CRISPR/Cas9 genome editing technology can overcome many limitations of traditional breeding,offering enormous potential for crop improvement and food *** the direct delivery of Cas9-single guide RNA(sgRNA)ribonucleoprotein(RNP)complexes to grapevine(Vitis vinifera)protoplasts has been shown before,the regeneration of edited protoplasts into whole plants has not been ***,we describe an efficient approach to obtain transgene-free edited grapevine plants by the transfection and subsequent regeneration of protoplasts isolated from embryogenic *** proof of concept,a single-copy green f luorescent protein reporter gene(GFP)in the grapevine cultivar Thompson Seedless was targeted and knocked out by the direct delivery of RNPs to ***/Cas9 activity,guided by two independent sgRNAs,was confirmed by the loss of GFP f *** regeneration of GFP−protoplasts into whole plants was monitored throughout development,confirming that the edited grapevine plants were comparable in morphology and growth habit to wild-type *** report the first highly efficient protocol for DNA-free genome editing in grapevine by the direct delivery of preassembled Cas9-sgRNA RNP complexes into protoplasts,helping to address the regulatory concerns related to genetically modified *** technology could encourage the application of genome editing for the genetic improvement of grapevine and other woody crop plants.

Today,planning procedures which are used in different fields of decision making such as determination of optimum planting patterns are considered as important management *** importance of optimum planting patterns is highlighted when we learn that agricultural measures may have environmental side *** economical analyses only focus on increasing economic gains of the farmers without regard to its environmental ***,one can argue that efficient managers should consider multiple purposes that cover both economic and environmental goals at the same *** study attempted to identify an optimum mixed model of organic and non-organic production systems using a combination of AHP(Analytical Hierarchy Process)approach andWeighted Goal Programming to consider environmental and economic indicators *** procedure was employed in the current design to determine and compare an optimum pattern of mixed planting of organic and non-organic *** study sample was cucumber,investigated in four farming systems:organic open field farming system,nonorganic greenhouse farming system,non-organic tunnel farming system and open field non-organic farming system in Jiroft,Kerman,with regard to paper *** the proposed MCDM(Multiple Criteria Decision Making)model,cucumber planting in open field non-organic farming system was replaced by open field organic farming *** and environmental indicators rose by 11.97%and 21.40%respectively in MCDM proposed plan in comparison with the existing farm plan,which indicates the feasibility of MCDM proposed plan in terms of economic and environmental indicators.

Transcranial direct current stimulation(tDCS)has been reportedly beneficial for different neurodegenerative *** has been reported as a potential adjunctive or alternative treatment for auditory verbal hallucination(AVH).This study aims to review the effects of tDCS on AVH in patients with schizophrenia through combining the evidence from randomized clinical trials(RCTs).The databases of PsycINFO(2000–2019),PubMed(2000–2019),EMBASE(2000–2019),CINAHL(2000–2019),Web of Science(2000–2019),and Scopus(2000–2019)were systematically *** clinical trials with RCT design were selected for final analysis.A total of nine RCTs were eligible and included in the *** RCTs were included in the final *** them,six RCTs reported a significant reduction of AVH after repeated sessions of tDCS,whereas three RCTs did not show any advantage of active tDCS over sham *** current studies showed an overall decrease of approximately 28%of AVH after active tDCS and 10%after sham *** tDCS protocols targeting the sensorimotor frontal-parietal network showed greater treatment effects compared with the protocols targeting other *** this regard,cathodal tDCS over the left temporoparietal area showed inhibitory effects on *** most effective tDCS protocol on AVHs was twice-daily sessions(2 mA,20-minute duration)over 5 consecutive days(10 sessions)with the anode over the left dorsolateral prefrontal cortex and the cathode over the left temporal *** patient-specific and diseasespecific factors such as young age,nonsmoking status,and higher frequencies of AVHs seemed to be the predictors of treatment *** together,the results of tDCS as an alternative treatment option for AVH show controversy among current literatures,since not all studies were ***,the studies targeting the same site of the brain showed that the tDCS could be a promising treatment option to reduce *** RCTs,with larger sample sizes,should be con

Bleomycins(BLMs), as tumor-seeking antibiotics, have been used for over 20 years in treatment of several types of cancers. Several radioisotopes are used in radiolabeling of BLMs for therapeutic and diagnostic purposes. An important points in developing new radiopharmaceuticals, especially therapeutic agents, is the absorbed dose delivered in critical organs. In this work, absorbed dose to organs after injection of ^(153)Sm-,^(177)Lu- and ^(166)Ho-labeled BLM was investigated by radiation dose assessment resource(RADAR) method based on biodistribution data in wild-type rats. The absorbed dose effect of the radionuclides was evaluated. The maximum absorbed dose for the complexes was observed in the kidneys, liver and lungs. For all the radiolabeled BLMs,bone and red marrow received considerable absorbed dose. Due to the high energy beta particles emitted by ^(166)Ho, higher absorbed dose is observed for ^(166)Ho-BLM in the most organs. The reported data can be useful for the determination of the maximum permissible injected activity of the radiolabeled BLMs in the treatment planning programs.

OBJECTIVE:To investigate immunomodulatory effects of Astragalus polysaccharides(APS)on the co-culture of peripheral blood mononuclear cells(PBMCs)with He La cervical cancer cell ***:To assess the proliferation of PBMCs,carboxyfluorescein succinimidyl ester(CFSE)-labeled PBMCs were co-cultured with He La cells and treated with different concentrations of *** of cell culture were collected for cytokines assay via enzyme-linked immunosorbent assay(ELISA).The impact of APS on the proliferation of PBMCs,induction of regulatory T cells(Tregs),and myeloid-derived suppressor cells(MDSCs)was carried out by flow ***:It was observed that APS could increase the proliferation of PBMCs co-cultured with He La cells(P0.05).Furthermore,ELISA results demonstrated that APS could decrease IL-10 and TGF-βconcentration(P<0.05).CONCLUSION:The above-mentioned characteristics showed that APS might be able to modulate immune responses and improve anti-tumor effects through increasing the proliferation of PBMCs and decreasing inhibitory cytokines secretion as critical mediators of immune suppression in the tumor microenvironment.
