Nicotine-induced transcriptional changes and mitochondrial dysfunction in the ventral tegmental area revealed by single-nucleus transcriptomics

Nicotine is widely recognized as the primary contributor to tobacco *** studies have indicated that molecular and behavioral responses to nicotine are primarily mediated by ventral tegmental area(VTA)neurons,and accumulating evidence suggests that glia play prominent roles in nicotine ***,VTA neurons and glia have yet to be characterized at the transcriptional level during the progression of nicotine ***,a male mouse model of nicotine self-administration is established and the timing of three critical phases(pre-addiction,addicting,and post-addiction phase)is ***-nucleus RNA sequencing in the VTA at each phase is performed to comprehensively classify specific cell *** changes occurred during the addicting and post-addiction phases,with the addicting phase displaying highly dynamic neuroplasticity that profoundly impacts the transcription in each cell ***,significant transcriptional changes in energy metabolism-related genes are observed,accompanied by notable structural alterations in neuronal mitochondria during the progression of nicotine *** results provide insights into mechanisms underlying the progression of nicotine addiction,serving as an important resource for identifying potential molecular targets for nicotine cessation.

Transcriptional landscape of rice roots at the single-cell resolution

There are two main types of root systems in flowering plants,namely taproot systems of dicots and fibrous root systems found in *** this fundamental split,our current knowledge of cellular and molecular mechanism driving root development is mainly based on studies of the dicot model ***,the world major crops are monocots and little is known about the transcriptional programs underlying cell-type specification in this ***,we report the transcriptomes of more than 20000 single cells derived from root tips of two agronomically important rice *** combined computational and experimental analyses we were able to robustly identify most of the major cell types and define novel cell-type-specific marker genes for both ***,we found divergent cell types associated with specific regulatory programs,including phytohormone biosynthesis,signaling,and response,which were well conserved between the two rice *** addition,we detected substantial differences between the cell-type transcript profiles of Arabidopsis and *** species-specific features emphasize the importance of analyzing tissues across diverse model species,including *** together,our study provides insight into the transcriptomic landscape of major cell types of rice root tip at single-cell resolution and opens new avenues to study cell-type specification,function,and evolution in plants.

The roots of legume plant play a crucial role in nitrogen fixation. However, the transcriptomes of different cell types of legume root and their functions remain largely unknown. Here, we performed single-cell RNA sequencing and profiled more than 22,000 single cells from root tips of Lotus japonicus, a model species of *** identified seven clusters corresponding to seven major cell types, which were validated by in situ hybridization. Further analysis revealed regulatory programs including phytohormone and nodulation associated with specific cell types, and revealed conserved and diverged features for the cell types. Our results represent the first single-cell resolution transcriptome for legume root tips and a valuable resource for studying the developmental and physiological functions of various cell types in legumes.

Ginger(Zingiber officinale),the type species of Zingiberaceae,is one of the most widespread medicinal plants and ***,we report a high-quality,chromosome-scale reference genome of ginger‘Zhugen’,a traditionally cultivated ginger in Southwest China used as a fresh vegetable,assembled from PacBio long reads,Illumina short reads,and high-throughput chromosome conformation capture(Hi-C)*** ginger genome was phased into two haplotypes,haplotype 1(1.53 Gb with a contig N50 of 4.68 M)and haplotype 0(1.51 Gb with a contig N50 of 5.28 M).Homologous ginger chromosomes maintained excellent gene pair *** 17,226 pairs of allelic genes,11.9%exhibited differential expression between *** on the results of ginger genome sequencing,transcriptome analysis,and metabolomic analysis,we proposed a backbone biosynthetic pathway of gingerol analogs,which consists of 12 enzymatic gene families,PAL,C4H,4CL,CST,C3’H,C3OMT,CCOMT,CSE,PKS,AOR,DHN,and *** analyses also identified the likely transcription factor networks that regulate the synthesis of gingerol ***,this study serves as an excellent resource for further research on ginger biology and breeding,lays a foundation for a better understanding of ginger evolution,and presents an intact biosynthetic pathway for species-specific gingerol biosynthesis.

The brain of the domestic pig(Sus scrofa domesticus)has drawn considerable attention due to its high similarities to that of ***,the cellular compositions of the pig brain(PB)remain *** we investigated the single-nucleus transcriptomic profiles of five regions of the PB(frontal lobe,parietal lobe,temporal lobe,occipital lobe,and hypothalamus)and identified 21 cell *** cross-species comparison of mouse and pig hypothalamus revealed the shared and specific gene expression patterns at the single-cell ***,we identified cell types and molecular pathways closely associated with neurological disorders,bridging the gap between gene mutations and *** reported,to our knowledge,the first single-cell atlas of domestic pig cerebral cortex and hypothalamus combined with a comprehensive analysis across species,providing extensive resources for future research regarding neural science,evolutionary developmental biology,and regenerative medicine.

Boron-rich layer removal and surface passivation of boron-doped p–n silicon solar cells

In boron-doped p+-n crystalline silicon（Si） solar cells, p-type boron doping control and surface passivation play a vital role in the realization of high-efficiency and low cost pursuit. In this study, boron-doped p+-emitters are formed by boron diffusion in an open-tube furnace using borontribromide（BBr3） as precursor. The formed emitters are characterized in detail in terms of shape of the doping profile, surface doping concentration, junction depth, sheet resistance and removal of the boron-rich layer（BRL）. In the aspect of BRL removal, three different methods were adopted to investigate their influence on device performance. The results demonstrate that our proposed chemical etch treatment（CET） with the proper etching time could be an effective way to remove the *** removal of the BRL, Al;O;/SiN;stacks are deposited by atomic layer deposition（ALD） and plasma-enhanced chemical vapor deposition（PECVD） to passivate the cell surface. It was found that a reasonably-high implied Voc of 680 mV has been achieved for the fabricated n-type Si solar cells.

蒸汽发生器传热管的在役自动涡流检验装置对定位器的定位精度要求较高,定位偏差使得探头磨损加剧甚至卡在传热管中。为了解决定位的精确计算问题,本文基于三自由度的检测机械臂,建立了机械臂关节的运动学模型,对正、逆运动学进行了定位计算求解,同时为解决安装机械臂时基座的位置偏差提出了误差消除的方法。试验结果表明,该定位计算方法的结果准确、稳定可靠。

核反应堆燃料组件长期服役过程中,锆合金与高温水作用会产生氧化形成一层氧化膜,由于膜层较薄,测量精度需要达到±5μm,测量精度的高要求为氧化膜厚度测量系统的研发带来难度。为此,本文在涡流提离效应的基础上研制了一套燃料棒氧化膜厚度测量系统,讨论了测量系统中的关键技术,同时对涡流提离效应的电磁场进行仿真。测试结果表明,该测量系统测量精度高、稳定可靠,可实现在役燃料棒氧化膜的厚度测量。

重点风险探井河探1井四开钻遇超高压油气层,二开Ø244.5 mm套管抗内强度60 MPa,关井能力不足,存在井控风险。为保障本井Ø177.8 mm套管回接固井的正常施工,通过室内实验开展加重材料及外加剂的优选,采用球形铁粉加重材料,配置出满足该井注水泥塞施工的密度2.60 g/cm^(3)水泥浆体系,在160℃、170 MPa条件下,48 h强度24.5 MPa,168 h强度28.1 MPa,滤失量23 mL。现场采用批混批注的方式,结合精细控压技术,实现了注水泥塞全过程的压力平衡,顺利完成了该井注水泥塞施工,水泥塞质量合格。应用结果表明,配置的2.60 g/cm^(3)超高密度水泥浆适用于类似的超高压固井施工,通过精细控压,可以解决钻井液密度不足以压稳地层、窄安全密度窗口固井时存在的压力波动问题,为后续同类井控压注水泥塞及控压固井积累了经验。
