Transgenic mice are widely used to delete or overexpress genes in a cell specific manner to advance knowledge of bone biology,function and *** numerous Cre models exist to target gene recombination in osteoblasts and osteoclasts,few target osteocytes specifically,particularly mature *** goal was to create a spatial and temporal conditional Cre model using tamoxifen to induce Cre activity in mature osteocytes using a Bac construct containing the 5’and 3’regions of the Sost gene(Sost ERT2 Cre).Four founder lines were crossed with the Ai9 Cre reporter *** founder line showed high and specific activity in mature *** and organs were imaged and fluorescent signal *** no activity was observed in 2 day old pups,by 2 months of age some osteocytes were positive as osteocyte Cre activity became spontaneous or‘leaky’with *** percentage of positive osteocytes increased following tamoxifen injection,especially in males,with 43%to 95%positive cells compared to 19%to 32%in *** signal was observed in any bone surface cell,bone marrow,nor in muscle with or without tamoxifen *** spontaneous signal was observed in any other ***,with tamoxifen injection,a few positive cells were observed in kidney,eye,lung,heart and *** other organs,28 in total,were negative with tamoxifen ***,with age,a muscle phenotype was apparent in the Sost-ERT2 Cre ***,although this mouse model may be useful for targeting gene deletion or expression to mature osteocytes,the muscle phenotype may restrict the use of this model to specific applications and should be considered when interpreting data.

Ethical decision-making in older drivers during critical driving situations: An online experiment

The present study examined the impact of aging on ethical decision-making in simulated critical driving scenarios.204 participants from North America,grouped into two age groups(18–30 years and 65 years and above),were asked to decide whether their simulated automated vehicle should stay in or change from the current lane in scenarios mimicking the Trolley *** participant viewed a video clip rendered by the driving simulator at Old Dominion University and pressed the space-bar if they decided to intervene in the control of the simulated automated vehicle in an online *** hierarchical models were used to analyze participants’responses,response time,and acceptability of utilitarian ethical *** results showed significant pedestrian placement,age,and time-to-collision(TTC)effects on participants’ethical *** pedestrians were in the right lane,participants were more likely to switch lanes,indicating a utilitarian approach prioritizing pedestrian *** participants were more likely to switch lanes in general compared to older *** results imply that older drivers can maintain their ability to respond to ethically fraught scenarios with their tendency to switch lanes more frequently than younger counterparts,even when the tasks interacting with an automated driving *** current findings may inform the development of decision algorithms for intelligent and connected vehicles by considering potential ethical dilemmas faced by human drivers across different age groups.

We present radial velocity(RV)curve templates of RR lyrae first-overtone(RRc)stars constructed with the Mg I b triplet and Hαlines using time-domain Medium-Resolution Survey spectra of seven RRc stars from large Sky Area Multi-Object Fiber Spectroscopic Telescope(lAMOST)Data Release ***,we derive the relation between the stellar RV curve amplitudes and g-band light curve amplitudes from Zwicky Transient Facility(ZTF)public *** those RRc stars without ZTF g-band light curves,we provide the conversions from the light curve amplitudes in ZTF r-and i-bands,Gaia G-band,and V-band from the All-Sky Automated Survey for Supernovae to those in ZTF *** validate our RV curve templates using the RRc star SV Scl and find the uncertainties of systemic RV are less than 2.11 km s~(-1)and 6.08 km s~(-1)based on the Mg I b triplet and Hαlines,*** calculate the systemic RVs of 30 RRc stars using the RV curve templates constructed with the Mg I b triplet and Hαlines and find the systemic RVs are comparable with each *** RV curve template will be particularly useful for obtaining the systemic RV of RRc using the lAMOST spectroscopy.

Garcinia cambogia extract(GC)with its active component consisting of hydroxycitric acid(HCA)is widely utilized for weight *** HCA salts are available,including calcium,magnesium,potassium and mixtures of ***,these salts exhibit different properties with some,but not all,improving glucose tolerance and blood ***,obesity-prone C57Bl/6J mice were fed a high-fat diet(HFD,45 kcal%fat)with or without GC(1%,w/w)for 16 *** active arm reduced visceral fat,adipocyte size and serum glucose,yet purportedly also exhibited hepatic collagen accumulation,lipid peroxidation and increased mRNA levels of genes related to oxidative *** latter findings are at odds with a large body of animal and human studies that have been conducted on the safety and efficacy of *** literature shows HCA to be protective against the liver toxicity associated with ethanol and dexamethasone administration,and to maintain serum aspartate aminotransferase,alanine aminotransferase and alkaline phosphatase at near normal *** both animal and clinical literature,elevated intakes of HCA per se have not led to signs of inflammation or *** compound has been found to reduce markers of inflammation in brain,intestines,kidney and serum.

A variety of Ca2+signals,in the form of intercellular pulses and waves,have beenreported to be associated with the various sequential stages of somitogenesis:from convergent extension and the formation of the paraxial mesoderm;during thepatterning of the paraxial mesoderm to establish segmental units;throughout theformation of the morphological boundaries that delineate the segmental units,andfinally from within the maturing somites as they undergo subsequent develop-ment and *** to both the technical challenges presented in imag-ing intact,developing embryos,and the subtle nature of the Ca2+transientsgenerated,they have proved to be difficult to ***,a combinationof cultured cell preparations and improvements in explant and whole embryo im-aging techniques has begun to reveal a new and exciting class of developmentalCa2+*** this chapter,we review the small,but expanding,number ofreports in the literature and attempt to identify common characteristics of thesomitogenic Ca2+transients,such as their mode of generation,as well as theirspatial and temporal *** may help to elucidate the significance andfunction of these intriguing Ca2+transients and thus integrate them into the com-plex signaling networks that orchestrate early developmental events.

Transporters Contributing to Iron Trafficking in Plants

This review will discuss recent progress in understanding the many roles of transporters in the whole-plant physiological processes that maintain iron （Fe） homeostasis. These processes include uptake from the soil via roots, control of transport from roots to above-ground parts of the plant, unloading of Fe from the xylem in above-ground parts, loading of Fe into mitochondria and plastids, transport of Fe to reproductive parts of the plant, and Fe mobilization during seed germination. In addition, we will discuss the mechanisms that plants use to cope with an apparently unintended conse- quence of Fe acquisition： the uptake of toxic heavy metals via Fe transporters. Rapid progress has been made in under- standing the transport processes involved in each of these areas in the last 5 years and this review will focus on this recent progress. We will also highlight the key questions regarding transport steps that remain to be elucidated.

Microbial community coalescence and nitrogen cycling in simulated mortality decomposition hotspots

Background The pulsed introduction of dead plant and animal material into soils represents one of the primary mechanisms for returning organic carbon(C)and nitrogen(N)compounds to biogeochemical *** of animal carcasses provides a high C and N resource that stimulates indigenous environmental microbial communities and introduces non-indigenous,carcass-derived microbes to the ***,the dynamics of the coalesced microbial communities,and the relative contributions of environment-and carcass-derived microbes to C and N cycling are *** test whether environment-derived,carcass-derived,or the combined microbial communities exhibited a greater influence on C and N cycling,we conducted controlled laboratory experiments that combined carcass decomposition fluids and soils to simulate carcass decomposition *** selectively sterilized the decomposition fluid and/or soil to remove microbial communities and create different combinations of environment-and carcass-derived communities and incubated the treatments under three temperatures(10,20,and 30℃).Results Carcass-derived bacteria persisted in soils in our simulated decomposition scenarios,albeit at low *** communities had higher respiration rates at 10 and 30℃ compared to soil or carcass communities ***,at higher temperatures,mixed communities had reduced diversity,but higher respiration,suggesting functional *** communities treatments also provided evidence that carcass-associated microbes may be contributing to ammonification and denitrification,but that nitrification is still primarily carried out by native soil *** Our work yields insight into the dynamics of microbial communities that are coalescing during carcass decomposition,and how they contribute to recycling carcasses in terrestrial ecosystems.
