A practical strategy to subcutaneous administered in-situ gelling co-delivery system of arsenic and retinoic acid for the treatment of acute promyelocytic leukemia

Arsenic trioxide(AtO) combined with all trans retinoic acid(AtRA) is the first choice for the treatment of low and medium risk acute promyelocytic leukemia(APL). Clinical studies reported that the combination of AtO and AtRA could achieve a significant curative effect. However, the retinoic acid syndrome, serious drug resistance and the short half-life in vivo which lead to frequent and large dose administration limit the application of AtRA. In addition, the preparations of arsenic are conventional injections and tablets in clinic, which has poor patients’ compliance caused by frequent long-term administration and serious side effects. In order to overcome the above limitations, a phospholipid phase separation gel(PPSG) loaded with AtO and AtRA was developed. AtO + AtRA-PPSG(AAP), as a biodegradable sustained-release delivery system, was the first achievement of co-delivery of hydrophilic AtO and lipophilic AtRA with high drug loading which is the main problem in the application of nano preparation. the prepared PPSG displayed high safety and biocompatibility. the drug in PPSG was released slowly and continuously in vivo and in vitro for up to 10 d, which could reduce the side effects caused by the fluctuation of blood drug concentration and solve the problem of the long treatment cycle and frequent administration. In vivo pharmacokinetics depicted that PPSG could improve the bioavailability, decrease the peak concentration, and prolong the t 1/2 of AtO and AtRA. Particularly, AAP significantly inhibited the tumor volume, extended the survival period of tumor-bearing mice, and promoted the differentiation of APL cells into normal cells. therefore, AtO + AtRA-PPSG not only could co-load hydrophilic AtO and lipophilic AtRA according to the clinical dosage, but also possessed the sustained-release and long-acting treatment effect which was expected to reduce administration time and ameliorate compliance of patients. thus, it had great potential for clinical transforma

Continuous Stirred tank Reactors (CStR) are used ubiquitously in chemical process industry for mixing, reactions and crystallizations. the purpose of this research is to study the effect of solute concentration (sodium chloride) on the stirrer speed in a CStR. the experimental method used in this research is the stop watch and beaker method. the behavior pattern of the fluid in a CStR can be experimentally verified as an alternative to the mathematical model. the tracer experiments were carried out. the effect of different parameters such as stirring speed, concentration at a steady time interval was analyzed. Experimental data obtained at 0.6 M and 160 rpm gave a curve that is in close agreement to that of theoretical or mathematical model. In other words, the results obtained at a concentration of 0.6 M and 160 rpm are in close agreement with that of early researchers. the paper is divided into five main sections: the first section immediately after the abstract is the introductory section that is basically a review of literature;the second section takes care of the materials and methods;the third section is the experimental procedure and this section takes the shut-down procedure and data processing into consideration;the fourth section is the discussion of experimental results and the last section is the conclusion of the paper.

AIM: to investigate physicians' knowledge including chronic hepatitis B(CHB) diagnosis, screening, and management in various stages of their ***: A voluntary 20-question survey was administered in Santa Clara County, CA where Asian and Pacific Islanders(API) account for a third of the population. Among the 219 physician participants,there were 63 interns, 60 second-year residents, 26 chief residents and 70 attending physicians. the survey asked questions regarding respondents' demographics,general hepatitis B virus knowledge questions(i.e.,transmission, prevalence, diagnostic testing, prevention,and treatment options), as well as, self-reported practice behavior and confidence in ***: Knowledge about screening and managing patients with CHB was poor: only 24% identified the correct tests to screen for CHB, 13% knew the next steps for patients testing positive for CHB, 18% knew the high prevalence rate among API, and 31% knew how to screen for liver cancer. Wald chi-square analysis determined the effect of training level on knowledge; in all cases except for knowledge of liver cancer screening(p = 0.0032), knowledge did not significantly increase with length in residency training or completion of ***: Even in a high-risk region, both medical school and residency training have not adequatelyprepared physicians in the screening and management of CHB.

Symptomatic neuromas are an all-too-common complication following limb amputation or extremity trauma,leading to chronic and debilitating pain for *** resection of symptomatic neuromas has proven to be the superior method of intervention,but traditional methods of neuroma resection do not address the underlying pathophysiology leading to the formation of a future symptomatic neuroma and lead to high reoperation *** approaches employ the physiology of peripheral nerve injury to harness the regeneration of nerves to their *** review explores the underlying pathophysiology of neuroma formation and centralization of pain *** compares the traditional surgical approach for symptomatic neuroma resection and describes three novel surgical strategies that harness this pathophysiology of neuroma formation to their *** traditional resection of symptomatic neuromas is currently the standard of care for amputation patients,but new techniques including the regenerative peripheral nerve interface,targeted muscle reinnervation,and intraosseous transposition have shown promise in improving patient pain outcomes for postamputation pain and residual limb *** neuromas are a chronic and debilitating complication following amputation procedures and trauma,and the current standard of care does not address the underlying pathophysiology leading to the formation of the *** techniques are under development that may provide improved patient pain outcomes and a higher level of care for symptomatic neuroma resection.

this study used six fields data alongside correlation heat map to evaluate the field parameters that affect the accuracy of bottom hole pressure(BHP)*** six oil field data were acquired using measurement while drilling device to collect surface measurements of the downhole pressure data while *** the two case studies,measured field data of the wellbore filled with gasified mud system was utilized,and the wellbores were drilled using rotary jointed drill *** Randomized tree and feed forward neural network algorithms were used to develop models that can predict with high accuracy,BHP from measured field *** modeling purpose,an extensive data from six fields was used,and the proposed model was further validated with two data from two new *** gathered data encompasses a variety of well data,general information/data,depths,hole size,and *** developed model was compared with data obtained from two new fields based on its capability,stability and *** result and model’s performance from the error analysis revealed that the two proposed Extra tree and Feed Forward models replicate the bottom hole pressure data with R2 greater than *** high values of R^(2) for the two models suggest the relative reliability of the modelling *** magnitudes of mean squared error and mean absolute percentage error for the predicted BHPs from both models range from 0.33 to 0.34 and 2.02%-2.14%,for the Extra tree model and 0.40-0.41 and 3.90%e3.99%for Feed Forward model respectively;the least errors were recorded for the Extra tree ***,the mean absolute error of the Extra tree model for both fields(9.13-10.39 psi)are lower than that of the Feed Forward model(10.98-11 psi),thus showing the higher precision of the Extra tree model relative to the Feed Forward *** has shown that underbalanced operation does not guarantee the improvement of horizontal well’s extension ability,because it mainly depends on t

Misfoldedα-synudein is a major component of Lewy bodies,which are a hallmark of Parkinson's disease(PD).A large body of evidence shows thatα-synuclein can aggregate into amyloid fibrils,but the relationship between a-synuclein self-assembly and Lewy body formation remains ***,we show,both in vitro and in a Caenorhabditis elegans model of PD,thatα-synuclein undergoes liquid-liquid phase separation by forming a liquid droplet state,which converts into an amyloid-rich hydrogel with Lewy-body-like *** maturation process towards the amyloid state is delayed in the presence of model synaptic vesicles in *** together,these results suggest that the formation of Lewy bodies may be linked to the arrested maturation ofα-synuclein condensates in the presence of lipids and other cellular components.
