Endoscopic retrograde cholangiopancreatography with rendezvous cannulation reduces pancreatic injury

AIM:To examine whether rendezvous endoscopic retrograde cholangiopancreatography(ERCP)is associated with less pancreatic damage,measured as leakage of proenzymes,than conventional ***:Patients(n=122)with symptomatic gallstone disease,intact papilla and no ongoing inflammation,were prospectively enrolled in this case-controldesigned ***-one patients were subjected to laparoscopic cholecystectomy and if intraoperative cholangiography suggested common bile duct stones(CBDS),rendezvous ERCP was performed intraoperatively(n=40).Patients with a negative cholangiogram constituted the control group(n=41).Another 41 patients with CBDS,not subjected to surgery,underwent conventional *** proenzymes,procarboxypeptidase B and trypsinogen-2 levels in plasma,were analysed at 0,4,8 and 24 *** proenzymes were determined in-house with a double-antibody enzyme linked immunosorbent *** amylase was measured by an enzymatic colourimetric modular analyser with the manufacturer’s *** samples were blinded at ***:Post ERCP pancreatitis(PEP)occurred in3/41(7%)of the patients cannulated with conventional ERCP and none in the rendezvous *** serum levels indicating pancreatic leakage were significantly higher in the conventional ERCP group compared with the rendezvous ERCP group regarding pancreatic amylase levels in the 4-and 8-h samples(P=0.0015;P=0.03),procarboxypeptidase B in the4-and 8-h samples(Ples(P=0.03).No differences in these markers were observed in patients treated with rendezvous cannulation technique compared with patients that underwent cholecystectomy alone(control group).Post procedural concentrations of pancreatic amylase and procarboxypeptidase B were significantly correlated with pancreatic duct cannulation and ***:Rendezvous ERCP reduces pancreatic enzyme leakage compared with conventional ERCP cannulation ***,l

Muscle RING-finger protein-1 (MuRFl) functions and cellular localization are regulated by SUMOl post-translational modification

The muscle RING-finger protein-1 (MuRFl) is an E3 ubiquitin ligase expressed in skeletal and cardiac muscle tissues and it plays important roles in muscle remodeling. Upregulation of MuRFl gene transcription participates in skeletal muscle atrophy, on contrary downregulation of protein expression leads to cardiac hypertrophy. MuRFl gene point mutations have been found to generate protein aggregate myopathies defined as muscle disorder characterized by protein accumulation in muscle fibers. We have discovered that MuRFl turned out to be also a target for a new post-translational modification arbitrated by conjugation of SUM01 and it is mediated by the SUMO ligases E2 UBC9 and the E3 PIASy/4. SUMOylation takes place at lysine 238 localized at the second coiled-coil protein domain that is required for efficient substrate interaction for polyubiquitination. We provided evidence that SUMOylation is essential for MuRFl nuclear translocation and its mitochondria accumulation is enhanced in hyper? glycemic conditions delivering a stabilization of the overall SUMOylated proteins in cultured myocytes. Thus, our findings add this SUM01 post-translational modification as a new concept to understand muscle disorders related to the defect in MuRFl activity.

以前的研究已经表明盘形凸轮的制造方法和凸轮从动件性能的动态质量之间在统计学上的重要关系。本文设计和制造的凸轮动态试验装置(CDTF)在调速和负载控制的条件下运行,可以采用各种技术加工而成的凸轮。动态试验的内容有:从动件的切向和法向加速度,从动件的切向和法向力,滚子的滑移也可以精确的测量。在凸轮试件运行之前,凸轮表面粗糙度可以由霍曼尔(Hommel)T—20S表面粗糙度测试计进行测量。跑合期后,在测量力、速度和加速度的情况下又可得到表面参数。加速度和力的数据可转换到频域。所有的试验均是随机的,数据要进行方差分析,以求得加速度有效功率谱和凸轮试件表面粗糙度参数之间的相关性。研究的凸轮试件是由连续数控制造技术采用1/°、1/2°、和1°的数值步长加工而成。试验结果表明:铣削和磨削的凸轮的动态性能具有很大的差异,但没有由于数值步长的不同而引起差异。一种可以替代铣削凸轮整体淬硬的热处理方法(离子氮化)已经显示出在动态性能方面有很大的改进。

Tissue distribution of perfluoroalkyl acids and health status in wild Mozambique tilapia(Oreochromis mossambicus) from loskop Dam, Mpumalanga, South Africa

This study examined concentrations of 15 perfluoroalkyl acids（PFAAs） in tissues from male Mozambique tilapia（Oreochromis mossambicus） collected at loskop Dam, Mpumalanga,South Africa in 2014 and 2016. Nine of the 15 PFAAs were detected frequently and were included in statistical analysis and included two of the most commonly known PFAAs,perfluorooctanesulfonic acid（PFOS）（median, 41.6 ng/g） and perfluorooctanoic acid（PFOA）（median, 0.0825 ng/g）. Of the tissues measured, plasma（2016 and 2014 median, 22.2 ng/g）contained the highest PFAA burden followed by（in descending order）： liver（median,11.6 ng/g）, kidney（median, 9.04 ng/g）, spleen（median, 5.92 ng/g）, adipose（median, 2.54 ng/g）, and muscle（median, 1.11 ng/g）. loskop Dam tilapia have been affected by an inflammatory disease of the adipose tissue known as pansteatitis, so this study also aimed to investigate relationships between PFAA tissue concentrations and incidence of pansteatitis or fish health status. Results revealed that healthy tilapia exhibited an overall higher（p-value 〈 0.05） PFAA burden than pansteatitis-affected tilapia across all *** analysis showed that organs previously noted in the literature to contain the highest PFAA concentrations, such as kidney, liver, and plasma, were the organs driving the difference in PFAA burden between the two tilapia groups. Care must be taken in the interpretations we draw from not only the results of our study, but also other PFAA measurements made on populations（human and wildlife alike） under differing health status.

Cardiac extracellular matrices(ECM)play crucial functional roles in cardiac *** studies have mainly focused on collagen,the major structural ECM in heart *** role of elastin in cardiac mechanics,however,is poorly *** this study,we investigated the spatial distribution and microstructural morphologies of cardiac elastin in porcine left *** demonstrated that the epicardial elastin network had location-and depth-dependency,and the overall epicardial elastin fiber mapping showed certain correlation with the helical heart muscle fiber *** compared to the epicardial layer,the endocardial layer was thicker and has a higher elastin-collagen ratio and a denser elastin fiber network;moreover,the endocardial elastin fibers were finer and more wavy than the epicardial elastin fibers,all suggesting various interface *** myocardial interstitial elastin fibers co-exist with the perimysial collagen to bind the cardiomyocyte bundles;some of the interstitial elastin fibers showed a locally aligned,hinge-like structure to connect the adjacent cardiomyocyte *** collagen-elastin combination reflects an optimal design in which the collagen provides mechanical strength and elastin fibers facilitate recoiling during ***,cardiac elastin fibers,along with collagen network,closely associated with the Purkinje cells,indicating that this ECM association could be essential in organizing cardiac Purkinje cells into“fibrous”and“branching”morphologies and serving as a protective feature when Purkinje fibers experience large deformations in *** short,our observations provide a structural basis for future in-depth biomechanical investigations and biomimicking of this long-overlooked cardiac ECM component.

Youth gender differences in alcohol use: A prospective study of multiple youth assets and the neighborhood environment

Research has identified factors (assets) that protect youth from engaging in risk behaviors including alcohol use. Very little research has examined asset/nonuse of alcohol associations by youth gender or determined if asset/nonuse of alcohol associations are influenced by the neighborhood environment. The purpose of the study was to determine if multiple youth assets and neighborhood factors are differentially associated with youth nonuse of alcohol by gender. Method: Five waves of data were collected annually from households (N = 1111) randomly selected to participate in the Youth Asset Study. Seventeen assets and 6 neighborhood factors assessed at waves 1 - 4 were analyzed longitudinally using marginal logistic regression to predict nonuse of alcohol at waves 2 - 5. Results: Sample mean age was 14.3 years, 52% female;39% white, 28% Hispanic, 23% African-American, and 9% other. Numerous assets were prospec- tively associated with alcohol nonuse for females (12 assets) and males (16 assets). Three assets were significantly more protective from alcohol use for males compared to females. Final modeling indicated that three assets were protective from alcohol use for both genders and that one asset was protective only for males. There were no significant associations between the neighborhood variables and nonuse of alcohol, and the neighborhood variables did not influence the asset/nonuse of alcohol associations. Conclusions: Youth assets appear to protect both genders from future alcohol use but males may benefit even more from asset-building prevention programming. Youth alcohol use and alcohol nonuse/asset associations may not be influenced by the neighborhood environment.

Non-redundant Contribution of the Plastidial FAD8 ω-3 Desatumse to Glycerolipid Unsatumtion at Different Temperatures in Arabidopsis

Plastidial ω-3 desaturase FAD7 is a major contributor to trienoic fatty acid biosynthesis in the leaves of Arabidopsis plants. However, the precise contribution of the other plastidial ω-3 desaturase, FAD8, is poorly understood. Fatty acid and lipid analysis of several ω-3 desaturase mutants, including two insertion lines of AtFAD7 and AtFAD8, showed that FAD8 partially compensated the disruption of the AtFAD7 gene at 22℃, indicating that FAD8 was active at this growth temperature, contrasting to previous observations that circumscribed the FAD8 activity at low temperatures. Our data revealed that FAD8 had a higher selectivity for 18：2 acyl-lipid substrates and a higher preference for lipids other than galactolipids, particularly phosphatidylglycerol, at any of the temperatures studied. Differences in the mechanism controlling AtFAD7 and AtFAD8 gene expression at different temperatures were also detected. Confocal microscopy and biochemical analysis of FAD8-YFP over-expressing lines confirmed the chloroplast envelope localization of FAD8. Co-localization experiments suggested that FAD8 and FAD7 might be located in close vicinity in the envelope membrane. FAD8-YFP over-expressing lines showed a specific increase in 18：3 fatty acids at 22℃. Together, these results indicate that the function of both plastidial ω-3 desaturases is coordinated in a non-redundant manner.
