Background:Mucopolysaccharidose. type.Ⅲ (MPS Ⅲ) are.a group of autosomai re.e.sive.lysosomal storage.dise.se.,cause. by mutations in ge.e. that code.for e.zyme. involve. in the.lysosomal de.radation of he.aran sulphate.he.aran sulfate.sulfamidase.(SGSH),a-N-ace.ylglucosaminidase.(NAGLU),he.aran sulfate.ace.yl-CoA:a-glucosaminide.N-ace.yltransfe.ase.(HGSNAT),and N-ace.ylglucosamine.6-sulfatase.(GNS).Me.hods:In this study,we.have.pe.forme. the.mole.ular analysis of the.SGSH,NAGLU and HGSNAT ge.e. in 10 patie.ts from 6 diffe.e.t MPS Ⅲ Tunisian ***:In the.SGSH ge.e.two mutations we.e.ide.tifie.:one.nove. (p.D477N) and one.alre.dy de.cribe. (p.Q365X).In the.NAGLU ge.e.two nove. mutations we.e.discove.e. (p.L550P and p.e.53X).For the.nove. misse.se.mutations found in the.e.two ge.e. we.pe.forme. an in silico structural analysis and the.re.ults we.e.consiste.t with the.clinical course.of the.patie.ts harboring those.***,in HGSNAT ge.e.we.found the.splice.ite.mutation c.234+1G>A that had alre.dy be.n re.orte. as re.ative.y fre.ue.t in MPS ⅢC patie.ts from countrie. surrounding the.basin of the.Me.ite.rane.n *** pre.e.ce.in two Tunisian MPS ⅢC familie. points to the.hypothe.is of its pe.i Me.ite.rane.n *** the.e.ce.tion of the.c.234+1G>A mutation,that was ide.tifie. in two unre.ate. AIM To e.aluate.the.influe.ce.of cre.tinine.me.hodology on the.pe.formance.of chronic kidne. dise.se.CKD)-e.ide.iology Collaboration Group-calculate. e.timate. glome.ular filtration rate.CKD-e.I-e.FR) for CKD diagnosis/staging in a large.cohort of diabe.ic patie.ts. Me.HODS Fasting blood sample. we.e.take. from diabe.ic patie.ts atte.ding our clinic for the.r re.ular annual e.amination, including laboratory me.sure.e.t of se.um cre.tinine.and *** Our re.ults indicate. an ove.all e.ce.le.t agre.me.t in CKD staging(kappa = 0.918) be.we.n the.Jaffé se.um cre.tinine.and e.zymatic se.um cre.tinine.base. CKDe.I-e.FR, with 9% of discordant case.. As compare. to the.e.zymatic cre.tinine. the.majority of discordance.(8%) we.e.positive. i.e., associate. with the.more.advance. CKD stage.re.classification, whe.e.s only 1% of case. we.e.ne.ative.y discordant if Jaffé cre.tinine.was use. for e.FR calculation. A minor proportion of the.discordant case.(3.5%) we.e.re.classifie. into clinically re.e.ant CKD stage.indicating mildly to mode.ate.y de.re.se. kidne. function(e. 1.73 m^2). Significant acute.and chronic hype.glycae.ia, asse.se.as plasma glucose.and Hb A1 c le.e.s far above.the.re.omme.de. glycae.ic goals, was associate. with positive.y discordant case.. Due.to a ve.y low fre.ue.cy, positive.discordance.is not like.y to pre.e.t a gre.t burde. for the.he.lth-care.provide.s, while.inte.sifie. me.ical care.may actually be.be.e.icial for the.small numbe. of discordant patie.ts. Le.shmania infantum using in vitro mode.s, re.re.e.t a nove. alte.native.as a control strate.y for the.transmission of le.shmaniasis and also provide.the.opportunity to de.e.t natural products or antimicrobial pe.tide. with diffe.e.t biological activitie.. In this study, we.e.aluate.the.le.shmanicidal and antimicrobial activitie. of Pantoe. ananatis, Ochrobactrum anthropi and e.te.obacte. cloacae. isolate. from the.dige.tive.tract of Lutzomyia e.ansi and the.susce.tibility of the.e.bacte.ia to commonly use. antibiotics. The.antagonistic e.fe.t of Pantoe. ananatis, Ochrobactrum anthropi and e.te.obacte. cloacae.was e.aluate. against six spe.ie. of human pathoge.ic bacte.ia and against stationary (Me.acyclic-like. and e.pone.tial promastigote. (Procyclic-like. of Le.shmania infantum (BCN-GFP strain) by co-culture.assays for 24 hours. The.activity of the.bacte.ial isolate. on Le.shmania infantum promastigote. was quantifie. by flow cytome.ry. The.susce.tibility of the.bacte.ial strains to clinically use. antibiotics was analyze. by antibiogram. The.highe.t pe.ce.tage.of inhibition was obse.ve. against e.pone.tial promastigote. with bacte.ial conce.trations of 108 CFU/ml of e.te.obacte. cloacae.(77.29% ± 0.6%) and Pantoe. ananatis (70.17% ± 1.1%). The.e.tracts produce. by thre. bacte.ial isolate. showe. similar biological activity (13 mm - 22 mm inhibition halos) against all te.te. bacte.ia;howe.e., significant diffe.e.ce. we.e.obse.ve. with re.pe.t to gram-positive.bacte.ia (P e.most active.antibacte.ial activity was displaye. against the.pathoge.ic bacte.ia Bacillus ce.e.s.
