This retrospective cohort study examined the relationship between self-reported participation in flexibility and muscular strengthening activities and the development of functional limitation(i.e.,once an individual has difficulty with or becomes unable to perform activities of daily living).Data were obtained from 1318 adults(mean age 49.59.7 years;98.7%Caucasian;14.9%female)enrolled in the Aerobics Center Longitudinal Study from 1979 to 2004 and free of functional limitation at ***-back health surveys were used to prospectively determine incident functional *** in muscle-strengthening and flexibility activities was assessed via *** logistic regression analyses were used to determine the odds ratios(OR)and corresponding 95%confidence intervals for developing functional limitation during follow-up based on participation in general and specific categories of flexibility(‘Stretching’,‘Calisthenics’,or‘Exercise Class’)and musclestrengthening activities(‘Calisthenics’,‘Free Weights’,‘Weight Training machines’,or‘Other’).Overall,42.6%of the sample reported incident functional *** adjusting for potential confounders(e.g.,age,sex,cardiometabolic risk factors),those who reported performing muscle-strengthening activities in general(n=685)were at lower risk of developing functional limitation[OR=0.79(0.63–1.00)].In addition,the specific flexibility activities of stretching(n=491)and calisthenics(n=122)were associated with 24%and 38%decreased odds of incident functional limitation,*** muscle-strengthening,stretching,and calisthenics activities are prospectively associated with decreased risk of incident functional limitation in generally healthy,middleaged and older ***,both public health and rehabilitation programs should highlight the importance of flexibility and muscle-strengthening activities during adulthood to help preserve functional capacity.

The transition metal manganese （mn） is indispensable for photoautotrophic growth since photosystem II （PSII） employs an inorganic mn4CaOs cluster for water splitting. Here, we show that the Arabidopsis membrane protein CHLOROPLAST mANGANESE TRANSPORTER1 （CmT1） is involved in chloroplast mn homeostasis. CmT1 is the closest homolog of the previously characterized thylakoid mn transporter PHOTOSyNTHESIS-AFFECTED mUTANT71 （PAm71）. In contrast to PAm71, CmT1 resides at the chloro- plast envelope and is ubiquitously expressed. Nonetheless, like PAm71, the expression of CmT1 can also alleviate the mn-sensitive phenotype of yeast mutant qomrl. The cmtl mutant is severely suppressed in growth, chloroplast ultrastructure, and PSII activity owing to a decrease in the amounts of pigments and thylakoid membrane proteins. The importance of CmT1 for chloroplast mn homeostasis is demonstratedby the significant reduction in chloroplast mn concentrations in cmtl-1, which exhibited reduced mn binding in PSII complexes. moreover, CmT1 expression is downregulated in mn-surplus conditions. The pam71 cmtl-ldouble mutant resembles the cmtl-f single mutant rather than pare71 in most respects. Taken together, our results suggest that CmT1 mediates mn2 uptake into the chloroplast stroma, and that CmT1 and PAm71 function sequentially in mn delivery to PSII across the chloroplast envelope and the thylakoid membrane.

The management of metastatic Thyroid Carcinoma: An Initial Presentation with Bony metastasis

Introduction: metastatic thyroid carcinoma presenting at the initial time of diagnosis is uncommon and the prognosis is unclear. Long term survival rates are variable ranging from 13% to 100%. This case report is presented to illustrate potential management and lend statistical power to future analysis of the correct treatment planning, mortality rates, and prognostic indications for an uncommon presentation of thyroid cancer. Case presentation: This patient is a 63 year old female who presented with new onset of progressive right hip pain. She was treated with a cortisone injection for presumed osteoarthritis but did not improve. Physical exam at the time was pertinent for a body mass index (BmI) of 38.4, mild systolic hypertension, difficulty walking secondary to the right hip pain, limited range of motion at the hip, and fullness of right thyroid gland with no palpable nodules. Laboratory evaluation including thyroid function tests was normal. However, CT and mRI scans revealed a 6.5 cm × 5 cm osteolytic expansive lesion on the right iliac crest with a soft tissue mass. In addition, an enlarged right thyroid lobe and small nodular densities in the lungs suspicious for metastatic disease were noted. A fine needle aspiration of the right ileum revealed metastatic follicular adenocarcinoma consistent with a thyroid primary. The patient was not a surgical candidate due to the extent of disease. She received 37.5 Gy to both the right iliac crest mass and the neck to include the thyroid lesion, followed by iodine-131 ablation and bisphosphonate therapy. Her disease was stable on her last follow up at 48 months. Conclusion: Further studies identifying independent variables such as age of the patient, site and extent of the disease and histology of the tumor are needed to help determine the true prognosis and proper management patients with this presentation. During LC-mS quantitation of drugs for pharmacokinetic assessment, usually metabolites are not quantified due to the unavailability of reference standards. If a metabolite is quantified without a reference standard, then it is assumed that the LC-mS response to a drug is similar to that of its metabolite and the standard curve, of the parent compound, is used to quantitate the metabolite. This approach could result in an over or underestimation of the metabolite. To evaluate the impact of mobile phase composition on LC-mS response, the mobile phases were interchanged between methanol, acetonitrile and a 50/50 mixture of methanol/acetonitrile. UHPLC flow rates were varied from 200-500 μL/min, with and without the addition of reverse composition of mobile phases, at the parent drug retention time. This change was necessary to achieve uniform mS responses for drugs and their metabolites. In this study, HRmS data, obtained using orbi-trap, resulted in a linear response over a wider dynamic range than that obtained using the linear ion trap. Overall, the parameters, required for achieving standard free quantitation, are dependent upon the mobile phase composition and LC flow rates.

In response to new European Union regulations,studies are underway to mitigate accumulation of toxic cadmium(Cd)in cacao(Theobroma cacao,Tc).This study advances such research with Cd isotope analyses of 19 genetically diverse cacao clones and yeast transformed to express cacao natural resistance-associated macrophage protein(NRAmP5)and heavy metal ATPases(HmAs).The plants were enriched in light Cd isotopes relative to the hydroponic solution withΔ^(114/110)Cd_(tot-sol)=−0.22±0.08‰.Leaves show a systematic enrichment of isotopically heavy Cd relative to total plants,in accord with closed-system isotope fractionation ofΔ^(114/110)C_(dseq-mob)=−0.13‰,by sequestering isotopically light Cd in roots/stems and mobilisation of remaining Cd to *** findings demonstrate that(i)transfer of Cd between roots and leaves is primarily unidirectional;(ii)different clones utilise similar pathways for Cd sequestration,which differ from those of other studied plants;(iii)clones differ in their efficiency of Cd *** yeast that expresses TcNRAmP5(*** natural resistance-associated macrophage gene)had isotopically lighter Cd than did *** suggests that NRAmP5 transporters constitute an important pathway for uptake of Cd by *** isotope signatures of transgenic yeast expressing HmA-family proteins suggest that they may contribute to Cd *** data are the first to record isotope fractionation induced by transporter proteins in vivo.

A new potential energy surface （PES） for the atmospheric formation of sulfuric acid from OH＋SO2 is investigated using density functional theory and high-level ab initio molecular orbital theory. A pathway focused on the new PES assumes the reaction to take place between the radical complex ***2 and H2O. The unusual stability of ***2 is the principal basis of the new pathway, which has the same final outcome as the current reaction mechanism in the literature but it avoids the production and complete release of SO3. The entire reaction pathway is composed of three consecutive elementary steps：（1） HOSO2＋O2-＋***2, （2） ***2＋H20-＋SO3·H2O·HO2, （3） ***2-＋H2SO4＋HO2. All three steps have small energy barriers, under 10 kcal/rnol, and are exotherrnic, and the new pathway is there- fore favorable both kinetically and therrnodynarnically. As a key step of the reactions, step （3）, HO2 serves as a bridge molecule for low-barrier hydrogen transfer in the hydrolysis of SO3. Two significant atmospheric implications are expected frorn the present study. First, SO3 is not released from the oxidation of SO2 by OH radical in the atmosphere. Second, the conversion of SO2 into sulfuric acid is weakly dependent on the humidity of air.
