller body moves under the influence of gravity, due to the gravitational effect of another large body. Newton’s classical gravitational theory addresses the interactions at low velocities. Einstein’s general relativity provides firm basis for gravitational interactions. Observations over past 100 years prove the mathematical precision and predictions of general relativity. Einstein’s special relativity forms the foundation of quantum physics. In this paper, the author applies concepts of special relativity to classical two body Newtonian gravitational problem. The study predicts a new mathematically viable condition that when a body moves at a specific velocity derived in this paper, the total energy of the moving body is zero. The specific velocity is a constant. At velocities far less than specific velocity, the total energy is negative and is equal to classical value of half the potential energy. At velocities, greater the specific velocity the total energy is positive. The specific velocity condition also enables determination of specific mass of gravitating body, as well as the specific distance of the moving body from gravitating body, at which the total energy of moving body is zero.

Development of Ultra Fast Liquid Chromatography (UFLC) Method for Fluorescence Detection of Domperidone in Human Serum and Application to Pharmacokinetic Study

A simple and sensitive fluorescence detection of domperidone by ultra fast liquid chromatographic method was developed and validated in human serum. For the evaluation of new drug delivery systems, conducting of pharmacokinetic studies in human volunteers is essential for approval to marketing after preclinical evaluation in animal models. The present method consists of protein precipitation, extraction of analytes from human serum into dichloromethane and separation using reversed-phase C18 column. Propranolol hydrochloride was used as an internal standard and the eluent was monitored by fluorescence detector at excitation 282 and emission 328 nm. The mobile phase used was 62:38 ratio of 10 mM phosphate buffer pH adjusted to 3.1 with OPA and methanol at a flow rate of 1 mL·min-1. The method was evaluated for assay, LLOD, LLOQ, recovery and stability studies. The retention times for domperidone and propranolol hydrochloride were found to be 6.36 and 7.94 minutes respectively. The intraday and inter-day coefficient of variation and percent error values of assay method were less than 5%;mean recovery was more than 96% for each analyte and the method was found to be precise, accurate and specific during study. The method was successfully applied for pharmacokinetic study of immediate and controlled release bioadhesive hot melt extruded buccal patches of domperidone after buccal administration to healthy human volunteers. The Cmax, Tmax, and AUC0–24 of domperidone from immediate and controlled release buccal patches were found to be 129.7 ng·mL-1, 1.5 h, 455.1 ng·h·mL-1 and 145.7 ng·mL-1, 5.25 h, 911.0 ng·h·mL-1 respectively. A simple, sensitive and reliable method for the fluorescence determination of domperidone in human serum by UFLC method was developed and validated.

α,β-amyrin, a natural triterpenoid ameliorates L-arginine-induced acute pancreatitis in rats

AIM: To study the benef icial effects of triterpene α,β-amyrin and the underlying mechanisms in an experimental pancreatitis model. METHODS: Acute pancreatitis was induced in five groups of rats (n = 8) by L-arginine (2 × 2.5 g/kg, intraperitoneal, 1 h apart) and 1 h later, they received a single oral dose of α,β-amyrin (10, 30 and 100 mg/kg),methylprednisolone (30 mg/kg) and vehicle (3% Tween 80). A saline (0.9% NaCl) treated group served as a normal control. Efficacy was assessed at 24 h by determination of serum levels of amylase, lipase and proinflammatory cytokines [tumor necrosis factor (TNF)-α and interleukin (IL)-6], pancreatic myeloperoxidase (MPO) activity, lipid peroxidation [thiobarbituric acid reactive substances (TBARS)], nitrate/nitrite levels, and the wet weight/body weight ratio. Tissue histology and the immunoreactivity for TNF-α and inducible nitric oxide synthetase (iNOS) were performed. RESULTS: α,β-amyrin and methylprednisolone treatments significantly (P < 0.05) attenuated the L-arginine-induced increases in pancreatic wet weight/body weight ratio, and decreased the serum levels of amylase and lipase, and TNF-α and IL-6, as compared to the vehicle control. Also, pancreatic levels of MPO activity, TBARS, and nitrate/nitrite were signifi cantly lower. Histological f indings and TNF-α and iNOS immunostaining further confirmed the amelioration of pancreatic injury by α,β-amyrin. CONCLUSION: α,β-amyrin has the potential to combat acute pancreatitis by acting as an anti-in? ammatory and antioxidant agent.
