Incidence and predictors of hypocalcemia in end-stage renal disease patients on denosumab therapy: A systematic review and metaanalysis

BACKGROUND Denosumab inhibits the receptor activator of nuclear factor *** markedly increases bone mineral density and has been proven to reduce the risk of ***,numerous adverse effects,notably hypocalcemia,are prevalent in patients with end-stage renal disease(ESRD).AIm To analyze the incidence and predictors of hypocalcemia caused by denosumab compared to control in patients with *** We conducted this study in accordance with the Preferred Reporting Items for Systematic Reviews and meta-analyses ***,Scopus,Cochrane Central,and EmBASE were systematically searched from inception through march *** original studies investigating the effects of denosumab on patients with ESRD compared to control were *** primary outcomes of our study were the incidence of mild,severe,and very severe *** outcomes included serum levels of intact parathyroid hormone,calcium,and *** results were pooled and analyzed using a random-effects *** Seven articles comprising 3240 patients were included in our *** treated with denosumab had a significantly increased incidence of mild hypocalcemia[risk ratio(RR):2.79;95%confidence interval(CI):0.99-7.91;P=0.05;I^(2)=37%]and of very severe hypocalcemia(RR:9.58;95%CI:1.58-57.98;P=0.01;I^(2)=49%).However,an increase in the occurrence of severe hypocalcemia was non-significant(RR:4.23;95%CI:0.47-38.34;P=0.20;I^(2)=96%).Alternatively,denosumab showed a significant decrease in serum intact parathyroid hormone[mean difference(mD):-433.20,95%CI:-775.12 to-91.28,I2=98%,P=0.01],while there was a non-significant decrease in phosphate(mD:-0.47,95%CI:-1.35 to 0.41,I^(2)=88%,P=0.30)and calcium levels(mD:-0.33,95%CI:-0.95 to 0.29,I^(2)=94%,P=0.29).CONCLUSION Our study demonstrated that denosumab is significantly associated with mild and very severe hypocalcemia in patients with ESRD making it necessary to detect and prevent this side effect of treatmen

Increased presence of effector lymphocytes during Helicobacter hepaticus-induced colitis

AIm： To identify and characterize drosophila mothers against decapentaplegic （SmAD）3-dependent changes in immune cell populations following infection with He- Iicobacter hepaticus （H. hepaticus）. mETHODS： SmAD3/ （n = L9） and colitis-resistant SmAD3＋/ （n = 24） mice （8-10 wk of age） were in- fected with/-/, hepaticus and changes in immune cell populations [T lymphocytes, natural killer （NK） cells, T regulatory cells] were measured in the spleen and mesenteric lymph nodes （msLNs） at 0 d, 3 d, 7 d and 28 d post-infection using flow cytometry. Genotype-dependent changes in T lymphocytes and granzyme B＋ cells were also assessed after 28 d in proximal colon tissue using immunohistochemistry. RESULTS： As previously observed, SmAD3＋, but not SmAD3＋/- mice, developed colitis, peaking at 4 wk post-infection. No significant changes in T cell subsets were observed in the spleen or in the msLNs between genotypes at any time point. However, CD4＋ and CD8＋/ CD62L＋＋ cells, an effector T lymphocyte population, as well as NK cells （NKp46/DX5＋） were significantly higher in the msLNs of SmAD3/ mice at 7 d and 28 d post-in- fection. In the colon, a higher number of CD3＋ cells were present in SmAD3＋ compared to SmAD3＋/- mice at base- line, which did not significantly change during infection. However, the number of granzyme B＋ cells, a marker of cytolytic lymphoo/tes, significantly increased in SmAD3＋ mice 28 d post-infection compared to both SmAD3＋/- mice and to baseline values. This was consistent with more severe colitis development in these animals. CONCLUSION： Data suggest that defects in SmAD3 signaling increase susceptibility to H. hepaticus-induced colitis through aberrant activation and/or dysregulation of effector lymphoo/tes.

The Pan-Eurasian Experiment modelling Platform(PEEX-mP)is one of the key blocks of the PEEX Research *** PEEX mP has more than 30 models and is directed towards seamless envir-onmental *** main focus area is the Arctic-boreal regions and *** models used in PEEX-mP cover several main components of the Earth’s system,such as the atmosphere,hydrosphere,pedosphere and biosphere,and resolve the physicalchemicalbiological processes at different spatial and temporal scales and *** paper introduces and discusses PEEX mP multi-scale modelling concept for the Earth system,online integrated,forward/inverse,and socioeconomical modelling,and other approaches with a particular focus on applications in the PEEX geographical *** employed high-performance com-puting facilities,capabilities,and PEEX dataflow for modelling results are *** virtual research platforms(PEEXView,Virtual Research Environment,Web-based Atlas)for handling PEEX modelling and observational results are *** over-all approach allows us to understand better physical-chemicalbiological processes,Earth’s system interactions and feedbacks and to provide valuable information for assessment studies on evaluating risks,impact,consequences,*** population,envir-onment and climate in the PEEX *** work was also one of the last projects of *** Zilitinkevich,who passed away on 15 February *** the finalization took time,the paper was actually submitted in 2023 and we could not argue that the final paper text was agreed with him.

Neurodegenerative disorders are characterized by disruptions to neuronal function and circuitry,leading to a variety of clinical syndromes depending on the affected neuroanatomic regions(Geula,1998).many proteinopathies implicated in neurodegenerative diseases are characterized by the pathologic accumulation of proteins into inclusions that are initially deposited in specific areas of the brain and spread widely with disease progression.
