Insect-induced galls are abnormal plant growths that can provide food and shelter to their inhabitants,resulting in stressed plant tissue that may alter the conditions for the colonization or proliferation of endophytic *** investigated the effect gall formation has on fungal endophyte communities and *** three closely-related gallforming aphid species that specialize on poplars,we characterized fungal endophyte diversity in galls and surrounding petiole and leaf lamina tissue.A total of 516 fungal endophyte samples were isolated from 272 tissue samples(32 leaves,31 petioles,and 209 galls),resulting in 23 distinct *** sharing a common host plant and often forming spatially contiguous galls,the endophyte profiles within the galls of each aphid species were distinct,not only from the galls of the other species,but also from surrounding plant *** results suggest that insect galls can affect the composition of fungal endophyte species in plant tissues,by altering either the colonization or proliferation of their endophytic ***,fungal endophytes may be important in the ecology and evolution of insect galls.

elevated expression of heat shock protein 90 （HSP90） has been found in kidneys and serum of systemic lupus erythematosus （SLe） patients and MRLIMp-FasIprIFasJpr（MRLIIpr） autoimmune mice. We investigated if inhibition of HSP90 would reduce disease in MRL/ Ipr mice. In vitro, pretreatment of mesangial cells with HSP90 inhibitor Geldanamycin prior to immune-stimulation showed reduced expression of IL-6, IL-12 and NO. In vivo, we found HSP90 expression was elevated in MRL/Ipr kidneys when compared to C57BL/6 mice and MRIJIpr mice treated with HSP90 inhibitor 17-DMAG. MRIJIpr mice treated with 17-DMAG showed decreased proteinuria and reduced serum anti-dsDNA antibody production. Glomerulonephritis and glomerular IgG and C3 were not significantly affected by administration of 17-DMAG in MRIJIpr. 17-DMAG increased CD8＋ T cells, reduced double-negative T cells, decreased the CD4/CD8 ratio and reduced follicular B cells. These studies suggest that HSP90 may play a role in regulating T-cell differentiation and activation and that HSP90 inhibition may reduce inflammation in lupus.

Postpartum involution is the process by which the lactating mammary gland returns to the pre-pregnant state after weaning. expression of tumor-promotional collagen, upregulation of matrix metalloproteinases, infiltration of M2 macrophages, and remodeling of blood and lymphatic vasculature are all characteristics shared by the involuting mammary gland and breast tumor microenvironment. The tumor promotional nature of the involuting mammary gland is perhaps best evidenced by cases of postpartum breast cancer (PPBC), or those cases diagnosed within 10 years of most recent childbirth. Women with PPBC experience more aggressive disease and higher risk of metastasis than nulliparous patients and those diagnosed outside the postpartum window. Semaphorin 7a (SeMA7A), cyclooxygenase-2 (COX-2), and collagen are all expressed in the involuting mammary gland and, together, predict for decreased metastasis free survival in breast cancer. Studies investigating the role of these proteins in involution have been important for understanding their contributions to PPBC. Postpartum involution thus represents a valuable model for the identification of novel molecular drivers of PPBC and classical cancer hallmarks. In this review, we will highlight the similarities between involution and cancer in the mammary gland, and further define the contribution of SeMA7A/COX-2/collagen interplay to postpartum involution and breast tumor progression and metastasis.

Dear editor, Chest pain is a frequent complaint of patients presenting to the emergency department (eD), and many of them are referred to the cardiology service for further investigation. At the Charles V. Keating emergency and Trauma Centre in Halifax, Nova Scotia, Canada, 4,800 (6.6%) of the approximately 73,000 patients per year register with a complaint of “chest pain”, and 20% of patients are referred to cardiology. Coagulation studies, specifi cally international normalized ratio (INR) frequently part of the “routine” panel of blood tests, are ordered for patients in the eD being investigated or treated for chest pain suspected to be cardiac in nature. Recent calls to examine how much of our practice is likely to benefit patients in any way have led us to question the clinical utility of routine use of these tests.

The usefulness of the apparent redox potential discontinuity(aRPD) in assessments of marine benthic habitat quality was explored at two intertidal mudflats along the north Pacific coast of Canada. Two transects were established at each intertidal site, with three sediment biogeochemistry cores collected from each transect four times over the summer of 2016. Measurements of the sediment pore water dissolved oxygen(DO) content and redox(eh) conditions were taken at the surface of the core(measured vertically), as well as at increasing depths(1 cm between readings) into the sediment(measured horizontally through predrilled holes in the biogeochemistry corer). While oxic, anoxic, oxidized, and reduced sediment pore water was observed above and below the aRPD, in general, sediment above the aRPD had higher DO content, and higher eh values than sediment below the aRPD. Therefore, the aRPD depth can be used as a relative indicator of sediment pore water DO and eh conditions: sediment with a deeper aRPD depth has more available DO, and the pore water has higher eh values(more oxidized or less reduced) than sediment with a shallower aRPD depth. As such, the aRPD depth is a useful parameter to include in models that assess the quality of marine benthic habitats.

Identification of Ca2+ signaling components in neural stem/progenitor cells during differentiation into neurons and glia in intact and dissociated zebrafish neurospheres

The development of the CNS in vertebrate embryos involves the generation of different sub-types of neurons and glia in a complex but highly-ordered spatio-temporal manner. Zebrafish are commonly used for exploring the development, plasticity and regeneration of the CNS, and the recent development of reliable protocols for isolating and culturing neural stem/progenitor cells(NSCs/NPCs) from the brain of adult fish now enables the exploration of mechanisms underlying the induction/specification/differentiation of these cells. Here, we refined a protocol to generate proliferating and differentiating neurospheres from the entire brain of adult zebrafish. We demonstrated via RT-qPCR that some isoforms of ip3 r, ryr and stim are upregulated/downregulated significantly in differentiating neurospheres, and via immunolabelling that 1,4,5-inositol trisphosphate receptor(IP3 R) type-1 and ryanodine receptor(RyR) type-2 are differentially expressed in cells with neuron-or radial glial-like properties. Furthermore, ATP but not caffeine(IP3 R and RyR agonists, respectively), induced the generation of Ca^(2+) transients in cells exhibiting neuron-or glial-like morphology. These results indicate the differential expression of components of the Ca^(2+) -signaling toolkit in proliferating and differentiating cells. Thus, given the complexity of the intact vertebrate brain, neurospheres might be a useful system for exploring neurodegenerative disease diagnosis protocols and drug development using Ca^(2+) signaling as a read-out.

Cyanobacteria may adversely impact aquatic ecosystems through oxygen depletion and cyanotoxin production. These cyanotoxins can also harm human health and livestock. In recent years, cyanobacterial blooms have been observed in several drinking water reservoirs in Kentucky, United States. In Kentucky, the paradigm is that phosphorous is the limiting nutrient for cyanobacteria growth. To explore this paradigm, an indoor microcosm study was conducted using hypereutrophic Guist Creek Lake water. Samples were collected and spiked with various combinations of locally used agricultural grade fertilizers, including ammonium nitrate, urea, and triple phosphate (calcium dihydrogen phosphate). Samples were incubated indoors for the photoperiod-specific to the time of the year. Cyanobacteria density, measured by phycocyanin, did not demonstrate increased growth with the addition of phosphate fertilizer alone. Cyanobacteria growth was enhanced in these conditions by the combined addition of ammonium nitrate, urea, and phosphorus fertilizer. Growth also occurred when using either ammonium nitrate or urea fertilizer with no additional phosphorus input, suggesting that phosphorus was not limiting the cyanobacteria at the time of sample collection. The addition of both nitrogen fertilizers (ammonium nitrate and urea) at the concentrations used in this study, in the absence of phosphorus, was deleterious to both the Chlorophyta and cyanobacteria. The results suggest further studies using more robust experimental designs are needed to explore lake-specific dual nutrient management strategies for preventing cyanobacterial blooms in this phosphorus-rich hypereutrophic lake and possibly other hypereutrophic lakes.

Background: The operative report is an essential component of communication, patient care, and safety;it is a JCAHO requirement and legal document. Almost 80% of residency program directors surveyed agreed on the importance of the operative report, yet less than 20% of programs provide formal resident training. Documentation has important implications in obstetrics as route and timing of future deliveries are often influenced by operative findings. Purpose: Examine completeness of cesarean operative reports using a validated instrument adapted from general surgery. Assess the impact of an educational resident in-service on the required elements and documentation in resident generated cesarean operative reports. Methods: A quality improvement project identified significant deficiencies in resident generated cesarean operative reports. We conducted a retrospective chart review of cesarean operative reports from November 2019 to May 2020 at Jersey Shore University Medical Center. Our study was conducted in three phases. In phase 1, a cohort of cesarean operative reports was examined using a structured evaluation tool. In phase 2, an educational resident in-service on standardization of operative notes was provided. In phase 3, another cohort of cesarean operative reports was examined using the same evaluation tool after the resident in-service. Analysis for improvement was performed. Results: Fifty pre and post-in-service cesarean operative report evaluations weree="font-family:Verdana;"> compared (N = 100).
