Genomic scanning enabling discovery of a new antibacterial bicyclic carbamate-containing alkaloid

Non-ribosomal peptides are a group of structurally diverse natural products with various important therapeutic and agrochemical *** pyrrolizidine alkaloids(PAs),containing a scaffold of two fused five-membered ring system with a nitrogen atom at the bridgehead,have been found to originate from a multidomain non-ribosomal peptide synthetase to generate indolizidine intermediates,followed by multistep oxidation,catalysed by single Bayer-Villiger(BV)enzymes,to yield PA *** bacterial PAs are rare in natural product inventory,bioinformatics analysis suggested that the biosynthetic gene clusters(BGCs)that are likely to be responsible for the production of PA-like metabolites are widely distributed in bacterial ***,most of the strains containing PA-like BGCs are not deposited in the public domain,therefore preventing further assessment of the chemical spaces of this group of bioactive ***,we report a genomic scanning strategy to assess the potential of PA metabolites production in our culture collection without prior knowledge of genome *** the strains tested,we found fifteen contain the key BV enzymes that are likely to be involved in the last step of PA ring *** one-strain-many-compound(OSMAC)method,supported by a combination of HR-MS,NMR,SMART 2.0 technology,and GNPS analysis,allowed identification and characterization of a new[5+7]heterobicyclic carbamate,legoncarbamate,together with five known PAs,bohemamine derivatives,from Streptomyces ***37,a Ghanaian soil *** absolute stereochemistry of legoncarbamate was determined by comparison of measured and calculated ECd *** displays antibacterial activity against *** ATCC 25922 with an MIC value of 3.1μg/***,a biosynthetic model of legoncarbamate and other bohemamines was proposed based on the knowledge we have gained so far.

Temporal dynamics of neonatal hypoxic-ischemic encephalopathy injuries on magnetic resonance imaging

Moderate to severe perinatal hypoxic-ischemic encephalopathy occurs in~1 to 3/1000 live births in high-income countries and is associated with a significant risk of death or neurodevelopmental *** assessment is important to help identify highrisk infants,to help families,and to support appropriate interventions.A wide range of monitoring tools is available to assess changes over time,including urine and blood biomarkers,neurological examination,and *** present,magnetic resonance imaging is unique as although it is expensive and not suited to monitoring the early evolution of hypoxic-ischemic encephalopathy by a week of life it can provide direct insight into the anatomical changes in the brain after hypoxic-ischemic encephalopathy and so offers strong prognostic information on the long-term outcome after hypoxic-ischemic *** review investigated the temporal dynamics of neonatal hypoxic-ischemic encephalopathy injuries,with a particular emphasis on exploring the correlation between the prognostic implications of magnetic resonance imaging scans in the first week of life and their relationship to long-term outcome prediction,particularly for infants treated with therapeutic hypothermia.A comprehensive literature search,from 2016 to 2024,identified 20 pertinent *** review highlights that while the optimal timing of magnetic resonance imaging scans is not clear,overall,it suggests that magnetic resonance imaging within the first week of life provides strong prognostic *** challenges limit the timing consistency,particularly the need for intensive care and clinical ***,although most reports examined the prognostic value of scans taken between 4 and 10 days after birth,there is evidence from small numbers of cases that,at times,brain injury may continue to evolve for weeks after *** suggests that in the future it will be important to explore a wider range of times after hypo

Optimization of thermophilic anaerobic-aerobic treatment system for Palm Oil Mill Effluent （POME）

Optimization of an integrated anaerobic-aero- bic bioreactor （IAAB） treatment system for the reduction of organic matter （Chemical Oxygen demand （COd）, Biochemical Oxygen demand （BOd） and Total Sus- pended Solids （TSS） concentrations） in Palm Oil Mill Effluent （POME） to legal standards with high methane yield was performed for the first time under thermophilic condition （50~C-55~C） by using response surface meth- odology （RSM）. The experiments were conducted based on a central composite rotatable design （CCRd） with three independent operating variables, organic loading rates in anaerobic compartment （OLRan） and mixed liquor volatile suspended solids （MLVSS） concentration in anaerobic （MLVSSa,） and aerobic compartments （MLVSSa）. The optimum conditions for the POME treatment were determined as OLRan of 15.6 g COd＇L-I＂d-1, MLVSSan of 43100mg.L l, and MLVSSa of 18600mg.L-1, where high aerobic COd, BOd and TSS removal efficiencies of 96.3%, 97.9%, and 98.5% were achieved with treated BOd of 56mg.L1 and TSS of *** meeting the discharge standard. This optimization study successfully achieved a reduction of 42% in the BOd concentrations of the final treated effluent at a 48% higher OLRan as compared to the previous works. Besides, thermophilic IAAB system scores better feasibility and higher effectiveness as compared to the optimized mesophilic system. This is due to its higher ability to handle high OLR with higher overall treatment efficiencies （more than 99.6%）, methane yield （0.31 L CH4＂ gl COdremoved） and purity of methane （67.5%）. Hence, these advantages ascertain the applicability ofthermophilic IAAB in the POME treatment or even in other high-strength wastewaters treatment.

Introduction:Western climate science and Traditional Ecological Knowledge(TEK)represent complementary and overlapping views of the causes and consequences of *** particular,observations of changes in abundance,distribution,phenology,or behavior of the natural environment(including plants and animals)can have a rich cultural and spiritual interpretation in Indigenous communities that may not be present in western science ***:Using interviews with Indigenous elders and other Traditional Knowledge holders,we demonstrate that assumptions about the nature,perception,and utilization of time and timing can differ across knowledge systems in regard to climate ***:Our interviewees’focus on relationality predisposes them to notice interactional changes among humans and other species,to be sensitive to smaller scale examples of change,to be more likely to see climate change as part of a broader time scale,and to link changes to a greater suite of socio-political phenomena,including the long arc of *** implication of this research and the interactions among humans and other species is that policies restricting Native and non-Native access to resources(i.e.,hunting and fishing)to certain calendar seasons may need to be revisited in a changing climate.

Impacts of wetting-drying cycles on short-term carbon and nitrogen dynamics in Amynthas earthworm casts

Effects of earthworm casts on soil nutrient dynamics and their responses to changing moisture availability in subtropical ecosystems remain poorly *** study aimed to examine short-term carbon(C)and nitrogen(N)dynamics and their interactions with wetting-drying cycles in three different structural forms(i.e.,granular,globular,and heap-like)of Amynthas earthworm *** rates of C and N mineralization in the earthworm casts were examined under two different wetting-drying cycles(i.e.,2-d and 4-d wetting intervals)using a rainfall simulation *** three simulated rainfall events,subsamples of the earthworm casts were further incubated for 4 d for the determination of CO2 and N2O *** results of this study indicated that the impacts of wetting-drying cycles on the short-term C and N dynamics were highly variable among the three cast forms,but wetting-drying cycles significantly reduced the cumulative CO2 and N2O fluxes by 62%-83%and 57%-85%,respectively,when compared to the control without being subjected to any rainfall *** C mineralization rates in different cast forms were affected by the amount of organic substrates and N content in casts,which were associated with the food preference and selection of ***,the cumulative N2O fluxes did not differ among the three cast *** wetting and drying of casts not only enhanced aggregate stability by promoting bonds between the cast particles,but also inhibited microbial survival and growth during the prolonged drying period,which together hindered decomposition and *** findings demonstrated that the interactions between the structural forms,aggregate dynamics,and C and N cycling in the earthworm casts were highly complex.
